Detection of phytoplasmal DNA in flowers and seeds from elm trees infected with elm yellows [Ulmus]

The occurrence of phytoplasmas associated with elm yellows (EY) was investigated in the reproductive structures (flowers, unripe and ripe fruits) of two EY-infected trees of the hybrid elm clone Lobel and two healthy trees, an Ulmus laevis and an U. japonica. Phytoplasma group-specific Polymerase Chain Reaction (PCR), restriction fragment length polymorphism analysis and sequencing of amplified fragments were carried out using as template DNA extracted from these reproductive structures. The flowers and fruits were dissected into parts (the flowers into anthers and ovaries, the fruits into seeds and membranaceous wings) and then examined separately. A total of 350 seeds from infected trees were sown, producing 24 plantlets, which were sampled for EY phytoplasma DNA one and five months after germination. Both flowers and seeds from the EY-infected trees were good sources for the extraction and PCR-amplification of EY phytoplasmas, but no EY phytoplasmas were detected in either the flowers and seeds of the two healthy trees or in samples collected from the 24 plantlets grown from seed

[E' stata studiata la presenza di fitoplasmi associati all'ingiallimento dell'olmo (EY) nelle strutture riproduttive (fiori, frutti non maturi e maturi) di due piante del clone ibrido Lobel dell'olmo infettate da EY e due piante sane, un Ulmus laevis e un U. japonica. Sono state effettuate la reazione a catena della polimerasi specifica per il gruppo di fitoplasmi, l'analisi del polimorfismo della lunghezza dei frammenti di restrizione e il sequenziamento dei frammenti amplificati, utilizzando come standard il DNA estratto da queste strutture riproduttive. I fiori e i frutti sono stati sezionati in parti (i fiori in antere e ovari, i frutti in semi e ali membranose) e quindi esaminati separatamente. E' stato messo a dimora un totale di 350 semi derivanti dalle piante infette, da cui sono state prodotte 24 piantine, da cui e' stato prelevato un campione di DNA del fitoplasma dell'EY uno e cinque mesi dopo la germinazione. Sia i fiori, sia i semi derivanti dalle piante infette da EY sono risultati buone fonti per l'estrazione e l'amplificazione PCR dei fitoplasmi dell'EY, ma non sono stati individuati fitoplasmi dell'EY sia nei fiori, sia nei semi delle due piante sane o in campioni derivanti dalle 24 piantine ottenute da seme]
