Effect of whey hydrolysate in fetal bovine serum replacement medium on protein synthesis mechanism of Hanwoo satellite cells

This study investigated the effects of whey-derived leucine in an FBS replacement medium on Hanwoo satellite cell protein synthesis. Control groups were F20 (20% FBS) and F1 (1% FBS), while treatments included F1W7, F1W14, and F1W21 (1% FBS + 7, 14, and 21 mg WPH, respectively). F1W14 showed the highest proliferation on days 4 and 6 among other treatment. mTOR and FGF2 mRNA levels were significantly higher in WPH groups than in F20, suggesting enhanced protein synthesis. MYOD1 and PAX7 were also upregulated, indicating increased muscle cell proliferation. Phospho-mTOR, p-4EBP1, and p-p70S6K1 levels were elevated in treatment groups compared to F1, likely due to leucine in WPH. These results suggest that 14 mg WPH in 1% FBS effectively enhances protein synthesis. However, optimizing cell adhesion remains a challenge. Future research should identify supplements to improve attachment while maintaining proliferation and explore leucine’s role in cultured meat production. This approach may contribute to the development of cost effective and standardized culture medium for scalable cultured meat production.
