Purification and physico-chemical characterization of the vegetations of Riemerella anatipestifer

In order to purify the vegetations of Riemerella anatipestifer (RA) four kinds of media for RA cultivation and four purification methods for vegetation extraction were compared. The results showed that Martin agar was the most suitable for growth of the bacteria leading to relatively more prolific vegetations and less impurities. Differential centrifugation following membrane filtration could effectively separate the vegetations from RA and successsfully purify them. No nucleic acids were detected in extracts from the vegetations and the SDS-PAGE pattern of vegetation was similar to that of the corresponding RA. In morphological study the vegetations could be classified into three kinds: big, medium and small ones. The big and medium vegetations contained a thick wall around its with a triangular or quadrilateral nucleus in the center. The small vegetations were round and surrounded by a very thin outer-membrane with a spot- or star-like nucleus. These results provide some scientific basis for taxonomical study of RA.
