Obtaining doubled haploids of Cucurbita pepo L. Texte intégral

Doubled haploids have been widely used worldwide in breeding programs and fundamental research as valuable homozygous material for about 100 years. The species Cucurbita pepo L. are represented by a huge variety of forms, include highly productive vegetable crops and have a wide distribution in the world. Despite the great economic importance, the creation of effective protocols to ensure stable production of doubled haploids in this species remains an urgent task. DH plants are of interest not only because of the acceleration of the breeding process, but also because of the realization of the huge potential of gametoclonal variability inherent in this highly polymorphic species. In this review, we analyzed the main technologies used for obtaining doubled haploids in vegetable crops of C. pepo: parthenogenesis in situ stimulated by treated/irradiated pollen, gynogenesis in vitro (unpollinated ovule culture in vitro) and androgenesis in vitro (anther/microspore culture in vitro). An analysis is presented of the research carried out from the beginning of the discovery of haploid plants to the current advances and evaluation of the prospects in the field of DH plant production. The main critical factors influencing the efficiency of each technology and its individual steps are considered. The developed technology of doubled haploids obtaining using non-pollinated ovary culture in vitro is presented. This technology allows to obtain up to 55 embryoids per one cultivated ovary (28 embryoids/ 100 cultivated ovules) To introduce haploid technologies into the breeding process it is necessary to evaluate the obtained plants for ploidy level. The use of direct counting of chromosomes in apical cells may present a certain difficulty in this species due to their large number (2n=40) and their small size. Depending on the level of laboratory equipment, ploidy determination using flow cytometry of cell nuclei and counting the number of chloroplasts in stomatal guard cells in the epidermis of the abaxial side of the leaf may be more convenient methods. The prospects for the use of molecular markers for assessment for homozygosity in DH technologies used, including C. pepo, are discussed in the review.

Relevance. The region continues to build new industrial greenhouses every year, as well as mass construction of winter film greenhouses in the farms of the population of Flat and foothill Dagestan. Today, there are more than 250 hectares of them.Methods. Experimental work was carried out in the greenhouse of LLC Agro-AS in 2019-2020. Carried out selection and evaluation of promising hybrids of tomato for the summer-autumn turnover in greenhouses. Studied the duration of interphase periods, estimated the percentage of fruit set in the inflorescence, determined the yield of tomato hybrids, and calculated the most economically promising ones for the natural and climatic conditions of Dagestan.Results. For the first time in the conditions of flat Dagestan, the following experimental data are proved the main elements of industrial technology for obtaining a high yield of tomatoes in protected ground structures using low-volume hydroponic technology on a coconut substrate were experimentally justified; highly productive tomato hybrids were identified for various protected ground structures; F1 hybrids were distinguished by good yield from the second decade of September to the second decade of December; hybrids such as Men-Shuai F1 and Mamston F1 provided 24.3 kg/m2 and 23.1 kg/m2 , respectively, according to the highest percentage of fruit setting, as well as the total and commercial yield. The term of planting seedlings in the substrate in the summer and autumn turnover in the conditions of the suburbs of Makhachkala on July 10 is economically effective, which ensures a high yield and net income per unit area. At the selling price of tomato fruits of 100-110 rubles per 1 kg, the profitability of production, depending on the timing of harvesting and sales, was 100-122%.

