Rugose wood of the grapevine in Yamanashi Prefecture

In 1992, the rugose wood symptom (pitting and / or grooving) of the woody cylinder was first recognized in Yamanashi. These studies intended to establish possible correlations of rugose wood with viruses and the way of controlling rugose wood. Indexing tests were carried out by grafting indicators 'St.G', 'kober 5BB', and 'LN33' to assess, respectively, Rupestris stem pitting (RSP), corky bark (CB), Kober stem grooving (KSG), and LN33 stem grooving (LNSG). Seven of nine infected sources (cv. 'Kyoho,' 'Neo muscat,' 'Rubi okuyama') were indexed as positive for RSP. These vines induced the wood pitting symptom in the indicator vitis rupestris 'St.G'. Two of nine infected sources, cv. 'Kaiji,' which induced stem pitting in 'St.G', also reacted in 'LN33'. Stem grooving developed on the indicator 'LN33', but this symptom was not accompanied with the leaf redding and swelling internode associated with CB in this indicator. Therefore, two infected cv. 'Kaiji' were indexed as positive for RSP and LNSG. Extracts from cortical scrapping that were affected by rugose wood were analyzed for the presence of Grapevine virus A (GVA), Grapevine virus B (GVB), Rupestris stem pitting associated virus (RSPaV), and Grapevine leafroll associated virus 1 (GLRaV-1), -2 (GLRaV2), and -3 (GLRaV3) using RT-PCR. All infected vines that indexed positive for RSP contained RSPaV. Two of nine contained RSPaV and GVA; one of nine contained RSPaV, GVB, and GVA. Grapevines for rootstock and scion without viruses were produced by shoot-tip culturing. Four rootstocks and seven scions were grafted with each other and planted in the field. After nine years, these grapevines were free from rugose wood symptoms and not infected by GVA, GVB, or RSPaV.
