Quick diagnosis of bovine leukosis using stump cytology

Because bovine leukosis (BL) has been found increasingly in slaughtered cattle in Japan in recent years, a quick diagnostic procedure for diagnosing BL is needed. We tested an immunostaining protocol using stump cytology that is easily performed and whose results could be obtained the same day. This immunostaining protocol provided the best staining quality when using acetone fixation with 3% H2O2 solution for blocking. The results of this immunostaining procedure in all five cases tested were identical to those when using a paraffin-embedded specimen. The immunostaining protocol by stump cytology tested in this study seemed to be useful for obtaining an immediate diagnosis of BL.
