Influence of osmotic and low-temperature stress factors on the development of Salvia sclarea L. in embryoculture in vitro | Влияние осмотического и низкотемпературного стрессовых факторов на развитие Salvia sclarea L. в эмбриокультуре in vitro

anglais. Creation of new genotypes resistant to abiotic stress factors is an important task of essential oil plants breeding. The purpose of the work was to study the effect of osmotic and low-temperature stresses on the development of clary sage zygotic germs in order to develop a selective system for creating forms resistant to these stress factors in vitro. The study involved 3 cultivars and 10 samples of Salvia sclarea L. differing in drought resistance. The effects of osmotic and low-temperature stresses were simultaneously tested in vitro. Under osmotic stress, the germs were cultured on Murashige and Skoog medium containing 3–5% sorbitol. Modeling of cold stress was carried out in 3 steps with different durations: hardening at 0…4 deg. C (3 or 8 days in different trial establishments; freezing with a decrease in temperature from 0 to –12...–14 deg. С; defrosting at 2…4 deg. C (2-4 days). After low-temperature stress, the germs were transplanted to a medium with the same sorbitol concentration and cultivated at 26 deg. C and illumination of 2–3 klx. It was found that as the sorbitol concentration increased to 5% and the temperature decreased to –8...–14 deg. C, all analyzed parameters (frequency of germination and developed seedlings, shoot and root length) decreased compared to the control. In genotypes with high drought resistance coefficients the inhibition of seedling development under the action of two stressors was much less than in unstable genotypes. The maximum correlation coefficients (from 0.696 to 0.870) were established between the field drought resistance of genotypes and the frequency of germination or developed seedlings. It is possible to reduce the total duration of low-temperature stress during in vitro selection from 19 to 8-10 days, and freezing from -8 ...-14 deg. C up to 3-5 days. A selective system was developed that can be used for in vitro selection or indirect assessment of S. sclarea forms having complex osmotic and low temperature resistance.

russe. Создание новых генотипов, резистентных к абиотическим стрессовым факторам - важная задача селекции эфиромасличных растений. Цель работы – изучить влияние осмотического и низкотемпературного стрессов на развитие зиготических зародышей шалфея мускатного для разработки селективной системы создания устойчивых к этим стрессовым факторам форм in vitro. В исследованиях использовали три сорта и 10 образцов Salvia sclarea L., различающихся по засухоустойчивости. Одновременно испытывали действие осмотического и низкотемпературного стрессов in vitro. При осмотическом стрессе зародыши культивировали на среде Мурасиге и Скуга, содержащей 3–5% сорбита. Моделирование холодового стресса проводили в три этапа с разной продолжительностью: закаливание при 0…4 град. С (3 или 8 сут в разных закладках опыта); промораживание при снижении температуры от 0 до –12…–14 град. С (от 4 до 11 сут); оттаивание при 2…4 град. С (2-4 сут). После низкотемпературного стресса зародыши пересаживали на среду с той же концентрацией сорбита и культивировали при 26 град. С и освещенности 2–3 клк. Установлено, что по мере повышения концентрации сорбита до 5% и снижения температуры до –8…–14 град. С все анализируемые параметры (частота прорастания зародышей, частота развившихся проростков, длина побега и корня) снижались по сравнению с контролем. У генотипов с высокими коэффициентами засухоустойчивости угнетение развития проростков при действии двух стрессоров было меньше, чем у неустойчивых генотипов. Максимальные коэффициенты корреляции (от 0,696 до 0,870) установлены между полевой засухоустойчивостью генотипов и частотой прорастания зародышей или частотой развившихся проростков. Возможно сокращение общей длительности низкотемпературного стресса при отборе in vitro с 19 до 8–10 сут, а промораживания до –8…–14 град. С – до 3–5 сут. Разработана селективная система, которую можно использовать для отбора или косвенной оценки in vitro форм S. sclarea с комплексной устойчивостью к осмотику и низкой температуре.
