Ex vivo und in vitro Studien zur Wirkung von Zylexis® auf die Immunantwort nach Impfung mit Resequin® NN plus unter besonderer Berücksichtigung der EHV-1 / -4 spezifischen Antikörperantwort, des Immunstatus und Zytokinmusters im Pferdeblut | Ex vivo and in vitro studies of the impact of Zylexis® on the immune response following vaccination with Resequin® NN plus with special consideration of the EHV-1/-4 specific antibody response, the immune status and the cytokine gene expression patterns in horse blood

Eine Infektion mit dem EHV-1/-4 beim Pferd kann mit schweren Krankheitssymptomen einhergehen und zu wirtschaftlichen Verlusten führen. Eine entscheidende Rolle in der Prophylaxe gegen EHV-1/-4 Infektionen nehmen die Impfstoffe ein. Die zurzeit auf dem Markt erhältlichen Impfstoffe stimulieren allerdings in der Regel nur die humorale Immunantwort. Ziel der vorgelegten Arbeit war es zu untersuchen, ob eine Verbesserung der humoralen und Stimulation der zellulären Immunantwort durch Kombination von Impfung und Immunstimulator Zylexis® möglich ist. Dazu wurde eine Gruppe von 20 Pferden in eine Zylexis®- (n = 11) und Placebo-Gruppe (n = 9) aufgegliedert. Die Tiere erhielten eine dreimalige Zylexis®- bzw. Placebo-Applikation im Abstand von 48 Stunden. Am Tag der dritten Zylexis®- bzw. Placebo-Injektion wurden die Pferde mit dem inaktivierten Kombinationsimpfstoff Resequin® NN plus vakziniert. Die Untersuchung der Immunantwort erfolgte zu 6 verschiedenen Zeitpunkten. 30 Tage vor Studienbeginn wurde ein Ausgangswert ermittelt. Die Entnahmen der Blutproben erfolgten 6 Stunden nach der zweiten Zylexis®-/Placebo-Applikation sowie nach der Verabreichung der dritten Zylexis®-/Placebo-Dosis zusammen mit der Impfung. Weitere Blutentnahmen fanden am 3., 7. und 14. Tag nach der Vakzinierung statt. Zu jedem Untersuchungszeitpunkt wurden die Antikörpertiter gegen EHV-1 und EHV 4 im Neutralisationstest (NT) und im indirekten Immunfluoreszenztest (IFT) gemessen sowie die Anzahl der peripheren mononukleären Blutzellen und die Expression der Zytokingene von IFN-γ, IL 2, IL-4 und IL-10 ermittelt. An zwei Untersuchungszeitpunkten (Tag -30 und 0) wurde der Immunstatus über die Firma LABOLKIN bestimmt. Im Vordergrund stand die Detektion von ausgewählten Zytokinexpressionsmustern mittels quantitativer „Real-Time“ TaqMan®-PCR. Als Parameter der zellulären Immunantwort wurde die Expression von IFN-γ und IL-2 untersucht und als Parameter der humoralen Immunantwort die von IL-4 und IL-10. Der Zytokin-mRNA-Nachweis mittels TaqMan®-PCR erfolgte direkt aus dem Pferdeblut (ex vivo) und vergleichend in vitro nach 24-stündiger Stimulation mit Concavalin A. Dabei wurde festgestellt, dass ex vivo Untersuchungen der Zytokin-mRNA mittels TaqMan®-PCR ausreichen, um Konzentrationsunterschiede darzustellen. Die Ergebnisse zeigten, dass die Anwendung von Zylexis® in Kombination mit dem Impfstoff Resequin® NN plus zu einer signifikanten Erhöhung der Anzahl von equinen PBMCs geführt hat. Darüber hinaus spiegelt die Induktion der Genexpression von IL-4 und IL-10 nach der Impfung sowohl bei den Zylexis®-behandelten Tieren als auch bei den Placebo-behandelten Pferden eine Aktivität der humoralen Immunantwort wieder. Diese korreliert mit dem Anstieg der EHV-1/-4 Antikörpertiter. So konnte ein Antikörperanstieg bei 19 (95%) Pferden im IFT und bei 7 (35%) Pferden im NT gezeigt werden. Der Nachweis von Virus neutralisierenden Antikörpern gelang vor allem bei Pferden, die ein Alter von mindestens über 4 Jahren hatten. Zwischen den Versuchsgruppen Zylexis® und Placebo konnten keine Unterschiede bezüglich der Höhe der virusspezifischen Antikörpertiter und der Zytokinexpression ermittelt werden. Eine Induktion von IFN-γ und IL-2 konnte in der vorliegenden Arbeit weder durch den Einsatz von Zylexis®, noch durch den Impfstoff allein festgestellt werden. Zusammenfassend lässt sich festhalten, dass nach Impfung mit Resequin® NN plus die Induktion der humoralen Immunantwort mit Anstieg der EHV-1/-4 Antikörpertiter und Expressionszunahme von IL-4 und IL-10 nachweisbar war. Durch Zylexis® konnte hierbei kein synergistischer Effekt erzielt werden. Eine nachweisliche Induktion der zellulären Immunantwort gemessen an der Expression von IFN-γ und IL-2 konnte weder durch den Impfstoff allein, noch in Kombination mit Zylexis® festgestellt werden. Zylexis® bewirkte allerdings in Kombination mit der Impfung eine signifikante Erhöhung der Anzahl von equinen PBMCs.

