Der Einfluss von Zink als Futterzusatzstoff auf die porzine intestinale Escherichia coli-Population beim Ferkel | The influence of zinc as a feed additive on the porcine intestinal Escherichia coli population in piglets

Seit 2006 sind antimikrobielle Fütterungszusatzstoffe in der Europäischen Union (EU) verboten. Um das Auftreten der verlustreichen Post-weaning Diarrhea (PWD) in der Ferkelaufzucht zu reduzieren, welche vor allem durch enterotoxische E. coli (ETEC) hervorgerufen wird, werden bislang verschiedene Strategien angewendet. Hierzu zählt die zum Teil exzessive Fütterung von Zinkoxid, welches nach Magenpassage als zweiwertiges Kation vorliegt (Zn2+). Gene, die Resistenzen gegen kationische Spurenelemente vermitteln, sind häufig mit antimikrobiellen Resistenzgenen co-lokalisiert. Daher wurde in früheren Studien die Hypothese aufgestellt, dass Zink-Fütterung eine der Ursachen für die Ausbreitung resistenter Bakterien darstellt. Bis heute sind die Effekte der zinkreichen Fütterung auf die intestinale Bakterienpopulation beim Schwein, v.a. auf die E. coli-Population, nicht vollständig geklärt. Vor diesem Hintergrund wurden in dieser Arbeit die phänotypischen und genotypischen Charakteristika einer re-präsentativen Auswahl von E. coli-Isolaten zur Aufklärung des Einflusses von Zink als Futterzusatzstoff auf die intestinale E. coli-Population beim Schwein untersucht. Insgesamt wurden 179 E. coli-Isolate aus einer Zinkfütterungsstudie mit Absatzferkeln betrachtet. 99 Isolate wurden aus der Zinkfütterungsgruppe (high-zinc fed group [HZG]) und 80 Isolate aus der Kontrollgruppe (control group [CG]) des letzten Beprobungstages (52. Tag) zufällig ausgewählt. Alle Isolate wurden auf Zinktoleranz, antimikrobielle und Schwermetall-Empfindlichkeiten geprüft. Dazu wurden minimale Hemmkonzentrationen (MHK) durch ein Mikrodilutionsverfahren ermittelt. Darüber hinaus wurden Ganzgenomanalysen zur Detektion von antimikrobiellen Resistenz-, Schwermetalltoleranz- und Biozidresistenzgenen sowie Viru-lenz-assoziierten und Bakteriozin-assoziierten Genen über eine in-house BLAST-basierte Pipeline (Treffer bei ≥ 90% Sequenzähnlichkeit) durchgeführt. Auf Grundlage eines Maximum Common Genome Alignements aller Isolate wurde ein phylogenetischer Baum erstellt. Um genetische Unterschiede der Abstammungslinie zu identifizieren, wurde eine Bayesian Analysis of Population Structure angewandt. In dieser Studie wurden drei verschiedene Zinkchlorid- (ZnCl2) MHK-Werte bei E. coli ermittelt: (1) 128 µg/ml (HZG: 2%, CG: 6%), (2) 256 µg/ml (HZG: 64%, CG: 91%) und (3) 512 µg/ml (HZG: 34%, CG: 3%; p < 0,001). Die enge Verteilung der ZnCl2-MHK-Werte lässt auf eine natürlich vorkommende E. coli-Population schließen. Zwischen höheren ZnCl2 MHK-Werten und Isolaten der Zinkfütterungsgruppe sowie niedrigen ZnCl2 MHK-Werten mit Isolaten der Kontrollgruppe bestand eine statistisch