Relevance. Since the increasing demand for agricultural products, especially at present in the conditions of import substitution, cannot be met without creating new highly productive varieties and hybrids.Material and methods. This article provides a description and comparative characteristics of economically valuable qualities and varieties of vegetable crops, the most common and in demand among the population, created by breeders of the All-Russian Research Institute of Irrigated Vegetable Growing and Melon Growing – a branch of the FSBSI "PAFSC RAS". The experiments were laid on the experimental fields.Results. As a result of many years of breeding work presented varieties belong to different variety types, they differ in groups of precocity, shape, size, color of fruits. Tomatoes – Astrakhan, Bulldog, Avdeevsky, Caspian, Malinovi shar, Moryana, Rychansky, Forward, Orange Avuri, Malinovi Supergol, etc. Varieties of sweet pepper – Atomor, Marble, Novichok VNIIOB, Sprinter, Lyudmila, etc. Varieties of eggplant – Albatross, Diamond, Astrakom, Nizhnevolzhsky, Swan, etc. Since 2021, the Gnomik tomato variety has been entered into the State Register, and the Zarnitsa sweet pepper variety is being tested. The Astrakhan region is also a center for the creation and production of melon crops, their fruits belong to dietary products, are used both fresh and for preservation, preparation of drinks, desserts, home cooking. Watermelon varieties – Astrakhan, Photon, Ilyasovsky, Portioned; melon – Lada, pumpkin – Kroshka and Kapelka, zucchini – Sosnovsky and Jubilee, squash – Tabolinsky.

Relevance. The study of the timing and duration of individual phases of the development of strawberry cultivars of different ecological and geographical origin in these climatic conditions is of great practical importance. The purpose of the study was the determination of the timing of the main phenophases of introduced strawberry cultivars in the Orel region, the compliance of the phenorhythms of the studied cultivars with climatic conditions, as well as the ranking of the cultivars by the terms of flowering and maturation.Methods and materials. The article presents the results of phenological observations for the period 2016-2019. 34 strawberry cultivars of domestic and foreign selection were studied. The research was carried out at the VNIISPK site of primary variety study.Results. Based on long - term observations, on the time of entry into the flowering and fruiting phase, the cultivars were ranked into early, medium and late-maturing. The influence of effective temperatures on the onset of phenophases was analyzed. The sum of effective temperatures necessary for the beginning of flowering of strawberry cultivars of different maturation periods in the conditions of the Orel region was determined. For the onset of strawberry flowering, it takes from126.68 to 260.37°effective temperatures. Early cultivars need the sum of effective temperatures of 126.68-197.55°, cultivars of middle maturation need 146.31-225.44°, late cultivars - 159.93-260.37 depending on the year of study. Fruiting occurs at the sum of effective temperatures in early cultivars from 351.73°C to 465.43°C, in middle-maturing cultivars from 390.96 to 535.44°C, in late-maturing cultivars from 450°C to 649°C.

Relevance. Modern agricultural production requires the creation of varieties that combine high productivity, quality, and adaptability. Taking into account the development trends of modern agriculture towards multiplicity, it is necessary to improve and expand the assortment, helping to meet the needs of both large and small producers of agricultural products. Due to the lack of material and human resources in the agricultural sector, there is a clear need for varieties and hybrids adapted to industrial cultivation and storage technologies. Existing zoned varieties of root chicory have a root crop length of 40 cm or more, and the soils of the area of cycoroseeding are mainly heavy in mechanical composition, where the use of digging devices to such a depth is impossible. Production requires varieties with a short root crop, which has the main mass concentrated in the upper part.Methods. The purpose of this work is to identify valuable genotypes in the soil and climate conditions of the non-Chernozem zone of the Russian Federation among the variety of varieties of chicory root of different ecological and geographical origin for use in the creation of the source material of selection by inter-port crosses.Results. When creating the source material for the selection of root chicory by the method of intervarietal crosses, one of the parents should choose local intrazonal varieties adapted to the soil and climatic conditions of the zone (Yaroslavsky, Gavrilov-Yamsky), and the second – off-zonal varieties with economically valuable qualities (short root crop, high yields and chemical-technological indicators, resistance to root rot) and donors of these traits (Kharpachi, Sleszka, BilogorkaOS-2, BilogorkaOS-3, Rexor, Wixor, Luxor).

