Vergleichende molekulare und funktionelle Analyse des „Locus of Enterocyte Effacement“ (LEE) im bovinen EHEC-Stamm RW1374 (O103:H2) | Molecular and functional comparative analysis of the “Locus of Enterocyte Effacement” (LEE) of bovine EHEC strain RW1374 (O103:H2)

Humane Infektionen mit enterohämorrhagischen E. coli (EHEC) der Serovar O103:H2 gewinnen in Deutschland zunehmend an Bedeutung. Da Wiederkäuer als transientes EHEC Reservoir mehrheitlich für die humanen Infektionen verantwortlich sind, haben wir die DNS einer Pathogenitätsinsel (PAI) des bovinen O103:H2 Stamms RW1374 analysiert, um putative Virulenzmerkmale zu identifizieren. Diese komplex zusammengesetzte PAI beinhaltet die Kernregion des „Locus of Enterocyte Effacement“ (LEE) und ein Homolog der genetischen O-Insel (OI-) 122, bestehend aus den virulenzassoziierten Genen efa1/lifA, ent, nleE, nleB und einigen mobilen Elementen. Sie hat eine Gesamtlänge von 111 kb und ist im pheV-tRNS-Locus inseriert. Die Kombination mehrerer komplexer Virulenzmerkmale könnte teilweise die hohe Virulenz der EHEC Serovar O103:H2 für den Menschen erklären. Beim Vergleich der PAI mit homologen Loci von humanen EPEC (O127:H6) und EHEC (O157:H7; O26:NM) sowie Kaninchen- spezifischen EPEC (O103:H2; O15:H-) zeigte sich die LEE-Kernregion weitgehend konserviert, während die OI-122 deutliche Unterschiede aufwies. In der LEE- Kernregion des RW1374 fehlen der Orf3 und das ERIC-Element, die Gene für EspF und Ler weichen von den Homologen der anderen Stämme ab. Der OI-122 fehlt das pagC Gen, das efa1/lifA Gen wird von zwei Insertionssequenzen (IS) flankiert und das Integrasegen am 5’-Ende der PAI durch ein IS629 Element zerstört. Um nähere Aufschlüsse über die Verbreitung der OI-122 zu erhalten, wurden 268 intestinale E. coli-Isolate mittels PCR und DNS-DNS-Hybridisierung auf das Vorkommen der Gene pagC, ent und efa1/lifA bzw. efa1’ untersucht. Während das trunkierte efa1’ Gen ausschließlich in EHEC der Serovar O157:H7 vorkam, wiesen 27 % der Isolate verschiedener Serovare das efa1/lifA auf. In drei Stämmen fanden wir Hinweise auf neuartige efa1/lifA Genvarianten. Das Gen ent besaßen 44 % und das Gen pagC 22 % der überprüften E. coli. Zwei oder drei der Gene identifizierten wir ausschließlich in Attaching-and-Effacing (A/E-) Läsionen- verursachenden E. coli (AEEC). Von den 164 AEEC hatten 31 % eine vollständige und 26 % eine unvollständige Version der OI-122. Die nicht-AEEC wiesen maximal eines der Gene auf. Die Ergebnisse weisen auf eine deutliche Assoziation zwischen der genetischen Insel und dem LEE hin. In einigen Stämmen konnten wir eine direkte Nachbarschaft der beiden Inseln feststellen. Die Ermittlung effizienter Strategien zur Minimierung der Infektionsgefahr mit EHEC und zur Bekämpfung der Verbreitung setzt möglichst genaue Kenntnisse über die Wirkungsweise der involvierten Virulenzfaktoren voraus. Aus diesem Grund wurden die Proteine Efa1/LifA und Ent des EHEC RW1374 weiterführend untersucht. In der Proteinsequenz des Efa1/LifA waren Homologien zu verschiedenen Toxinen (ToxB der O157:H7 EHEC, LCT der Clostridien spp., putatives Zytotoxin von Chlamydia caviae) und Adhäsinen (TC0437 von Chlamydia muridarum, YopT von Yersinia spp., PvRBP2 und Rhop von Plasmodium spp., Interaptin von Dictyostelium discoideum) sowie einige Protein-Motive zu finden. Die Expression von Efa1/LifA konnten wir mittels RT-PCR nachweisen. Trotz einiger Probleme bei der Klonierung und Kultivierung ließen sich zwei interne Proteinfragmente (LifA2 und LifA3) fremdexprimieren, aufreinigen und zur Generierung polyklonaler Antikörper verwenden. Während im Immunoblot keine Detektion des Efa1/LifA im Wildstamm nachzuweisen war, konnten wir im Zellkulturverfahren eine spezifische Anfärbung der Bakterienhülle des adhärenten EHEC-Stamms RW1374 mittels Immunfluoreszenz und konfokaler Laserscan Mikroskopie sichtbar machen. Der virulenzassoziierte Faktor Ent zeigte Sequenzhomologien zu den Enterotoxinen von enteroinvasiven E. coli (EIEC)/Shigella spp. (ShET-2) sowie Yersinia spp. (SenA) und wurde nur unter Eisenmangel-Bedingungen exprimiert. Eine Fremdexpression des Ent gelang uns nicht, was wir auf eine mögliche Toxizität zurückführten.

