Expression of the glossy2 gene of maize during plant development [Zea mays L.]

anglais. The glossy2 (gl2) gene of maize participates to the synthesis-deposition of the juvenile epidermal wax layer of maize leaves. In this paper, the expression of the gene was analysed at the RNA and protein levels. The results showed that in wild type plants the gl2 gene is transcribed in young leaves, in coleoptiles, in husks, in immature ears and in silks. Neither transcripts were found in roots of wild type plants nor in any tissues of plants homozygous for the recessive gl2-ref allele. Antibodies against the gl2 protein revealed its predominant presence in leaves with a juvenile wax phenotype and protein detection was consistent with gene transcription in wild type coleoptiles, husks, immature ears and silks, but not in roots, anthers and seeds. Sequence data bank analysis revealed homologies of the gl2 protein of maize with eighteen sequences from several plant species. Four of these proteins are induced in species different from maize during the hypersensitive response to plant pathogens. One protein is present in maize silks. A further set of four proteins represent polypeptides responsible for CoA-dependent acyl transfer in plants. The results provided contribute to assign a putative function to the gl2 gene
Afficher plus [+] Moins [-]italien. [Il gene glossy2 (gl2) del mais partecipa alla sintesi-deposizione dello strato ceroso epidermico giovanile delle foglie del mais. In questa nota, l'espressione del gene e' stata analizzata a livello dell'RNA e delle proteine. I risultati hanno dimostrato che nelle piante normali il gene gl2 e' trascritto nelle foglie giovani, nei coleottili, nei cartocci, nelle spighe immature e nelle sete. Non sono stati trovati trascritti ne' nelle radici del tipo normale, ne' in alcun tessuto di piante omozigoti per l'allele recessivo gl2-ref. Anticorpi contro la proteina gl2 hanno rivelato la sua presenza predominante nelle foglie con un fenotipo giovanile ceroso e la presenza di proteine era in accordo con la trascrizione del gene nei coleottili del tipo ordinario, nonche' nei cartocci, nelle spighe e nelle sete immature, ma non nelle radici, nelle antere e nei semi. L'analisi effettuata con la banca dati delle sequenze ha evidenziato omologie delle proteine gl2 del mais con 18 sequenze provenienti da diverse specie vegetali. Quattro di queste proteine sono prodotte in specie diverse dal mais nel corso della risposta di ipersensibilita' ai patogeni. Una proteina e' presente nelle sete del mais. Un ulteriore gruppo di 4 proteine rappresenta i polipeptidi responsabili del trasferimento di acili dipendente dal CoA. I risultati presentati contribuiscono ad attribuire una funzione apparente al gene gl2]
