Der Einfluss phytogener Futterzusatzstoffe auf die Nährstoffverdaulichkeit und Aktivität verschiedener intestinaler Enzyme sowie Expression von Nährstofftransportern bei Absetzferkeln | Influence of phytogenic feed additive on the digestibility and the activity of some intestinal enzyme and gene expression of nutrient transporters in weaning piglets

In der vorliegenden Studie wurde der Einfluss eines phytogenen Futterzusatzstoffes, zweier polyphenolreicher Apfelextrakte sowie vier phytogener Substanzen auf die Adhäsion zwischen einem enteropathogenen Escherichia coli-Stamm und der intestinalen porcinen epithelialen Zelllinie IPEC-J2 in vitro getestet. Weiterhin wurden in einem Fütterungsversuch 24 weibliche und 24 kastrierte männliche abgesetzte Ferkel im Alter von 28 Tagen in zwei aufeinanderfolgenden Versuchsreihen auf vier Fütterungsgruppen aufgeteilt. Das Ferkelaufzuchtfutter enthielt in der Kontrollgruppe 0,2 % Weizenkleie. Der Versuchsgruppe B wurde 0,2 % der Gruppe C 0,2 % des phytogenen Ferkelfutters Fresta F und der Versuchsgruppe D 0,2 % eines polyphenolreichen Apfelextraktes hinzugefügt. Einmal wöchentlich erfolgte die Erhebung der zootechnischen Daten. An den Versuchstagen 25 –28 wurden die Tiere zur Probenentnahme euthanasiert und Blut-, Kot-, Urin-, Digestaproben aus dem Ileum sowie Gewebeproben aus dem Jejunum entnommen. Zusätzlich zu den Leistungsdaten wurde die scheinbare praecaecale Verdaulichkeit und die Gesamtverdaulichkeit des Rohproteins sowie der Aminosäuren ermittelt. Aus Gewebeproben des Jejunums wurden die Bürstensaummembranen isoliert, aus denen anschließend der Proteingehalt sowie die Aktivitäten der intestinalen Enzyme Saccharase, Maltase, Laktase, Aminopeptidase N und der alkalischen Phosphatase bestimmt wurden. Jejunumproben wurden auch zur Bestimmung der Genexpression der intestinalen Enzyme Saccharase, Maltase, Laktase, Aminopeptidase N sowie des natriumabhängigen Glukosetransporters-1 (SGLT-1) herangezogen. Weiterhin wurden elektrophysiologische Messungen am Jejunum in Ussingkammern durchgeführt. Die antioxidative Kapazität des Blutplasmas wurde mithilfe des FRAP-und des TEAC-Tests aus dem Plasma bestimmt. Die Ergebnisse der in vitro- Untersuchungen zeigten signifikante Reduktionen der Adhäsion der E.coli- Bakterien an den IPEC-J2-Zellen durch Zugabe von Bockshornkleesamen, Pektin, der beiden Apfelextrakte und des Zusatzes Fresta F®. Die phytogenen Substanzen Glucoraphanin und Sulforaphan bewirkten keine signifikante Reduktion der Adhäsion. Die Ergebnisse des Fütterungsversuchs zeigten keine signifikanten Unterschiede hinsichtlich der Leistungsparameter. Die praecaecale Verdaulichkeit der Aminosäure Valin erhöhte sich in der Bockshornkleegruppe verglichen mit der Kontrollgruppe und der Fütterungsgruppe D (Apfelextrakt). Die praecaecale Verdaulichkeit von Isoleucin erhöhte sich in der Fütterungsgruppe D (Apfelextrakt) verglichen mit den Fütterungsgruppen B (Bockshornkleesamen) und C (Fresta F®). Die Gesamtverdaulichkeit der Aminosäuren Threonin, Valin, Isoleucin und Lysin erhöhte sich der Apfelextraktgruppe gegenüber den anderen drei Versuchsgruppen. Die Gesamtverdaulichkeit von Leucin und Tyrosin erhöhte sich ebenfalls in der Apfelextraktgruppe verglichen mit den Fütterungsgruppen B und C. Der Zusatz der phytogenen Futterzusatzstoffe hatte keinen Einfluss auf die Aktivitäten der intestinalen Enzyme Laktase, Maltase, Saccharase, Aminopeptidase N und der alkalischen Phosphatase. Auch die antioxidative Kapazität im Blutplasma wurde durch die Futterzusätze nicht beeinflusst. Die elektrophysiologischen Messungen in der Ussingkammer zeigten keine Unterschiede zwischen den Fütterungsgruppen. Die Ermittlung der Genexpression mithilfe der PCR zeigte eine Verringerung der Expression der Saccharase in den Fütterungsgruppen B (Bockshornkleesamen) und C (Fresta F®) verglichen mit der Kontrollgruppe. Die Ergebnisse zeigen, dass der Einfluss auf die Adhäsion von E. coli und der IPEC-J2-Zelllinie für weiterführende Studien interessant sein könnte. Ein Einsatz entsprechender Zusatzstoffe könnte zur Vorbeugung bakterieller Durchfallerkrankungen in der Absetzphase bei Ferkeln sinnvoll sein.

