Development of an ic-ELISA and an immunochromatographic strip assay for the detection of aconitine

The medicinal properties and toxicity of aconitine in traditional Chinese medicine have caused widespread concern. In this study, we screened a highly specific and sensitive monoclonal antibody with an IC₅₀ of 3.65 ng/mL generated by immunizing mice and cell fusion, and the limits of detection of the ic-ELISA were 1.13 and 11.76 ng/mL. We then developed a colloidal gold test strip based on the antibody. For rapid detection of aconitine in herbs, the visual limit and detection limit of the test strip are 10 and 25 ng/mL, respectively; the results are obtained within 5 min and are visible to the naked eye. Therefore, the colloidal gold test strip is very suitable for on-site detection and quality screening of aconitine in Chinese herbal medicine.
