Breeding profiles at the periparturient stage in captive red foxes (Vulpes vulpes) mating naturally or subjected to artificial insemination in Japan

Introduction: Breeding profiles at the periparturient stage in red foxes which mated naturally or were subjected to artificial insemination were retrospectively surveyed using 130 vixens during their reproductive seasons of 2012–2017 in Japan. Material and Methods: Natural mating vixens were encouraged a maximum of three times with the same male, while artificial insemination was conducted using frozen-thawed semen with the bovine semen extender as a diluent. Results: With natural mating, conception rates after one, two, and three copulations were 55.8%, 68.0%, and 85.7%, respectively, showing a significant difference between the rates for one and three copulations. Conception rates with artificial insemination were 82.4%. Mean gestation periods were between 52.1 and 53.3 days in all groups. Mean litter sizes were 3.7–4.3 cubs with natural mating, and 4.4 cubs with artificial insemination. Although some sporadic and inconsistent changes in litter sizes were noted between primiparous and multiparous groups, these were of doubtful clinical importance. Conclusion: This is the first report from Japan concerning basic breeding events of red fox vixens in captivity.

With the exception of the domestic cat, all felid species (Felidae) are currently threatened with extinction in their natural habitat. To develop effective and optimal wild cat conservation programmes with assisted reproductive technology (ART) it is necessary to combine advances from different disciplines of science, starting from the biology of the species, through research into the population and habitat, assisted reproductive technologies, establishment of gene banks, developing bioinformatic systems, and ending with biodiversity and endangered species management. In the last few years knowledge of felid reproduction has expanded considerably thanks to comparative studies utilising the domestic cat as a research model for endangered wild cats. Basic reproductive techniques utilised in both domestic cat breeding and rescuing wild felid populations that are threatened with extinction include semen collection and cryopreservation, artificial insemination, oocyte collection, in vitro maturation, in vitro fertilisation, somatic cloning, and embryo transfer. The main directions in which assisted reproductive technologies are being developed in wild cat conservation implementations and the contribution of Polish research centres in advancing these methods are presented.

Introduction: The aim of this study was to investigate the relationship between body condition score (BCS) determined on the dry-off day, calving day, and in the first month of lactation, its changes during the dry period and early lactation, and periparturient indices and fertility in high-producing dairy cows. Material and Methods: The experiment was conducted in two herds: A and B, located in Western Poland. The studies were conducted on 116 and 108 Polish Holstein-Friesian dairy cows respectively, with an average milk yield of >10 000 kg/305-day lactation. The experiment included the dry period (-56 d to the calving day), the calving day, and early lactation (from +1 to +56 d). The experimental factor was BCS (0 to 5-point scale). The BCS was performed by one person on day -56, on parturition day (in the first 12 h after calving) and on day 30 of lactation. Results: A decrease in BCS (≥-0.25) in herd A during the dry period accelerated the planned calving period by 7.3 d. In the group of cows with BCS <3.25 on the dry-off day, the lowest artificial insemination index (1.80), the shortest period of insemination services (25 d), and days open (87 d) were recorded. Moreover, cows with BCS < 3.25 at calving had the shortest days open (91 d). BCS >3.50 in the first month of lactation (30 d) resulted in the extension of uterine involution period (56 d). Improvement of BCS during the dry period shortened the anoestrus (60 d) in herd A and the period of insemination service (60 d) in herd B. However, in this group (IM BCS ≥ 0.25) of cows the day of the highest artificial insemination index (2.50) in herd B was analysed. Conclusion: The body condition on the dry-off day and at calving, as well as its deterioration in the first month of lactation, have a considerable effect on fertility indices in dairy cows, thus confirming the advisability of its regular monitoring during routine operations connected with the management of a dairy cattle herd.

Introduction: The aim of this study was to evaluate the dependence between ejaculate traits, sperm morphology, and ejaculate volume in Duroc boars. Material and Methods: The analysis involved 121 ejaculates collected from 12 Duroc boars kept in three artificial insemination centres located in central Poland. Ejaculates were collected manually at one-month intervals, over a period of 10 months. At least 10 ejaculates were collected from each boar. The material was divided by ejaculate volume and each ejaculate was assigned to one of three volume groups: 160 mL and lower, 161–200 mL, and 201 mL and higher. The ejaculates were assessed to identify the basic physical traits and determine the incidence of morphological abnormalities in the spermatozoa, specifying major and minor abnormalities. Furthermore, the morphological structure indices for the spermatozoa were also calculated. Results: In large-volume ejaculates, spermatozoa were more elongated in shape, their heads were more elongated and had the largest flagella. With an increase in the ejaculate volume, sperm concentration in the ejaculate decreased. Moreover, while the total number of sperm in the ejaculate increased, the number of insemination doses obtained from a single ejaculate were higher. Conclusion: The volume of ejaculate has little impact on the occurrence of morphological abnormalities and the size of sperm cells. Ejaculate volume is important for the shape of the sperm cells.

