In der pflanzenphysiologischen Grundlagenforschung werden Chlorophyll-Fluoreszenzmessungen seit langem eingesetzt, um den Zustand des Photosyntheseapparates, insbesondere des Photosystems II, zu charakterisieren. Aus praktischen Gründen wurde daraus das Ziel abgeleitet, mit Hilfe von Chlorophyll-Fluoreszenzmessungen die Vitalität von Pflanzen mit einer zerstörungsfreien Methode zu beurteilen. Im Rahmen eines Forschungsprojekts zum Spitz-Ahorn soll die Vitalität von Baumschulpflanzen verschiedener Herkünfte mit Chlorophyll-Fluoreszenzmessungen erfasst werden, wobei das Gerät PAM-2500 der Fa. Walz (Effeltrich, Deutschland) zum Einsatz kommt. Dieses Gerät kann mit Hilfe der Puls-Amplituden-Modulation (PAM) die Messsignale von sich überlagerndem Licht trennen. In einem Vorversuch wurden getopfte Spitzahorn-Pflanzen einem Trockenstress ausgesetzt und mit Kontrollpflanzen verglichen, wobei sog. Rapid Light Curves (RLC) aufgenommen wurden. Die daraus abgeleiteten Parameter wie die maximale Elektronentransportrate bei Lichtsättigung (ETRmax) und der Lichtsättigungskoeffizient verringerten sich in der Versuchsgruppe der trockengestressten Pflanzen deutlich im Vergleich zu den Kontrollpflanzen, jedoch nicht vor dem Erscheinen phänotypischer Symptome. Diese aus RLCs gewonnenen Parameter reagieren schon einige Tage früher als konventionell gemessene Fv/Fm-Werte bzw. die effektive Quantenausbeute Y(II). Wegen der hohen Streuungen, auch innerhalb und zwischen den Pflanzen der Kontrollvariante, erscheint ein erfolgreicher Einsatz der Chlorophyll-Fluoreszenzmessungen unter Freilandbedingungen in der Baumschule eher fraglich.

anglais. In basic plant physiological research, chlorophyll fluorescence measurements have long been used to characterize the state of the photosynthetic apparatus, especially photosystem II. For practical reasons, this led to the goal of using chlorophyll fluorescence measurements to assess the vitality of plants using a non-destructive method. As part of a research project on Norway maple, the vitality of nursery plants of different provenances will be assessed with chlorophyll fluorescence measurements, using the PAM-2500 instrument from Walz (Effeltrich, Germany). This instrument can use the Pulse Amplitude Modulation (PAM) method to separate the measurement signals from superimposed light. In a preliminary experiment, potted Norway maple plants were exposed to drought stress and compared with control plants, recording so-called Rapid Light Curves (RLC). The derived parameters such as maximum electron transport rate at light saturation (ETRmax) and light saturation coefficient decreased clearly in the experimental group of drought-stressed plants compared to control plants, but not before the appearance of phenotypic symptoms. These parameters obtained from RLCs respond several days earlier than conventionally measured Fv/Fm values or the effective quantum yield Y(II). In addition, high scattering of the measured values was shown, even within the plants of the two control variants, a successful application of the method under field conditions in the nursery seems rather questionable.