E. coli (EHEC) strains of serotype O103:H2 are increasingly associated with human infections in Germany. As ruminants have been implicated as the main reservoir and source of human infections, the DNA of a pathogenicity island (PAI) of the bovine O103:H2 strain RW1374 was analysed in regard to the presence of putative virulence factors. This complex PAI spans over 111 kb and comprises the core-region of the "locus of enterocyte effacement" (LEE), and a homologue of the O-island (OI-) 122, including its virulence-associated genes efa1/lifA, ent, nleE, nleB and some mobile elements. It is inserted in the pheV-tRNA-locus. The high level of pathogenicity EHEC serovar O103:H2 causes in human infections could be attributed to this combination of multiple complex virulence factors. Comparison with homologue loci of human EPEC (O127:H6) and EHEC (O157:H7; O26:NM), and rabbit-specific EPEC (O103:H2; O15:H-) respectively, revealed the LEE core-region of the PAI to be largely conserved, whereas the OI-122 differed considerably. Regarding the LEE core- region, RW1374 lacks the orf3 and ERIC (enteric repetitive intergenic consensus) element present in the other strains, and showed variations of genes encoding EspF and Ler. The OI-122 of RW1374 lacks the pagC gene, it's efa1/lifA gene is flanked by two insertion sequences (IS) and the integrase gene at the 5' terminus of the PAI is disrupted by an IS629 element. To clarify the distribution of OI-122, we performed PCR and DNA-DNA hybridization assays on 268 intestinal E. coli isolates in search for the genes pagC, ent and efa1/lifA or efa1', respectively. While the truncated efa1' gene was exclusively present in EHEC O157:H7, the efa1/lifA homologue was detected in 27 % of isolates belonging to different serovars. Three isolates revealed new variations of efa1/lifA genes. In 44 % of the E. coli strains we detected gene ent, and in 22 % gene pagC. A combination of two or three of these genes occurred only in attaching-effacing E. coli (AEEC). Among the 164 AEEC isolates, 31 % contained a complete and 26 % an incomplete version of OI-122. In non-AEEC isolates we found no more than one of these genes. Our results suggest a strong association between the two islands, and for some isolates they could be demonstrated to be located in immediate genetical neighbourhood. The design of efficient strategies apt to minimize the risk of infection and to control the spreading requires an exact knowledge about the mode of action of the involved virulence factors. Thus, the proteins Efa1/LifA and Ent of RW1374 were studied in more detail. The protein sequence encoding Efa1/LifA showed homologies to different toxins (ToxB of EHEC O157:H7, LCT of Clostridium spp., putative cytotoxin of Chlamydia caviae) and adhesins (TC0437 of Chlamydia muridarum, YopT of Yersinia spp., PvRBP2 and Rhop of Plasmodium spp., Interaptin of Dictyostelium discoideum), as well as to some protein motives. Using RT-PCR we could provide evidence for the expression of Efa1/LifA. In spite of some difficulties concerning cloning and cultivation, two internal protein fragments (LifA2 and LifA3) could be overexpressed, purified and used for preparation of polyclonal antibodies. While no detection of Efa1/LifA could be achieved using immunoblot assays on the wild-type, in in vitro assay of bacterial adherence a specific stain of the bacterial wall of adherent EHEC RW1374 was yielded using immunofluorescens and confocal laserscan microscopy. The virulence associated factor Ent was found to be characterized by sequence-homologies to enterotoxins of enteroinvasive E. coli (EIEC)/Shigella spp. (ShET-2) and Yersinia spp. (SenA). It was exclusively expressed in iron-deficiency situations. Foreign expression of Ent could not be realized, which we attributed to an assumed toxicity.