In the present study, the influence of a phytogenic feed additive, two polyphenol-rich apple extracts and four phytogenic substances on the adhesion between an enteropathogenic E. coli and an IPEC-J2-cell line was examined. In a feeding trial, 24 female and 24 castrated male piglets (weaned at the age of 28 days) were assigned to four feeding groups in two successive series of experiments. The starter diet contained 0.2 % wheat bran in the control group A, 0.2 % fenugreek seeds in group B, 0,2 % of the phytogenic feed additive Fresta F® in group C and 0.2 % of a polyphenol-rich apple extract in group D. The zootechnical data were determined once a week. After a feeding period of four weeks, the piglets were euthanized and samples of blood, urine, digesta of the ileum and tissues of the jejunum were collected. In addition to the performance data, the apparent ileal digestibility and the total digestibility of crude protein as well as amino acids was determined. The tissues of the jejunum were used to isolate the brush border membranes to determine content of protein and the activity of the intestinal enzymes maltase, succrase, Laktase, aminopetidase n and the alkaline phosphatase. The gene expression of the intestinal enzymes maltase, succrase, Laktase, aminopetidase n and the alkaline phosphatase and the sodium-glucose transporter (SGLT-1) were determined in the tissue of the jejunum. Furthermore, electrophysiological measurements of jejunal tissues were conducted in the Ussing chamber. The antioxidative capacity of the blood plasma was established with the FRAP-and the TEAC-Assay. The results of the in vitro investigations showed significant reductions of the adhesion between E.coli and IPEC-J2-cells by addition of the Fresta F®, fenugreek seeds, pectin and both apple extracts. There was no significant reduction of the adhesion when using Glucoraphanin and Sulforaphan. The results of the feeding trial did not show any significant differences between the feeding groups in regard to the performance data. The analyses of the ileal digestibility of the amino acid valine in group B (fenugreek seeds) showed a significant decrease compared to the groups A (control) and D (apple extract). The ileal digestibility of isoleucine showed a significant increase in group D (apple extract) compared to group B (fenugreek seeds) and C (Fresta F®). The analyses of the total digestibility of the amino acids threonine, valine, isoleucine and lysine showed a significant increase in group D compared to the three other groups. The total digestibility of leucine and thyrosine showed a significant increase in group D compared to groups B and C. The additions of the phytogenic feed additives did not have any influence on the activities of the intestinal enzymes Laktase, maltase, succrase, aminopeptidase n and the alkaline phosphatase. The feed additives did not influence the antioxidative capacity of the blood plasma either. The electrophysiological measurements in the Ussing chamber also did not show any differences between the feeding groups. The determination of the gene expression using PCR showed a reduction of the gene expression of succrase in the treatment group B (fenugreek seeds) and C (Fresta F®) compared to the control group. The results assumed that the influence on the adhesion between E. coli and IPEC-J2 could be of interest for further investigations. The use of appropriate additives could be useful for the prevention of bacterial diarrhea in weaning piglets.
