Culture and Sesquiterpene Analyses of Cells and Reǵenerated Plantlets of Pogostemon cablin Benth. (Patchouli)

Culture of cells of Pogostemon cablin Benth. on both solid and liquid media is described. In these cultures no free, volatile sesquiterpenes typical of the parent plant were detected by capillary gas liquid chromatography and combined gas liquid chromatography-mass spectrometry using methods sensitive to 4 X io(~I0)g sesquiterpene per mg fresh weight of plant tissue. Under the culture conditions used, cultures freshly initiated from explants regenerated shoot-like structures. These were developed into autotrophic plantlets. The morphology of these plantlets was unlike that of the parent plant, although glandular trichomes were present as in the parent plant. Only unidentifiable trace amounts of mass fragmentation patterns typical of sesquiterpenes could be detected in these plantlets, even when grown under similar conditions to the parent plants.
