Morphological and molecular characterization of cattle hydatid cysts in Khorramabad, Iran

BACKGROUND: Hydatidosis is one of the zoonotic diseases which affect animals and human beings at the larval stage of Echinococcusgranulosus, thereby playing a role in exacerbating the economic and health problems in Iran. OBJECTIVES: This study was conducted to investigate the strains of Echinococcusgranulosus isolated from cattle hydatid cysts, in Khorramabad, in the Lorestan Province, west of Iran. METHODS: Twenty six isolates of hydatid cyst of cattle from lung (24 samples) and liver (2 sample) organs were collected from Golshan Slaughterhouse, in Khorramabad. All of the samples were transferred to the laboratory for morphometric characterization and molecular study. In morphological characterization, blade length of large (LBL) and small (SBL) hook and the ratio of blade length to total length in large (LBL/LTL) and small (SBL/STL) hooks and total length of large (LTL) and small (STL) hooks were measured. In molecular study, a partial sequence of cytochrome oxidase 1 (COX1) with 440 bp in length was amplified applying primers J3 and J4.5. Genomic DNA sequencing was performed by Sanger’s method. RESULTS: The morphological results showed that there is no significant difference between isolated from cattle hydatid cyst and sensu stricto strain (P<0.05). The results of molecular studies support the findings of morphological characterization. All sequences showed 100% identity with sensu stricto strain. CONCLUSIONS: The results from this study showed that sensu stricto strain (G1) is a causative agent of cattle hydatid cyst in Khorramabad. On the other hand, the cattle play a role in enabling the parasite to complete its cycle. Hence in order to execute a control program for minimizing the effects of this disease, the cattle shall be considered as a source of infection for dogs. The results of this study could be helpful in designing such control program in the region.

Examination of 528 (450 males and 78 females) dromedary camels slaughtered at Cairo abattoir revealed that a total of (93) 17.6 % were infected with hydatidosis. Post mortem examination revealed that infection was restricted only in the lungs and the liver of infected camels. Among the 93 hydatidosis infected camels, lungs were the most frequently infected 88 (94.623%) compared with liver 5 (5.376%). ELISA test using partially crude antigen and purified antigen is important for the early diagnosis of cystic echinococcosis as most cases in the early stages of infection are asymptomatic. Sensitivity of ELISA using the crude antigen was 82.758% while sensitivity of the partially purified antigen was 79.310 %. On the other hand specificity of the crude antigen was estimated as 62.5 % and specificity of partially purified antigen as 75.0 %.

A cross-sectional study to determine the prevalence of bovine cysticercosis and hydatidosis was conducted from October 2007 to March 2008 in cattle slaughtered at the Jimma municipal abattoir. Cyst distribution and viability of bovine cysticercosis and hydatidosis were also determined. A total of 512 carcasses were inspected of which 15 (2.93%) and 161 (31.44%) were infected with Taenia saginata metacestodes and hydatid cysts, respectively. From a total of 109 cysticerci collected from infected carcasses, 47 (43.12 %) were viable. The anatomical distribution of the cysticerci was, shoulder muscle (39.5°%), heart (33.9 %), neck muscle (13.8°%), tongue (10.1°%), masseter muscles (1.8°%) and diaphragm (0.9°%). Of the 1171 hydatid cysts collected 223 (19.0°%) were fertile, 505 (43.1 °%) sterile, 49 (29.8 °%) calcified and 94 (8.0 °%) contained pus. A greater proportion of fertile cysts were found in the lungs than in other organs. It was concluded that these zoonotic cestodes deserve due attention to safeguard public health, and that further studies are needed on genotyping, epidemiology and public health importance of Echinococcus granulosus in the study area.

Hydatidosis, caused by the larval stage of the dog tapeworm, Echinococcus granulosus is considered to be an important condition in herbivores such as cattle, buffaloes, sheep and goats. Due to its presence in the food animals meant for meat purpose, the disease is considered to be of economic and public health significance owing to the impact it produces on the meat industry by condemnation of the affected organs. Food animals such as sheep, goats get infected by accidental ingestion of contaminated feed and water with the eggs of the tapeworm. Development of the cysts in the intermediate host such as sheep and goats occurs in the lungs, liver and other visceral organs. The disease remains asymptomatic in most cases in spite of the presence of the hydatid cysts for a long period. A study was undertaken to find out the incidence of the hydatid cysts in sheep and goats in the Corporation slaughter house by examination of the carcasses of the slaughtered sheep and goats. By the study, an overall incidence of 6.5% in sheep and 5.8% in goats were observed with the sheep harbouring more number of fertile hydatid cysts which plays a major role in dissemination of infection to dogs.