signifikante Assoziation (lineares gemischtes Regressionsmodell, p = 0,005). Dieses Ergebnis kann auf einen Selektionsvorteil ver-schiedener kommensaler E. coli-Populationen in Anwesenheit hoher Zinkmengen im Schweinefutter hinweisen. Die Zinkchlorid-Toleranzwerte in dieser Studie zeigen jedoch keinen statistischen Zusammenhang zum Auftreten eines Antibiotikaresistenz-Phänotyps. Isolate der höheren ZnCl2 MHKs von 512 µg/ml wiesen zum Teil (n=19/36) zwei neue F (engl. Fertility) -Plasmide auf, die neben diversen antimikrobiellen Resistenzgenen (aadA1, dfrA1, sul1, sul3 und tetA) auch Schwermetalltoleranzgene (merRTPCDAB und arsRP) sowie Biozidresistenzgene (qacL und unterbrochenes qacE [qacEΔ1]) beinhalteten. Zink-resistenzgene, die für andere Spezies bereits beschrieben sind, wurden auf den Plasmiden nicht lokalisiert. Die bisher publizierte Literatur hatte verschiedene Gene als verantwortlich für eine Zinkresistenz gesehen, einschließlich zitB und zntA, die für Zink-Exporter codieren, und pit, welches für einen unspezifischen anorganischen Phosphat-Transporter codiert. Ganzgenom-Untersuchungsdaten von Genen, die an der Zinkhomöostase beteiligt sind, offenbaren ein vollständiges Vorhandensein nahezu aller bislang genannten Gene und Faktoren in den E. coli-Isolaten dieser Studie, ein Zinkresistenzgen wurde nicht identifiziert. Die phylogenetischen Analysen zeigten keine Assoziation zwischen den ermittelten Zinkchlorid-Toleranzwerten und der phylogenetischen E. coli-Populationsstruktur in der Studie. Die Bayesian Analysis of Population Structure-Cluster spiegelten ausschließlich die Sequenz- resp. Serotypen wider. Das Ergebnis weist darauf hin, dass eine Zinktoleranz keine Eigenschaft eines bestimmten phylogenetischen Hintergrundes ist. Insgesamt traten weniger Virulenz-assoziierte Gene (41% vs. 65% in Isolaten der Kontrollgruppe, p < 0,001) und Bakteriozin-assoziierte Gene (22% vs. 35% in Isolaten der Kontrollgruppe, p = 0,002) in Isolaten der Zinkfütterungsgruppe auf. Hier zeichnet sich ein Effekt der Zinkfütterung auf das Vorkommen dieser Gene in der intestinalen E. coli-Population ab. Die Verteilung bestimmter Virulenzdeterminanten wies auf deutliche Unterschiede im Auftreten verschiedener assoziierter E. coli-Pathotypen (enterotoxische E. coli [ETEC], enteropathogene E. coli [EPEC], Shigatoxin bildende E. coli [STEC]) in beiden Fütterungs-gruppen hin (1% HZG, 14% CG). Zink scheint nicht nur die intestinale Zusammensetzung von porzinen E. coli-Populationen hinsichtlich der Phänotypen mit erhöhter Zinktoleranz zu be-günstigen, sondern auch die Populationsdichte potenziell pathogener Isolate zu verringern. Weitere Untersuchungen über die Mechanismen, die an der Entwicklung der Zinktoleranz in E. coli beteiligt sind, müssen folgen, insbesondere Forschungsarbeiten zur zinkinduzierten Stressantwort durch vergleichende Transkriptom-Analysen.