The rabbit corpus luteum becomes an estradiol-dependent tissue by day 6 of gestation, and adequate estrogen is critical to avoid pregnancy failure. The aim of this study was to investigate the effect of parity (primiparous or multiparous), day of mating (11 or 21 d postpartum), and suckling status (suckling or nonsuckling) on various reproductive traits in hybrid rabbit does (n = 96). Ovarian structures on day 6 after coitus were evaluated by means of ultrasonography. Blood samples were collected that day, and the serum was analyzed for estradiol-17β by radioimmunoassay (RIA). Parity and suckling had significant effects on mating rate (P < 0.01 and P < 0.05, respectively). More does accepted the male on day 11 than on day 21 (P < 0.05). Ovulation frequency was significantly affected by parity (P < 0.05), day of mating (P < 0.01), and suckling (P < 0.01). Fewer ovarian large follicles and lower estradiol-17β levels were detected in suckling compared with nonsuckling rabbits (P < 0.01). Since estrogen concentrations are commonly used to assess follicular growth and steroidogenic capacity, the lower hormonal levels in the suckling rabbits may reveal poorer ovarian activity, which could result in reduced reproductive efficiency. Our observations confirm the existence of a partial antagonism between lactation and reproduction in rabbits. Further research is needed to elucidate these phenomena, including when artificial insemination is done. Ultrasonography could represent a noninvasive and reliable method for studying several reproductive functions and dysfunctions in rabbits.

OBJECTIVE To evaluate use of flunixin meglumine as a treatment to postpone ovulation in mares, mare fertility after flunixin meglumine treatment during estrous cycles, and effects of flunixin meglumine on function of the corpus luteum after ovulation. ANIMALS 13 healthy mares. PROCEDURES A single-blinded, placebo-controlled, crossover study was conducted. Flunixin meglumine (1.1 mg/kg, IV, q 24 h) or lactated Ringer solution (placebo treatment) was administered for 2 days to mares with a dominant follicle (≥ 35 mm in diameter) and behavioral signs of estrus. Mares then were bred by artificial insemination. Number of days to ovulation from initial detection of a follicle ≥ 30 mm in diameter, uterine edema score, and pregnancy were determined by ultrasonography; the examiner was unaware of the treatment of each mare. Serum progesterone concentrations were evaluated 5 and 12 days after ovulation by use of radioimmunoassay. RESULTS Data were available for 45 estrus cycles of the 13 mares. Number of days to ovulation from initial detection of a follicle ≥ 30 mm was not significantly affected by administration of flunixin meglumine versus the placebo. Per-cycle pregnancy rate was not significantly different between flunixin meglumine (20/24 [83%] breedings) and the placebo (13/19 [68%] breedings). Flunixin meglumine did not significantly affect behavioral signs of estrus, uterine edema, or serum progesterone concentrations. CONCLUSIONS AND CLINICAL RELEVANCE Findings did not support the use of flunixin meglumine to postpone ovulation in mares.

The objectives were to determine the effects of feeding supplements rich in omega-6 or omega-3 fatty acids (FA) during the late gestation to the early postpartum and breeding periods on reproduction and milk FA profile in beef cows. For each of two years, at the beginning of period 1 (mid-December), 72 beef cows, calving in January or February, were assigned to diets supplemented with roasted flaxseed (Flax) or roasted soybean (Soybean). For each of two years, after 11 wk (end of period 1), 18 cows of 36 in the Flax group were switched to the soybean supplement and 18 cows of 36 in the Soybean group were switched to the flax supplement (start of Period 2). Cows were bred by timed artificial insemination (TAI) in week 5 of period 2. The FA composition of the milk reflected the FA profile of the oilseed supplements. There were no differences in pregnancy rates among the 4 groups. The treatments had no effect on plasma prostaglandin metabolite levels or ratios at 4 to 11 d postpartum. At 5 to 6 d post- TAI, pregnant cows fed Flax in period 1 had lower (P < 0.05) plasma prostaglandin F metabolite (PGFM) levels and PGFM to prostaglandin E metabolite (PGEM) ratio than cows fed Soybean, but there were no significant differences at 19 to 20 d post-TAI. Cows pregnant from TAI and fed Flax in period 2 had higher (P < 0.05) serum progesterone levels at 5 to 6 d post-TAI than cows fed Soybean, but there was no difference at 19 to 20 d post-TAI. The dietary treatments had no effect on pregnancy rates, but there were some effects on plasma PGFM levels, PGFM to PGEM ratios, and serum progesterone levels. The FA supplements influenced the FA composition of milk.

A 3 year-old female Afghan hound came to the Veterinary Referral Hospital of the College of Veterinary Medicine, Seoul National University for artificial insemination (AI) with frozen semen. In order to inseminate, semen was frozen in USA 3 years ago. Frozen semen was sent by air from Santiago to Seoul for AI. The stud died 2 years ago, so we could only use a limited amount of frozen semen in that estrus cycle. The number of total motile spermatozoa was 59.4×10∨6 and the total volume was 1.2 ml. The frozen spermatozoa were thawed in 70℃ water for 8 sec, which were then deposited at the bilateral uterine horns by a surgical method.

A six-year-old Siberian husky bitch was presented to Small Animal Gynaecology and Obstetrics Unit, Madras Veterinary College Teaching Hospital, with a history of proestrus bleeding and prolonged breeding failure due to shyness. Based on progesterone value (8 ng/mL, artificial insemination was done. On day 45 post AI, ultrasonographic examination revealed the presence of a viable foetus and lateral abdominal radiography showed the presence of a single foetus. Based on the observations, the owner was made aware of chances of risk of dystocia due to single puppy syndrome and the case was kept under observation. However, the case was presented on day 65 of gestation with signs of inappetence and greenish black vaginal discharge for the past 6 hrs. Based on the Ultrasound examination, Radiography and progesterone concentration, the case was diagnosed as dystocia due to single puppy syndrome. Under local anaesthesia, episiotomy was performed and a viable foetus (650 gm) was delivered. The episiotomy wound was closed in simple interrupted suture pattern using polyamide (1-0). After one week of postoperative care, the bitch had an uneventful recovery.
