anglais. The Nenets reindeer breed is the most numerous in the Russian Federation, has unique adaptive abilities and plays a key role in the life of numerous peoples of the Far North. It is relevant to search for and implement various approaches that will expand and clarify the available data on the state of genetic diversity and phylogenetic relationships of regional populations of the Nenets breed. The aim of the work is to validate the developed a test system for the isolation of complete mitochondrial genomes of reindeer and to conduct a phylogenetic analysis of domestic populations of the Nenets breed in comparison with the wild tundra population of reindeer. The test system is based on the amplification of six overlapping mtDNA fragments, which, after purification and quality control of the libraries, were sequenced using NGS technology and performed on a miSeq device (Illumina, USA). Approbation of the test system for the isolation of complete mitochondrial genomes of the reindeer was carried out on representatives of six regional populations of the Nenets breed (n=63), as well as the wild reindeer (n=62). The length of the complete mitogenome was 16367 base pairs, with the following distribution of nucleotide frequencies: A - 33.70%, T - 30.04%, G - 13.16%, C - 23.10%. All studied individuals of the Nenets breed were characterized by high haplotype and nucleotide diversity (Hd = 0.993±0.005; pi = 0.00391±0.00068). No clear population clustering of the studied reindeer samples was observed. Phylogenetic analysis revealed the presence of four main clusters with a mixed composition according to the distribution of haplotypes on the median network. As a result of the study, a test system was developed and tested for the isolation of complete reindeer mitochondrial genomes, new data were obtained on the phylogenetic relationships not only of groups of the Nenets reindeer of various geographical origins, but also of domestic and wild populations of this animal species.

Ненецкая порода северного оленя – самая многочисленная в Российской Федерации, обладает уникальными адаптационными способностями и играет ключевую роль в жизни многочисленных народов Крайнего Севера. Актуален поиск и внедрение различных подходов, которые позволят расширить и уточнить имеющиеся данные о состоянии генетического разнообразия и филогенетических взаимоотношениях региональных популяций ненецкой породы. Цель исследований – выполнить апробацию разработанной тест-системы для выделения полных митохондриальных геномов северных оленей и провести филогенетический анализ домашних популяций ненецкой породы, в сравнении с дикой тундровой популяцией северных оленей. Разработанная тест-система основана на амплификации шести перекрывающихся фрагментов мтДНК, которые после очистки и проверки качества библиотек секвенировали с использованием технологии NGS на приборе miSeq (Illumina, США). Апробацию тест-системы выделения полных митохондриальных геномов северного оленя проводили на представителях шести региональных популяций ненецкой породы (n=63) и диких северных оленей (n=62). Длина полного митогенома составила 16367 пар оснований со следующим распределением частот нуклеотидов: А – 33,70%, Т – 30,04%, G – 13,16%, C – 23,10%. Все исследуемые особи ненецкой породы характеризовались высоким гаплотипическим и нуклеотидным разнообразием (Hd = 0,993±0,005; пи = 0,00391±0,00068). Четкой популяционной кластеризации исследуемых выборок северных оленей не наблюдали. Филогенетический анализ выявил наличие четырех основных кластеров со смешанным составом согласно распределению гаплотипов на медианной сети. Разработана и апробирована тест-система для выделения полных митохондриальных геномов северного оленя, получены новые данные о филогенетических связях группировок ненецкой породы северного оленя различного географического происхождения, а также домашних и диких популяций этого вида животных.
