Makroskopische, mikroskopische und morphometrische Vergleichsstudie des Gastrointestinaltraktes von kommerziellen B.U.T. Big 6 Puten und Wildputen | Macroscopic, microscopic and morphometric comparative study of the gastrointestinal tract of commercial B.U.T. Big 6 turkeys and wild turkeys

Die Pute ist eines der wichtigsten praxis- und wirtschaftsrelevanten Geflügelarten. Erkrankungen des Magen-Darm-Traktes und die Effizienz der Verdauung haben in der Geflügelzucht eine hohe wirtschaftliche Bedeutung. In der Literatur findet man wenig über die Anatomie, Histologie und das Wachstum des Gastrointestinaltraktes unter dem Einfluss der Domestikation. Durch genetische Selektion der Hausputen änderte sich sowohl die Wachstumsgeschwindigkeit als auch die Wachstumsstruktur der Tiere. Folgend wurde eine zusammenhängende anatomische, histologische und morphometrische Darstellung des Magen-Darm-Traktes einer modernen selektierten Hausputenlinie (British United Turkeys, B.U.T. Big 6) mit einer nicht selektierten Wildform (Wild Canadian Turkeys, WCT) im Vergleich verschiedener Altersstadien erstellt. Es wurden dazu 25 Hausputen sowie 48 kanadische Wildputen als nicht geschlechtlich vorsortierte Eintagsküken eingestallt. Die Gruppe der Hausputen wurde an 5 Terminen mit je 5 Tieren im Alter von 4, 8, 12, 16 und 20 Lebenswochen anatomisch beurteilt, fotografiert, beprobt und die Organlängen vermessen. Die Gruppe der Wildputen wurde mit je 8 Tieren pro Termin ebenfalls im vierwöchigen Intervall, beginnend mit der 4. Lebenswoche und einer zusätzlichen Gruppe im Alter von 24 Wochen anatomisch beurteilt, fotografiert und beprobt. Von je 5 Tieren wurden die Organlängen vermessen. Anatomisch und histologisch können bei beiden Putenrassen bis auf die Zunge und den subserösen Fettansatz keine nennenswerten Unterschiede detektiert werden. Die Wildputen weisen längere und schlankere Zungen mit zahlreicheren und spitzeren Papillen auf und lagern mehr subseröses, perivaskuläres Fettgewebe ein als die Hausputen. Bei den morphometrischen Vermessungen bestätigen sich die erwartet homogen höheren Körpermassen der Hausputen im Vergleich zu den geringeren und breiter gestreuten Körpermassen der Wildputen. Die Vermessung des Magen-Darm- Traktes zeigt korrespondierende Werte. Die Hausputen der vorliegenden Untersuchung weisen alle ein kontinuierliches, rasches Darmwachstum auf. Die Darmlänge im gesamten, sowie auch die einzelnen Darmabschnitte, sind in allen Altersstadien länger als die der Wildputen. Bei den Hausputen korreliert die Länge des Dünndarmes, der Caeca und des Colorectums stark mit der Körpermasse. Bei den Wildputen korreliert nur die Länge der Dünndarmabschnitte mittel mit der Körpermasse. Das Duodenum weist bei beiden Putenlinien die längsten Zotten des Dünn- und Dickdarmes auf. Im Dünndarm verändert sich der Vergrößerungsfaktor durch Zotten im Altersverlauf: die Wildputen bilden in den ersten 8 Wochen eine größere Oberfläche durch Zotten als die Hausputen, danach stagniert dieser Faktor. Bei den Hausputen hingegen steigt der Wert kontinuierlich an. Die Wildputen zeigen in beiden vermessenen Dünndarmabschnitten in allen Altersstadien einen geringgradig größeren Anteil der Oberflächenvergrößerung durch Krypten als die Hausputen. Im Dickdarm gibt es deutliche Unterschiede in den Zottenlängen sowohl innerhalb seiner einzelnen Abschnitte als auch im Vergleich zum Dünndarm. Die Zotten im Caecum sind bei beiden Spezies deutlich kürzer als die Zotten der Dünndarmabschnitte. Im Corpus caeci haben beide Putenrassen aller Altersstadien längere Zotten als im Apex caeci. Im kürzesten Darmabschnitt, dem Colorectum, sind die Zotten bei beiden Spezies kürzer als im Dünndarm und im Corpus caeci aber länger als im Apex caeci. Die Höhe der Enterocyten ist bei beiden Putenarten in allen Altersstadien im Dünn- und im Dickdarm ähnlich. Bei beiden Spezies ist der Darm mit Lymphgewebe durchsetzt, die Wildputen weisen ein diffuses Verteilungsmuster auf, bei den Hausputen formieren sich die Zellen eher zu Haufen. Fasst man diese Ergebnisse zusammen, lässt sich vermuten, dass bei gleicher Fütterung eine genetische, durch Domestikation und Züchtung geprägte, höhere Wachstumsrate mit inem längeren Darm und längeren Zotten assoziiert ist: der Darm der Hausputen ist grundsätzlich länger als der der Wildputen; mit 20 Wochen um das 1,7fache, die Zotten im Dünndarm um das 1,4fache. Die Morphometrie der für die Zellerneuerung wichtigen Krypten und der absorptiv tätigen Epithelzellen der Zotten zeigt derartig starke Unterschiede zwischen den beiden untersuchten Spezies jedoch nicht auf. Die weitestgehende Konstanz der Kryptentiefe und der Epithelzellenhöhe im Altersgang zwischen der 4. und 20. (Hausputen) bzw. 24. (Wildputen) Lebenswoche entspricht scheinbar einem physiologischen Optimum, das bei den Epithelzellen der Zotten bei ca. 40 μm, bei den Krypten im Bereich zwischen 120 μm (Hausputen) und 150 μm (Wildputen) liegt.

