Callus induction and plant regeneration from immature and endosperm-supported mature embryos of winter oat (Avena sativa L.)

An efficient endosperm-supported callus induction and plant regeneration from mature embryos in 10 genotypes of oat (Avena sativa L.) have been established as an alternative method for immature embryo culture. Fifteen days after anthesis, immature embryos were aseptically dissected from seeds and placed with the scutellum upwards on a solid agar medium containing the inorganic components of MS and 2 mg 2,4-D per liter. Mature embryos were moved slightly on the imbibed seeds. The seeds with moved embryos were placed furrow downwards in dishes containing 8 mg 2,4-D per liter for callus induction. The developed calli and regenerated plants were maintained on 2,4-D-free MS medium. Plants regenerated from both embryo cultures were vernalised and grown to maturity in soil. Variability among genotypes was observed for callus induction, weight of callus, regeneration capacity of callus, culture efficiency and number of regenerated plants for both explant types. Endosperm-supported mature embryos had also a high frequency of embryo culture responses as immature embryos. The results suggested that endosperm-supported mature embryos might be useful totipotent targets for particle acceleration-mediated transformation

[Sono state messe a punto un'efficiente induzione del callo mantenuto dall'endosperma e la rigenerazione delle piante da embrioni maturi in 10 genotipi di avena (Avena sativa L.) come metodo alternativo per la coltura di embrioni immaturi. Quindici giorni dopo l'antesi, embrioni immaturi sono stati separati asetticamente dai semi e posti, con lo scutello rivolto verso l'alto, su un terreno solido di agar che conteneva i componenti inorganici del MS e 2 mg di 2,4-D per litro. Gli embrioni maturi sono stati leggermente mossi sui semi imbevuti. I semi con embrioni mossi sono stati posti rivolti con il solco verso il basso in dischi contenenti 8 mg di 2,4-D per litro per l'induzione del callo. I calli sviluppati e le piante rigenerate sono stati mantenuti su un terreno MS senza 2,4-D. Le piante rigenerate da ambedue le colture embrionali sono state vernalizzate e allevate fino alla maturita' su suolo. E' stata osservata una variabilita' fra i genotipi riguardo all'induzione del callo, al peso dei calli, alla capacita' di rigenerazione del callo, all'efficienza della coltura e al numero di piante rigenerate per ambedue i tipi di espianto. Gli embrioni maturi mantenuti dall'endosperma presentavano anche un'alta frequenza di risposte alla coltura embrionale come gli embrioni immaturi. I risultati fanno ritenere che gli embrioni maturi supportati dall'endosperma potrebbero rappresentare target totipotenti utili per una trasformazione tramite bombardamento con microproiettili]
