Antifungal effect of chitosan on the mycelial growth of Ganoderma boninense, Basal Stem Rot (BSR) pathogen of oil palm in vitro

Ganoderma boninense isolates (GBB 7, GBB 18, GBLS, and GBU) were collected from BSR infected oil palm and was identified microscopically, while pure mycelia culture was confirmed by PCR techniques. GBLS is the most aggressive isolate screened through pathogenicity test using rubber wood block inoculation technique. After 20 days, all of the concentrations (0.5, 1.0, 1.5, 2.0 % w/v) significantly reduced in vitro mycelial growth of GBLS when compared with control treatment. Chitosan exhibited a fungistatic effect on mycelial growth of G. boninense and markedly reduced radial growth with increased concentration. The highest inhibition of radial growth (PIRG) was observed with chitosan at 2.0 % w/v. The germination of G. boninense basidiospores treated with 1000 and10000 ppm of chitosan were affected significantly after 14 days incubation period. Chitosan also caused morphological changes in G. boninense mycelium including the occurrence of small vesicles due to coagulation of fungal cytoplasm and formation of excessive abnormal hyphal branching at higher concentrations.