In 2006, the European Union banned the use of antimicrobial growth promoters in animal production. Since then, different alternative strategies were used to reduce the economic losses due to post-weaning diarrhea (PWD) associated with particularly enterotoxigenic Escherichia (E.) coli (ETEC) in piglet farming. One of the most prominent strategies is supplementation of pig fed with zinc oxide (ZnO). The low pH in the stomach allows the formation of the divalent cation as a soluble salt (Zn2+). Of note, genes conferring resistance against cationic trace elements are often co-localized with genes conferring antimicrobial resistance in bacteria. Previous studies hypothesized that zinc feeding selects for antimicrobial resistant bacteria due to co-resistances harbored by on mobile genetic elements. However, effects of high-zinc diets on porcine intestinal bacterial populations, especially E. coli, are not fully understood yet. Therefore, the aim of this study was the phenotypic and genotypic characterization of a representative selection of E. coli isolates to clarify the influence of zinc as feed additive on the porcine intestinal E. coli population. In total, 179 E. coli isolates, consisting of a group of 99 high-zinc fed (HZG) isolates and a corresponding control group (CG) of 80 isolates were collected from piglets sacrificed at the end of a Zn feeding trial (52nd day). All isolates were screened for zinc tolerance, antimicrobial- and biocide susceptibilities by determination of minimum inhibitory concentrations (MICs). In addition, in silico whole genome screening (WGS) of all isolates for genes conferring antimicrobial resistance, heavy metal tolerance and biocide resistance was carried out. In addition, virulence-associated- and bacteriocin-associated genes were identified using an in-house BLAST-based pipeline (hits based on ≥ 90% sequence identity). The maximum common genome alignment was used to generate a phylogenetic tree. Bayesian analysis of population structure was applied to identify genetically distinct lineages based on the constructed phylogeny. In this study, porcine E. coli yielded three different levels of zinc chloride (ZnCl2) MIC: 128 µg/ml (HZG, 2%; CG, 6%), 256 µg/ml (HZG, 64%; CG, 91%) and 512 µg/ml (HZG, 34%; CG, 3%; p < 0.001). The close distribution of ZnCl2 MICs seems to reflect a naturally occurring E. coli population. A statistically significant association was revealed between “higher” ZnCl2 MIC values and isolates representing high zinc-fed group and “lower” ZnCl2 MIC values with isolates of the control group (linear mixed regression model, p = 0.005), indicating a selective advantage of distinct commensal E. coli populations in the presence of high amounts of zinc in the piglets’ diet. The different zinc tolerance levels investigated in this study lacked a particular association with the occurrence of an antibiotic resistance phenotype profile. The isolates with the 512 µg/ml ZnCl2 MIC particularly harbored two novel F (Fertility) plasmids (n= 19/36) carrying antimicrobial resistance genes (aadA1, dfrA1, sul1, sul3 and tetA), heavy metal tolerance genes (merRTPCDAB and arsRP) and biocide resistance genes (qacL and disrupted qacE [qacEΔ1]). Zinc resistance genes that had been previously detected among other species were not identified on the plasmids. WGS data screening for genes involved in zinc homeostasis revealed an almost ubiquitous presence of most factors tested, including zitB, zntA both encoding zinc exporters and pit encoding unspecific inorganic phosphate transporter formerly described as important for zinc resistance. The phylogenetic analysis showed no association between zinc tolerance levels and the phylogenetic E. coli population structure in the study. The Bayesian Analysis Population Structure cluster reflected just the sequence resp. serotypes. The results suggested that zinc tolerance lacks a particular association with a specific phylogenetic background. Overall, fewer virulence-associated genes (41% vs. 65% in isolates from control group, p<0.001) and bacteriocin-associated genes (22% vs. 35% in isolates from control group, p=0.002) were detected in isolates from the high-zinc fed group. The results indicate an effect of high-zinc supplementation of the piglets’ diet on the occurrences of these genes among intestinal E. coli populations: The distribution of typical virulence determinants demonstrated obvious differences in the occurrence of distinct E. coli pathotypes (enterotoxigenic E. coli [ETEC], enteropathogenic E. coli [EPEC] and shiga toxin-producing E. coli [STEC]) in both feeding groups (1% in HZG, 14% in CG). Zinc seems not only to favor the intestinal composition of porcine E. coli population regarding the phenotypes with increased zinc tolerance, but also to reduce the abundance of potentially pathogenic isolates. Further research on mechanisms involved in development of zinc tolerance in E. coli is needed, especially regarding the zinc-induced stress response, for instance by employment of comparative transcriptomics.