EHV-1/-4 infections in horses not only cause severe clinical symptoms, but also economical losses. Vaccination plays an important role in preventing EHV-1/-4 infections. Vaccines currently available on market usually only stimulate the humorale immune response. The aim of the present study was to evaluate whether it is possible to improve the humoral immune response and whether the cellular immune response could be stimulated by the combination of the inactivated vaccine Resequin® NN plus and the immune modulator Zylexis®. A group of 20 horses were divided into a Zylexis-group (n = 11) and a Placebo- group (n = 9). The animals of both groups received three applications of Zylexis® or a Placebo respectively at 48 hour intervals. The horses were vaccinated with the inactivated combination vaccine Resequin® NN plus simultaneously to the third Zylexis®-/Placebo application. The examination of the immune response was carried out at 6 different time points. Initial values were determined 30 days before the beginning of the study. Blood samples were collected 6 hours after the second and third combined application of Zylexis®-/Placebo and Resequin® NN plus. Further blood samples were collected on days 3, 7 and 14 after vaccination. At every time point of investigation the antibody titer against EHV-1 and EHV-4 was measured with neutralization test and indirect immunofluorescence test, the number of peripheral mononuclear blood cells and the cytokine gene expression of IFN-γ, IL-2, IL-4 and IL-10 were examined. The immune status was determined at two time points (day -30 and day 0) by LABOKLIN. Foremost we studied the gene expression of selected cytokines using quantitative “real-time” TaqMan®-PCR. For the cellular immune response the gene expression levels of IFN-γ and IL 2 and for the humoral immune response the levels of gene expression of IL-4 and IL-10 were used as parameters. The cytokine mRNA was analyzed with TaqMan®-PCR directly from the horses’ blood (ex vivo) and for comparison after stimulation with Concavalin A for a period of 24 hours (in vitro). We noticed that in vivo examinations of cytokine mRNA using TaqMan®-PCR is sufficient to detect differential gene expressions. The results showed that the administration of Zylexis® in combination with the vaccine Resequin® NN plus resulted in significantly increased numbers of equine PBMCs. Further more the induction of the gene expression of IL-4 and IL-10 after vaccination equally mirrored an activity of the humoral immune response in the Zylexis®- and Placebo-treated horses. These findings correlate with the serological reactions of EHV-1/-4 antibodies. Thus the IFT and NT antibody titers increased in 19 (95%) animals and 7 (35%) horses respectively. The virus neutralizing antibodies could be detected especially in horses aged at least 4 years. No significant differences regarding the level of virus specific antibody titers and cytokine gene expression were noted between the Zylexis- and Placebo-groups. An induction of the IFN-γ and IL-2 in the present study could neither be detected with Zylexis® nor using the vaccine alone. In conclusion after vaccination with Resequin® NN plus an induction of the humoral immune response was detectable shown as increased EHV-1/-4 antibody titers and increased gene expressions of IL 4 and IL-10. No synergistical effect was achieved by Zylexis®. A detectable induction of the cellular immune response as measured by the gene expressions of IFN-γ and IL-2 was neither observed when applying Zylexis® alone, nor Resequin® NN plus nor using both in combination. However Zylexis® caused a statistically significant increase in equine PBMC number in combination with the vaccine.
