IN VITRO PROPAGATION OF KORONAKI AND CORATINA OLIVE CVS.

Experiments carried out through 2016 revealed that a proper protocol for the in vitro propagation of Koronaki and Coratina olive cvs. as follows : Sterilization of stem node explants was carried out successfuly with copper sulphate at 50mg/l for Koronaki cv. And Ampicillin at 150mg/l for Coratina cv. both cultured on Olive medium.Multiplication on MS medium enriched with BAP at 1.5mg/l for both cvs. Rooting of Koronaki on half sterngth OM medium enriched with IBA at 0.5mg/l and Coratina on half sterngth MS medium enriched with NAA at 0.1mg/l. Acclimization of Koronaki plantlets derived from half strength OM rooting medium and acclimatized on peat : sand (1:1) and Coratina plantlets derived from MS rooting medium and acclimatized on peat :sand : vermiculite (1:1:1).

Stem node explants of three almond (Prunus dulcis Mill) cultivars (Om – Elfahm, M-Dalt and Ne Plus Ultra) and the hybrid rootstock “Hansen” cv. were suc-cessfully established and proliferated on Murashige and Skoog medium (MS) sup-plemented with benzyl adenine (BA) at 0.0 - 2.0 mgl -1 and indol butyric acid (IBA) at 0.0 - 0.1 mgl-1 .The highest bud development percentage of stem node explant was obtained with medium contained BA at 2 mgl-1 with or without IBA at 0.1 mgl-1 for M – Dalet and Ne Plus Ultra almond cultivars compared with other treatments. The longest shoot during establishment stage was recorded by “Hansen” rootstock cultured in MS with BA at 2.0 mgl-1 plus IBA at 0.01 mgl-1.Using BA at 1.0 mgl-1 plus IBA at 0.1 mgl-1 and BA at 2.0 mgl -1 plus 0.05 or 0.1 mgl -1gave the highest number of proliferated shoots for Om – Elfahm, M-Dalt and Ne Puls Ultra cvs. and “Hansen”, respectively in the 3rd subculture. The best average proliferated shoot length was achieved with medium supplemented with BA at 1.0 mgl-1 plus IBA at 0.05 mgl-1 or 0.1 mgl-1 for M-Dalt, Ne Puls Ultra and “Hansen” respectively, where-as, using BA at 2.0 mgl -1 plus IBA at 0.1 mgl -1 recorded the longest average prolif-erated shoot for Om- Elfahm cultivar. Microshoots were rooted in modified Bourgin and Nitsch medium supplemented with IBA or NAA at 1.0 mgl-1 only or combination (0.2 IBA mgl-1 + NAA 0.4 mgl-1, IBA 0.4 mgl-1 + NAA 0.8 mgl-1 and IBA 0.5 mgl-1 + NAA 1.0 mgl-1). The large callus (more than 10 mm diameter) was recorded with “Hansen” rootstock and Om – Elfahm treated by IBA at 0.4 mgl -1 plus NAA at 0.8 mgl -1. The greatest rooting percentage (66.67%) was obtained with medium contained IBA at 0.2 or 0.4 mgl-1 plus NAA at 0.4 or 0.8 mgl-1 for “Hansen” rootstock and Ne Plus Ultra shoots.

Nodal stem segments of in vitro growing somatic embyogenesis and seedling-derived plantlets of sweet orange, C. sinensis (L.) of (Washington navel, shamouti and blood orange cvs.); grapefruit, C. paradisi (L.); Local mandarin, C. deliciosa (Tenor); lemon, C. limon; (L.); citron, C. medica (L.); lime, C. aurantifolia (Christm); rough lemon, C. jambhiri (Lush) and sour orange, C. aurantium (L.) were cultured on MS medium supplemented with different concentrations of BA (0.5,1.0 and 2.0 mg/L) alone or in combination with NAA (0.5 mg/L) for shoot multiplica-tion. The results indicated that somatic embryogenesis-derived plantlets can be suc-cessfully propagated in vitro with a good vigor compared with germinated seedlings. A large number of proliferated shoots were produced when these explants were cul-tured on MS medium containing 0.5mg/L of BA combined with 0.5mg/L of NAA. Most obtained shoots were rooted on half-strength MS medium containing 0.5 mg/L of NAA. These in vitro grown plantlets were then successfully transferred to green house through acclimatization process.

Study was conducted during 2008 - 2009 on five local grape cultivars: Gubery, Assemy, Aswed, Razeql and Bayadh at the faculty of AgricultureThamar University . Cultivars were collected from Rawdha and Bany Husheish area - Sana'a Governorate. Morphological and physical characters of berry were determined and the chemical analysis for juice was also assessed. Finally, cuttings were taken from all, cultivars and cultivated at the nurser.Data were collected and analyzed by SAS system according to the least significant difference at the probability level of 0.05 and the results showed that: Assemy cultivar gave the maximum value of berry weight of 8.5 g, whereas the lowest rate was for Razeqi cv. of 1.2 g. According to berry dimensions, berry length ranged between 1.6 - 2.6 cm and berry width 1.35-2.43 cm. Razeql cv. gave the highest content of T.S.S (24.5%), whereas Gubery cv. contained the lowest value of 20.37%. pH value was between 3.98-4.77 and organic acids were between 0.35 -0.80%. Aswed cv. gave the highest rate of rooting percentage of 58% meanwhile, the lowest rate was found In the Razeqi cv. (6.75).
