Manipulation of the expression of heme activation protein HAP5c gene in transgenic plants

The HAP52/3/5 complex heme activation protein is a CCAAT binding factor, composed of three essential subunits. In yeast it has been demonstrated that this complex activates the transcription of a wide array of respiratory genes, and the mutation in any of its subunits results in reduced growth of non fermentable carbon sources. In the present study, the counterpart of the yeast HAP5 gene in Arabidopsis HAP5c was expresses, using the 35S cauliflower mosaic virus promoter, in the antisense and sense orientations in Arabidopsis and tobacco plants, respectively. Vacuum infiltration method was used to transform Arabidopsis, while tobacco plants were transformed by the particle bombardment method. Transgenic Arabidopsis carrying the antisense HAP5c exhibited three kinds of phenotypes.
