Patlıcanda Kotiledon Eksplantından Organogenezis Sağlanması Üzerinde Çalışmalar

Vegetatif propagation of plant materials obtained in vitro conditions or in a limited number can be madeimportant. In this study, we aimed to determinate the composition of aculture medium suitable for in vitro plantregeneration of eggplants also to investigate the effect of genotypes to regeneration capacity. As plant materialtwo open pollinated eggplant varieties (Long Purple and Topan), one local hybrid genotype (3x8) and a wildspecies (Solanum integrifolium) was used. Cotyledon pieces of seedlings were used as explants and thatseedlings were obtained from eggplant seeds which are germinated in vitro conditions. The composition of thebasic MS medium was used and this medium was enriched with two different carbohydrate sources (glucose,1.5% or 2% sucrose). "2 mg/l zeatin and 0.01 mg/l NAA," "4 mg/l NAA," "10 mg/l NAA", are added as growthregulators. Experiment was established according to randomized block experimental design. 50 cotyledon pieceexplants were cultured for each application. The cultures were placed in a climate room in the darkness for 3weeks and then photoperiodic system was applied. After one month observations made in cultures, callusformation rate, presence of organogenesis (shoot and root formation) was determined and the amounts perexplant was detected. Different response among genotypes was observed on to effect of culture mediumcomposition. For the formation of shoots, it could be necessary to transfer the callus tissues into the new mediacompositions including cytokinins. It is required the further investigations about the optimization of this method.
