Embriogênese em Ilex paraguariensis St. Hil. : aspectos do suspensor e endosperma

Estudos realizados no gênero flex, família Aquifoliaceae, ordem Celastrales, mostram que embriões pertencentes a este gênero permanecem rudimentares, em estágio de coração, quando os frutos estão maduros, sendo que as sementes necessitam de um período de 6 a 8 meses para germinar, com uma taxa de germinação muito baixa. As características apresentadas fazem com que a produção de mudas de erva-mate seja lenta e a quantidade não suficiente para a demanda. A análise morfoanatômica do embrião e endosperma de flex paraguariensis, durante a embriogênese tardia, mostram a presença de suspensor, mesmo em fases mais avançadas. Apresenta também modificações nas reservas do endosperma nessas fases. Tendo por base essa análise, o presente trabalho objetiva apresentar um estudo criterioso do suspensor desta espécie, desde sua formação, até sua degradação durante a embriogênese final, bem como do embrião e endosperma, na embriogênese. As diferentes fases da embriogênese e endospermogênese foram analisadas e descritas utilizando métodos anatômicos tradicionais, para microscopia óptica, microscopia eletrônica de varredura e de transmissão. Acompanhando o desenvolvimento ontogenético desta espécie, o trabalho caracteriza o ciclo reprodutivo, as flores pistiladas, o processo de fecundação, os frutos, bem conío o embrião e o endosperma. O endosperma é classificado como celular, segundo o sistema EODP. O tipo de divisão celular, que ocorre na embriogênese inicial, permite classificar o desenvolvimento embrionário como pertencente ao tipo Cariofiláceo. Durante a embriogênese inicial, as fases analisadas vão desde a formação do zigoto, passando por proembrião bicelular até proembrião pós-octante. Durante a embriogênese final, as fases analisadas vão desde embrião piriforme até embrião em estágio de torpedo (maduro), quando ocorre a germinação. O aporte de nutrientes ao embrião esporofitico, nas fases iniciais de seu desenvolvimento, ocorre predominantemente via suspensor, pois o embrião xenofitico não está totalmente desenvolvido. Quando o embrião xenofitico apresenta as reservas estruturadas, a utilização destas é observada através das modificações de suas características morfológicas que obedecem a um gradiente, tanto no sentido micrópila-calaza como no sentido tegumento-embrião. Suas variações de síntese, absorção e translocação morfologicamente são: (1) formação de vesículas de membrana simples que se fundem formando vacúolos; (2) deposição de proteína amorfa no seu interior; (3) surgimento de adensamentos protéicos (globóides ou pró-globóides) na massa fundamental; (4) formação de lipídios ao longo de toda a margem do vacúolo protéico; (5) início da cavitação intema do vacúolo, evidenciando sinais de reutilização das reservas; (6) fragmentação do conteúdo protéico; (7) adensamento do material fragmentado com visualização óptica ou eletro-transparente do vacúolo, com início da degradação dos lipídios; (8) vacúolo com pouco material adensado no seu interior e proteína identificada no citoplasma como fragmentos ou adensamentos, sem qualquer sinal de lipídios contornando a estrutura; (9) vacúolo sem qualquer material protéico individualizado no seu interior, sendo constituído apenas por uma matriz solúvel. Em particular, o suspensor assume três distintos padrões de comportamento: (1) a lise através de Morte Celular Programada da célula basal e das células contíguas a esta, durante a embriogênese inicial, (2) alise caracterizada por rompimento das paredes das células de comunicação entre a hipófise e embrião, na embriogênese final, e (3) a permanência do suspensor íntegro, em embriões em estágio de torpedo. A manutenção do suspensor para a continuidade do desenvolvimento embrionário, tanto quanto a dinâmica das reservas lipoprotéicas e seu processo de translocação, são de fundamental importãncia para a conclusão do desenvolvimento embrionário.

Previous papers on the flex genus, Aquifoliaceae family, Celastrales arder have shown that the embryos stay rudimentary, in the heart shaped stage, even when the fruits are ripe. The seeds need a period of 6 to 8 months to germinate, with a very low germination rate. Due to these facts, the production of seeds of maté (flex paraguariensis St. Hil.) is slow and scarce. The morphoanatomical analysis of the embryo and endosperm, during late embryogeny, has shown the presence of the suspensor even in advanced stages. This analysis has also shown changes in the storage and translocation of the endosperm reserves. Based on these results, the present work aims to detail the embryo and endosperm ontogeny, and the entire suspensor development, from its origin to degradation. The different stages of embryo and endosperm development were analyzed and described using the traditional anatomical methods for optical, as well as scanning and transmission electron microscopy. Along the ontogenetic development, the work describes the reproduction cycle of this species, its pistilate flowers, fruits, fertilization process, and embryo and endosperm ontogeny. The endosperm was classified as cellular, according to the EODP system. The type of cellular division during early embryogeny leads to the classification of the embryo development as being of Caryophyllad type. During early embryogeny, differerít stages were analyzed, beginning willi zygote formation, then bicellular stage all the way to the post-octant proembryos. During late embryogeny, the analysis was performed of stages ranging from the pyriform to the ripe torpedo embryos, when germination occurs. The increase of nutrients to the sporophytic embryo during early stages of development occurs mainly through the suspensor. This is because the xenophytic embryo is not completely developed. When the xenophytic embryo contains structured reserves, their mobilization is observed through the characteristic morphological changes. These changes follow a gradient in the micropyle-chalaza direction, as well as in the integument-embryo direction. Different steps of syntheses or translocation of reserves can be classified as: (1) formation of single membrane vesicles binding into vacuoles, (2) deposition of amorphous protein into them, (3) appearance of proteinous spots (globoids or progloboids) in the ground mass, (4) formation of lipids around the proteinous vacuole, (5) beginning of the intemal cavitation of the vacuole, showing signs of reserve degradation and translocation, (6) fragmentation of the proteinous bodies, (7) adhesion of fragmented material with optically or electrotransparent visualization of the vacuole, and the beginning of lipid degradation, (8) vacuoles with little dense material and protein identified in the cytoplasm as fragmentary or adhesion masses, without any sign of lipids around them, (9) vacuoles without any sign of proteinous material, being solely constituted by a soluble matrix. The suspensor showed three distinct developmental behavior pattems: (1) the lysis by programmed cell death of the basal cell and its contiguous cells during early embryogeny, (2) the lysis by the breaking of the cell wall between hypophysis and embryo during late embryogeny, and (3) the maintenance of the intact suspensor in embryos until the torpedo stage. The suspensor is not only a characteristic structure during early embryogeny stages, but its mainten ance as well as the dynamics of lipid and protein storage, and the translocation process are essential for the conclusion of embryo development.