Relevance. According to the Federal Law of 16.07.1998 No. 101-FZ (as amended on 05.04.2016) "On State Regulation of Ensuring the Fertility of Agricultural Land", owners, owners, users, including tenants of land plots are obliged to: carry out the production of agricultural products in ways that ensure the reproduction of the fertility of agricultural land, as well as excluding or limiting the adverse impact of such activities on the environment. Environment.Materials and methods.The research was carried out on a stationary field experience in the Podolsk district of the Moscow region in 2011-2012. On experience, five-floor crop rotation will be deployed in time: 1) oats; 2) barley with planted perennial herbs; 3) perennial herbs of the first year of use; 4) perennial herbs of the second year of use; 5) winter wheat. The plots were located in the upper part of the slope with a slope of 14% (weakly pitched) and in the lower part of the slope with a slope of 7% (gentle).Results. It was found that on average annually in the slope areas of the non-chernozem region soils lose from the layer 0-40 cm with a surface runoff of phosphorus 0.7-6.0 kg / ha, potassium 2.3-28.9 kg / ha, calcium 7.3-45.8 kg / ha. With anti-erosion methods of tillage on a slope with a steepness of 7%, the migration of chemical elements was 2.2 times less compared to similar options on the slope with a steepness of 14%. Surface runoff redistributes substances along the length of the slope, therefore, it is necessary to take this into account during chemical reclamation of the slopes soils and carry out differentiated application of fertilizers and lime. After chemical analysis of the arable layer of soil, it turned out that the maximum content of humus, mobile forms of phosphorus and exchange potassium is noted in the lower part of the slope, which is associated with the processes of washing and alluvium of the soil.

Relevance and methods. Leek is one of the most promising onion crops for industrial cultivation in the Non-Chernozem zone, which has medicinal properties and high nutritional value. The results of studying the technological features of leek cultivation in the open ground of the Moscow region are presented. The studies were carried out in 2019-2020 on the territory of the “Vegetable Experimental Station named after V.I. Edelstein". We studied the influence of the feeding area of leek seedlings on the biometric parameters of plants and the quality of products. The object of the study is the variety Karantansky, experimental options: 1) seedling feeding area – 6 cm2 (control), 2) seedling feeding area – 8 cm2 , 3) seedling feeding area – 10 cm2 . The objects of research to study the content of ascorbic acid were leek varieties: late-ripening – Karantansky (standard), Bandit, Dobry Molodets, mid-ripening – Columbus, Hobot slona.Results. It is noted that the feeding area during the seedling period does not significantly affect the biometric parameters of leek plants: leaf area, plant weight, leek pseudostems diameter. Therefore, during the period of growing seedlings, from the point of view of rational use of the area in the seedling department, it is recommended to use the minimum feeding area – 6 cm2 . It was also found that the studied varieties accumulate ascorbic acid in different ways in various plant organs. The maximum content of ascorbic acid was noted in the leek pseudostems of the control late-ripening variety Karantanskiy – 23.6 mg%. The minimum values are for Dobry Molodets (11.8 mg%) and Hobot Slona (11.9 mg%).

Relevance. It is possible to significantly increase the multiplication factor of varieties of winter garlic by growing from air bulbs (bulbs). For successful culture using air bulbs, it is very important to correctly determine the most productive fraction for each variety and calibrate it for sowing.Material and methodology. In our collection, there are more than 70 samples of winter garlic collected from different regions of Russia and two CIS countries. The air bulbs were calibrated using a set of round sieves with apertures of 3, 5, 7, 10 mm.Results. On average, the samples of the collection have a high percentage of ripening of air bulbs - 75- 83%. The number of underdeveloped flowers on average for samples of different origins varies slightly – 37-48 pcs. in the inflorescence. The mass of inflorescence on average in specimens of local origin (Sverdlovsk region) is 4.2 g, of different origin (Ukraine, Uzbekistan, 5 regions of Russia) – 4.9 g, of southern origin (Dagestan) – 5.6 g. In specimens of local origin air bulbs of medium (5-7 mm) and small fraction (3-5 mm), different origin – large (7-10 mm) and medium fraction, southern origin – large, medium and small fraction. For reproduction by air bulbs, samples 9/5-1. 9/5-2 of local origin were allocated (Sverdlovsk region); 9/4 (Ukraine), 8/15, 10/5, 10/10 (Moscow), 9/3-1, 9/3-2 and grade Shadeyka (Perm); sample-5, 10/11, 10/13 (Dagestan).

