Effects of DA-6 soaking of cucumber seeds on cold-resistance of seedlings

The seeds of cucumber (Cucumis sativus L. 'Aoshu No.6') were soaked with an aqueous solution of hexanoic acid-2(diethylamino) ethyl ester (DA-6). The seedlings pretreated with 10-20 μg•L-1 DA-6 had a higher germination rate, chlorophyll content and growth rate than those untreated ones under room temperature (22-25°C). Under cold temperature (4-6°C, 3 d) and following room temperature recovery, the treated seedlings not only increased activities of superoxide dismutase (SOD), but also increased peroxidase (POD), which could maintain lower active oxygen and H2O2 levels. This in turn would reduce cell membrane damage resulting in a lower rate of electrolyte leakage and hence a higher seedling survival. In contrast, the untreated plants only increased SOD activities, not POD, so their H2O2 could maintain higher level, and have higher rate of electrolyte leakage resulting in lower survival percentage. The experiment suggested that DA-6 soaking cucumber seeds can improve cold-resistance of the seedlings, and the soaking method is simple to use, and the DA-6 concentration of soaking seeds is much lower than that of other methods.
