Physico-biochemical characteristics of Kadaknath and broiler chicken semen under storage condition

Attempts have been made to investigate the physical and biochemical characteristics of Kadaknath and broiler chicken semen under liquid storage 3–5°C at 24 hrs. Kadaknath and broiler chicken semen was collected by abdominal massage method and diluted (1:2) with CARI semen diluent and examined for various physical and biochemical characteristics. Semen volume was found 0.21±0.07 ml in kadaknath whereas nearly double in case of broiler (0.43±0.05 ml). No significant difference was found in sperm motility between fresh and preserved (24 h) semen in kadaknath whereas, a significant (P<0.05) reduction in sperm motility was investigated in preserved broiler chicken semen. A reduction (P<0.05) in per cent live counts associated with enhanced dead and morphologically abnormal spermatozoa were noticed in preserved semen in both the breeds of chicken. Sperm concentration was found slightly higher in broiler chicken than kadaknath. Biochemical characteristics of Kadaknath and broiler semen revealed a significant increase (P<0.05) in GOT activity and MBRT in preserved (24 h) semen at low temperature when compared to fresh semen. Semen acidity was noticed close to neutral pH in each breed. Profile of protein, GPT, cholesterol and ALP enzymes exhibited no significant difference between fresh and preserved semen. However, cholesterol and ALP enzyme activity was higher in broiler chicken biological fluid than Kadaknath. It was concluded that during liquid storage, quality of semen was deteriorated as evidenced by physical and biochemical characteristics of Kadaknath and broiler chicken semen.
