Nitric oxide regulates mitochondrial thioredoxin/ glutaredoxin system of peach fruit during cold storage

The mitochondrial thioredoxin/glutaredoxin system plays an important role in modulating redox balance to prevent oxidative damage caused by excessive reactive oxygen species (ROS). In this study, 'Feicheng' peaches were dipped, respectively, in 15 μmol L-1 nitric oxide (NO) solution, 5 μmol L-1 c-PTIO (2-(4-carboxyphenyl)-4,4,5,5- tetramethylimidazoline-1-oxyl-3-oxide, NO scavenger) solution and deionized water (control) for 30 min, and then stored at 0°C. The activities of thioredoxin reductase (TR), glutaredoxin reductase (GR), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR) and the contents of glutaredoxin (Grx), GSH and GSSG, were measured during storage in 'Feicheng' peaches. The results showed that, treatment with 15 μmol L-1 NO significantly increased TR activity and Grx content in first 5 d compared with the control and the c-PTIO treatment. The peaks of TR activity and Grx content in NO-treated peaches appeared 5 d earlier than those of the control and the treatment with c-PTIO. The content of GSH in NO-treated peaches was higher than that of the control and the c-PTIO treatment in the first 5 d. The content of GSSG in NO-treated peaches was higher than that of the control during storage at low temperature. Compared with the control and treatment with c-PTIO, treatment with 15 μmol L-1 NO significantly increased the activities of GR and APX in the first 10 d and the activities of MDHAR and DHAR in the first 5 d during storage. These results suggested that NO could help to delay the redox imbalance via promoting the mitochondrial thioredoxin/glutaredoxin system of peaches during cold storage.