Relevance. Currently, the use of secondary plant-orgin metabolites with high biological activity is becoming an important part of a comprehensive plant protection program. The mechanism of their action is aimed at activating molecular biological and physiological systems that increase the ability of the plant organism to withstand the adverse effects of biotic and abiotic nature. The purpose of these studies is to screen the selectivity of the physiological activity of phytoregulators in relation to different varieties of vegetable beans.Methodology. Plants of two mid-ripening varieties of vegetable beans were twice sprayed with solutions of plant biological products of different chemical composition, isolated from 12 plant species of different families. Control – treatment with water, standards - solutions of Pharmayod and Fitolavin. During the growing season, an analysis of the development of diseases of various etiologies on seed crops was carried out. Seed productivity of plants was taken into account after harvesting and threshing the beans, the sowing qualities of the obtained seeds were studied in accordance with the relevant recommendations and GOST 12042-80, and the biological effectiveness (BE) of the action of phytopreparations was calculated.Results. Two-fold treatment of beans with most of the tested preparations reduced the total percentage of affected plants by more than 25%. The protective effect of the studied glycosides differed significantly depending on their chemical structure, disease etiology and cultivar responsiveness. Among them, there are preparations based on steroidal glycosides - Moldstim and Mestim, which have the most pronounced immunomodulatory effect on the resistance of vegetable beans to phytopathogens. Treatment with Mestim in the selected concentration led to a decrease in seed productivity of plants. Other phytopreparations showed a neutral or stimulating effect on this trait, increasing the proportion of germinating seeds in the yield structure by 3-26% relative to control. The certain varietal specificity has been identified by the total effectiveness of the action. Irioid glycosides from Linaria genistifolia and the sum of iridoids and flavonoids from Linaria vulgaris showed the greatest activity on the responsive cultivar Lika. On the SiBemol variety, there is Moldstim, a steroidal glycoside of the furostanol series, isolated from Capsicum annuum. Their biological effectiveness was higher than that of Fitolavin and Pharmayod standards.

Relevance. The purpose of the research was to produce a new medium-ripe, high-yielding variety of vegetable peas with the usual type of leaf to expand the variety in the now used conveyor variety selection of the Krymsk EBS VIR Branch.Materials and methods. The experience was laid on the fields of the Krymsk EBS VIR Branch (Russia, Krasnodar Region, Krymsk) in 2019-2020. Varieties and strain were evaluated in a competitive variety testing. Sowing was carried out with a selection seeder SKS-6-10. The plot area was 10 m2 , the experiment was repeated four times. In the phase of technical ripeness, the sheaves were selected for description and the yield was taken into account. The conservation of green peas was done at the Tech Evaluation Lab.Results. Competitive variety testing in the mid-ripeness group was held by two strains 2014/7 and 344/16. The strain 2014/17 for green pea yields surpassed the standard "Parus" and G-344/16 in both years of study. Fresh and processed green peas were rated by the Tasting Commission at 5.0 and 4.8 points, respectively. High yield is achieved due to the paired beans on the peduncle and a large number of grains in the bean, up to 10 pieces. The grain in technical ripeness has a dark green color and the linear dimensions are 9.0x8.1 mm. In biological maturity, the seeds are intensely green, angular-compressed with a mass of 1000 pieces. 188-225 g.Conclusion. The results of field and laboratory analyses carried out on the complex of economicvaluable characteristics made it possible to distinguish the strain 2014/7, which in 2020 was transferred to the State Variety Testing under the name "Patriot". It is recommended as a supplement to the existing set of vegetable pea varieties of the Krymsk EBS VIR Branch in the mid-ripe group.
