Immuno-stimulating and anti-metastatic effects of polysaccharides isolated from Glycyrrhiza glabra stalks

This study was conducted to evaluate the immunostimulatory and anti-metastatic activities of polysaccharides isolated from Glycyrrhiza glabra (GGP). GGP mainly consists of galacturonic acid (41.7%) and glucose (30.9%), characteristic of typical pectic polysaccharides. GGP-stimulated peritoneal macrophages secrete various cytokines. In an ex vivo assay of natural killer (NK) cell activity, intravenous administration of GGP enhanced the cytolytic effects of NK cells against YAC-1 lymphoma (up to 5.3 folds compared to the negative control (NC) administration). In experimental lung metastasis induced by Colon26-M3.1 carcinoma, prophylactic intravenous administration of GGP inhibited lung cancer up to 63% compared to NC, and dose-dependently increased TNF-α production. These results indicate that the anti-cancer metastatic effect is mediated by the activation of macrophages and NK cells. Thus, GGP exhibits a great potential as a functional food ingredient to improve immune function and prevent metastasis.
