Screening of highly-polymorphic RFLP [restriction fragment lenght polymorphism] probes in Oryza sativa L.

anglais. The abilities of 185 RFLP probes on the detection of polymorphisms in Oryza sativa were studied. Eight-five rice varieties, including 54 indica varieties and 31 japonica varieties, were selected and treated as random samples for the indica and japonica subspecies, respectively. Of the total 185 probes, 174 probes were used for both subspecies and 11 were used for indica subspecies only. The numbers of polymorphic probes were 121, 115 and 128 for intra-indica, intra-japonica and inter-subspecies, respectively. Large variation was observed on the frequencies of polymorphism, mainly due to the variation among probes. Based on the frequency of polymorphism detected by each probe, highly-polymorphic probes were screened. Altogether, 88, 91 and 95 probes were recommended to be used as an initial set of RFLP markers for gene mapping in populations derived from crosses of intra-indica, intra-japonica and inter-subspecies, respectively. These highly-polymorphic probes were distributed over all the 12 chromosomes of rice, and tended to cluster in some specific intervals rather than evenly scatter over the rice genome

italien. [E' stata studiata la capacita' di 185 sonde RFLP nell'identificazione del polimorfismo in Oryza sativa. Ottantacinque varieta' di riso, comprendenti 54 indica e 31 japonica, sono state selezionate e trattate come campioni casuali, rispettivamente per le sottospecie indica e japonica. Delle 185 sonde, 174 sono state utilizzate per ambedue le sottospecie e 11 solamente per la sottospecie indica. I numeri di sonde polimorfiche erano 121, 115 e 128, rispettivamente, per intra-indica, intra-japonica e inter-sottospecie. E' stata osservata un'ampia variazione sulle frequenze del polimorfismo, dovuta in gran parte alla variazione fra le sonde. Sulla base della frequenza del polimorfismo identificata da ogni sonda, sono state selezionate sonde altamente polimorfiche.
