Estudo experimental da resposta imune celular de bovinos vacinados com o peptídeo sintético sbbo23290 no controle da babesiose por Babesia bovis (Babes, 1888) | Experimental study of cellular immune response of bovines immunizated with synthetic peptide sbbo23290 to the control of babesia bovis’s (babes, 1888) babesiosis

Bovinos da raça holandesa entre 3-4 meses de idade receberam quatro imunizações subcutâneas com o peptídeo sintético SBbo23290 anti-Babesia bovis mais saponina, e foram desafiados com amostra virulenta de B. bovis, sendo avaliados quanto ao desenvolvimento da resposta imune celular. Para isso, foi determinado o perfil de linfócitos circulantes (CD4 + , CD8 + , CD21 + e WC1 + ) por citometria de fluxo e seguimento dos eventos microscópicos ocorridos em linfonodos através de técnicas diferenciais (Hematoxilina & Eosina e Verde Metil Pironina) e de imunohistoquímica (Peroxidase anti-peroxidase). Os estudos histológicos mostraram reatividade evidente de centros germinativos a partir do quinto dia após a primeira imunização. A hiperplasia dos cordões medulares foi mais evidente aos três dias após a terceira imunização. O antígeno SBbo23290 foi detectado em células SBbo23290 positivas durante todo o experimento, concomitantemente às alterações histológicas em órgãos linfóides. Quando foi determinada a composição fenotípica dos linfócitos de sangue periférico circulantes, observou-se no grupo peptídeo um aumento progressivo de linfócitos WC1+ até os sete dias após a segunda coleta, que decaiu ligeiramente após a terceira inoculação, aumentando novamente após o desafio com a amostra virulenta de B. bovis, o que não aconteceu com os demais grupos experimentais, havendo diferença estatística entre os grupos. Os linfócitos CD4 + e CD21 + decresceram após a primeira inoculação, aumentando após a segunda inoculação, e decaindo nas demais coletas, havendo diferença estatística entre os grupos. A porcentagem de linfócitos T totais no grupo peptídeo foi superior aos demais grupos durante quase todo o experimento, sendo que a maior porcentagem foi obtida após a segunda coleta, não havendo diferença estatística entre os grupos nas diferentes coletas. Sendo assim, pode-se concluir que o peptídeo sintético induz de maneira eficaz uma resposta imunológica antígeno- específica, na qual encontram-se envolvidos mecanismos celulares e humorais.

Tree-four month old bovines of the Holstein Freesan breed received four under–skin immunization of the synthetic peptideo 23290 anti-Babesia bovis plus saponina. They were challenged with an virulent sample of B. bovis and they were evaluated considering the development of the cellular immune answer through identification of CD4 + , CD8 + , CD21 + , and WC1 + T lymphocytes by flow citometry and microscopic evaluation of lymph nodes using the techniques H&E and Pironin Methil Green, such as PAP. The lymphonodes analysis demonstrated germinal centers reactivity at the 5 th day after the first immunization. Medullars cords hyperplasia was more evident three days after the third immunization. SBbo32290 antigen was detected in SBbo23290 positive cells during whole experiment, and simultaneous to the histology alterations in lymphonodes. Evaluating phenotypic composition of the peripheral blood lymphocytes, a progressive increase of WC1 + lymphocytes was detected, which decreased after third immunization, and an increment was observed again after the challenge. It was not observed in the another experimental groups. The CD4 + and CD21 + lymphocytes decreased after first immunization, increased after the second immunization, and decreased again in the following immunizations; the variation was statistically significant between the groups. The variation in total number of lymphocytes wasn’t statistically significant, although an increment of this cells was observed in the peptide group xvalmost whole experiment, comparing with the another groups. We can conclude that the syntetic peptideo induct an effective immunological specific-antigen answer, in with cellular and humoral mechanisms were involved.
