A Procedure for the determination of total protein in human milk

Extract: The colorimetric method of Lowry and associates was adapted to a simple procedure for total protein in human milk. The mean value for 19 random samples ranging from 2 to 448 days in lactation was 13.3 plus or minus 4.9 mg of protein per ml of milk. The 13 mature milks (greater than 30 days in lactation) averaged 10.7 plus 1.5 mg per ml. The principal difficulty in developing a suitable modification was turbidity caused by the milk lipids. Two solutions to the problem were found. For routine analysis of milks, removal of fat globules is the practical solution. The extraction of lipids after color development shows promise as a useful alternative for systems high in lipids, such as isolated milk fat globules. (Author)
