The effect of 6-dimethylaminopurine, a protein kinase(s) inhibitor involved in nuclear envelope (re)organization and chromatin remodelling, was investigated on embryos reconstructed by nuclear transplantation. Metaphase II oocytes (n. 121) were enucleated and reconstructed with 16-32 cells embryos; 67 were treated for two hours, during and after fusion, with 2 mM 6-DMAP, while the others served as a control. After seven days culture into ligated oviduct in ewes the development to blastocyst stage was significantly higher (82% vs 25%, p0.001) in 6-DMAP group. [Su embrioni ricostruiti mediante trapianto nucleare e' stato studiato l'effetto della 6-dimetilaminopurina (6-DMAP), un inibitore delle chinasi proteiche interessate alla (ri)organizzazione della membrana nucleare e al rimodellamento della cromatina. Oociti in metafase II (n. 121) sono stati enucleati e ricostruiti con embrioni allo stadio di 16-32 cellule; 67 di questi sono stati trattati per due ore, durante e dopo la fusione, con 2 mM di 6-DMAP, mentre gli altri servivano come controllo.