Turkeys are one of the most important and relevant poultry species in veterinary medicine and economy. Gastrointestinal diseases as well as digestive efficiency are of high economic significance in poultry breeding. In literature, there is little to find concerning the anatomy, histology and the growth of the gastrointestinal tract under the influence of domestication. Due to genetic selection of domesticated turkeys, changes in both the animal’s growth rate and in their growth structure occurred. In this study, a continuous anatomical, histological and morphometrical investigation of the gastrointestinal tract of a modern selected turkey breeding line (British United Turkeys, B.U.T. Big 6) in comparison with a none selected wild turkey line (Wild Canadian Turkeys, WCT) at different stages of age is presented. For this purpose, 25 domesticated and 48 Canadian wild day-old turkey chicks of random gender were housed. 5 animals of the domesticated turkey group and 8 animals of the wild turkey group were anatomically evaluated, photographed and sampled in a 4 week interval at the age of 4, 8, 12, 16 and 20 weeks. In the wild turkey group, anatomical evaluation, photography and sampling was additionally carried out at the age of 24 weeks. In 5 animals of each group, the organ length was also measured on the particular examination dates. Comparing anatomical and histological findings in the examined turkey breeds, differences are found merely in the amount of subserous adipose tissue and in the morphological appearance of the animal’s tongues. The tongues of wild turkeys are longer, slimmer and have more pointed lingual papillae. Wild turkeys also show a higher storage of subserous perivascular fatty tissue compared to domesticated turkeys. Morphometric measurements confirm a homogeneously higher body mass in domesticated turkeys compared to the generally lower body mass of wild turkeys – spread over a wider range within the group. Measurements of the gastrointestinal tract reveal analogous findings: the domesticated turkeys in this study show a continuous rapid growth of their intestinal tract. The complete intestinal tract and the individual intestinal segments are longer at each investigated age than in wild turkeys. In domesticated turkeys, the length of duodenum and jejunoilium, of caeca and colorectum correlates significantly with their body mass. In wild turkeys, only the length of the small intestinal segments shows a medial correlation with their body mass. The duodenum shows the longest villi of the intestinal system in both examined turkey groups. In the small intestine, the ‘growth factor attributable to villi’ changes with the progression of age: compared to domesticated turkeys, wild turkeys form a larger surface by small intestinal villi during their first 8 weeks of age, subsequently this factor stagnates. In domesticated turkeys however, this parameter increases continuously. In the two measured small intestinal segments, wild turkeys show a slightly higher ratio of surface increase by crypts than domesticated turkeys. In the large intestine, significant differences of villi length within the individual segments, as well as in comparison to the small intestine can be noted. Villi of the caecum appear considerably shorter than villi of the small intestinal segments in both turkey species. In the corpus of the caecum, both species at all ages show longer villi than in the apex caeci. In the shortest intestinal segment, the colorectum, villi are shorter than in the small intestine and in the corpus of the caecum, but longer than in the apex caeci in both turkey species. The height of the enterocytes in the small and large intestine is similar in both turkey species at all ages. In both species, the intestine is interstratified with lymphatic tissue. Wild turkeys show a diffuse distribution pattern of lymphatic cells, whereas in domesticated turkeys the cells tend to form clusters. In summary, the results of this study lead to the assumption that, feeding on an identical diet, a genetic higher growth rate, characterized by domestication and breeding, is associated with an increased length of intestines and villi: the intestines of domesticated turkeys are generally longer than those of wild turkeys; at 20 weeks of age by the factor 1.7, and the small intestinal villi by the factor 1.4. However, morphometric evaluation of the crypts, which play an essential role in cell replacement, and of the epithelial cells of the villi, which regulate absorption, does not reveal marked discrepancies between the two examined species. The predominant consistency of crypt depths and height of epithelial cells at the age of 4 to 20 weeks in domesticated turkeys, respectively 4 to 24 weeks in wild turkeys, seems to correlate with a physiological optimum, which is approximately 40 μm in the epithelial cells of the intestinal villi, and a range of between 120 μm (domesticated turkeys) and 150 μm (wild turkeys) for the crypts.
