Reproductive consequences of deficiency of uridine monophosphate synthase in Holstein cattle

Cattle heterozygous for deficiency of uridine monophosphate synthase required more breeding services per calving when mated to other heterozygotes than did matings of normal cattle. Gestation length, number of breeding services per calving, and days from breeding to pregnancy examination were monitored on 759 complete gestations, 76 false-positive pregnancy diagnoses, 14 false-negative pregnancy diagnoses, and 413 negative pregnancy diagnoses in a dairy herd between 1983 and 1987. For complete gestations, 15 heterozygote X heterozygote matings required 3.13 +/- 0.37 breeding services per calving, which was significantly more than the 2.05 +/- 0.05 breeding services required for normal X normal matings; gestation length and days from breeding to pregnancy examination were similar between mating types. For false-positive pregnancy diagnoses, females diagnosed pregnant, but subsequently found not to be pregnant, 5 heterozygote X heterozygote matings averaged 51 +/- 23 days of gestation, which was less than the 93 +/- 6 days required for 71 normal matings; services and days from breeding to pregnancy examination were similar between mating types. All false-negative pregnancy diagnoses, females diagnosed not pregnant but later found to be pregnant, were made on cattle with normal matings. For negative pregnancy diagnoses, heterozygous matings averaged 0.3 more breeding services per examination than normal matings.

Red blood cell populations separated by density centrifugation were compared in a dynamic assay of osmotic stress. Red blood cells from Beagles genotypically normal and nonanemic (nonaffected), Beagles with inherited hemolytic anemia (anemic), and Beagles presumed to be carriers of the anemia trait (trait carriers) were examined for rate and extent of swelling after exposure to the ionophore A23187 in a medium containing calcium and potassium chloride. Comparisons were made between RBC populations separated on the basis of density. Significant differences were observed in the rates of cell swelling in Rbc populations separated by density between nonaffected and anemic Beagles. The response of RBC from Beagles presumed to carry the anemia trait was similar to that of RBC from nonaffected dogs. One phenotypic expression of this inherited abnormality of Rbc in Beagles was an accelerated rate of RBC swelling under osmotic stress, and this swelling response diminished with increasing RBC density.

During the investigation of nuclear DNA of boas of Belarusian meaty breed there was studied the polymorphism of EPOR, MUC4 and IGF-2 genes with the further determination of frequency of alleles and genotypes. Among boars tested for the polymorphism EPOR gene there was stated high percentage of heterozygous samples (EPOR**st): 43,3% for Belarusian meaty breed boars and 66,7% for Belarusian Kripnaya Belaya breed. In course of analysis of the genetic structure by MUC4 gene there was revealed the predominance of animals with the desirable d allele combination (MUC**cc): 60% for Belarusian Kripnaya Belaya breed and 82,2% for Belarusian meaty breed; by IGF-2 gene with homozygotic manifestation of recessive alleles: 46,4% and 63,4%, respectively. In the conditions of the Republic of Belarus there were determined the preferred boar genotypes with EPOR, MUC4 and ICF-2 genes and combinations which made it possible to increase the fattening and meaty qualities of posterity. The research results showed that in the selection breeding schemes it was necessary to consider not only maternal genotype, but also male genotypes giving preference to EPOR**st, MUC4**ss and ICF-2**qq genotypes
