Genetic relationship of Salmonella isolates found in subcutaneous abscesses in leopard geckos (Eublepharis macularius)

The article describes the occurrence and phylogenetic relationship of Salmonella isolates found in subcutaneous abscesses of leopard geckos. The aim of the study was to determine the cause of the abscesses and to characterise isolated Salmonella strains. Samples of abscesses from five animals and internal organs (lungs, liver, and gut) of three of them were tested for Salmonella according to the PN-EN ISO 6579:2002/A1:2007 standard. The antimicrobial resistance was evaluated by minimal inhibitory concentrations and the genetic similarity of the isolates was assessed with pulsed field gel electrophoresis (PFGE). In total, seventeen Salmonella isolates belonging to five different serovars were found to be susceptible to all tested antimicrobials except streptomycin. The serovars were S. Hadar, S. Fluntern, S. Tennessee, S. enterica subsp. salamae 55:k:z₃₉, and S. Kentucky. Up to three serovars from different organs were isolated from the same individual. In two geckos, Salmonella were detected in the lungs. In three serovars, XbaI-PFGE typing revealed indistinguishable isolates from organs and abscesses. Multiple Salmonella serovars might be involved in abscess formation and infections. The occurrence of the same PFGE profiles of the isolates may testify to the role of opportunistic organisms in causing infection.

Introduction: Data collection on the Salmonella occurrence is crucial in effective implementation of different actions or control programmes aiming to protect consumers’ health and to reduce the level of Salmonella prevalence in farm animals. The goal was to describe Salmonella serovar distribution along the food chain in Poland during 2010–2015 and to identify their epidemiological importance.Material and Methods: Slide agglutination according to White-Kauffmann-Le Minor scheme was used to identify Salmonella serovars of 6,928 isolates originating from animals, food, feeds, and fertilisers.Results: In total, 160 Salmonella serovars were identified. Differences in serovar distribution were observed depending on animal species. Among isolates from hens, S. Enteritidis and S. Infantis were the most prevalent. Serovar pattern in turkeys differed from those in hens, with S. Kentucky, S. Newport, S. Saintpaul being the most prevalent. Monophasic S. Typhimurium was predominant in pigs. Serovars found in food reflected those observed among livestock animals. Nine out of the ten most prevalent serovars in animals and humans were also found in organic fertilisers.Conclusion: Serotyping of large number of isolates from different sources is essential for insight on emerging serovars and trends of Salmonella occurrence. This may increase the value of epidemiological data and result in updating of Salmonella control programmes to target further epidemiologically important serovars in animals and better protection of consumers’ health.

Salmonellosis is a zoonotic disease, and Salmonella spp. can sometimes be found in dogs and cats, posing a risk to human health. In this study, the prevalence and antimicrobial susceptibility of faecal Salmonella were investigated in pet dogs and cats in Xuzhou, Jiangsu Province, China. Faecal samples from 243 dogs and 113 cats, at seven pet clinics, were tested between March 2018 and May 2019. Each Salmonella isolate was characterised using serotyping and antimicrobial susceptibility tests. The prevalence of Salmonella was 9.47% in dogs and 1.77% in cats. Among the 25 isolates, eight serotypes of Salmonella enterica subsp. enterica were detected, S. Kentucky (n = 11), S. Indiana (n = 5), and S. Typhimurium (n = 4) predominating. S. Derby, S. Toucra, S. Sandiego, S. Newport, and S. Saintpaul all occurred singly. The 23 Salmonella strains found in dogs were from seven different serovars, while the two strains in cats were from two. The highest resistance rates were found for tetracycline (92%), azithromycin (88%), cefazolin (84%), nalidixic acid (80%), ampicillin (80%), ceftriaxone (80%), and streptomycin (76%). Resistance to three or more antimicrobial agents was detected in 24 (96%) isolates. Most of the S. Kentucky and S. Indiana isolates were multi-drug resistant to more than 11 agents. The carriage rate was far higher in dogs than in cats from Xuzhou. Some isolated strains were highly resistant to antimicrobials used to treat infections in humans and pets, which may raise the risk of humans being infected with multi-drug resistant Salmonella via close contact with pets.

A temporal study was carried out to determine Salmonella prevalence, trends, major serovars, and their clusters from environmental samples, in poultry breeder flocks in Ontario between January 1998 and December 2008. Surveillance data were obtained from the Ontario Hatchery and Supply Flock Policy. Logistic regression with a random effect for flock was used to identify factors [poultry type, year (trend) and season] associated with the prevalence of Salmonella. A cluster detection test was used to identify clusters of common serovars. The period prevalence of Salmonella was 47.4% in broiler-breeder, 25.7% in layer-breeder, and 19.6% in turkey-breeder flocks. The overall trend in the prevalence of Salmonella was decreasing for all breeder types, due primarily to decreasing trends of Salmonella Heidelberg. The seasonal effects varied by year with the highest probability of Salmonella occurring in different seasons. The 4 most common serovars identified were Salmonella Heidelberg, Kentucky, Hadar, and Typhimurium in broiler-breeders; Salmonella Heidelberg, Brandenburg, Thompson, and Typhimurium in layer-breeders; and Salmonella Heidelberg, Saintpaul, Brandenburg, and Muenster in turkey-breeders. Salmonella Enteritidis was infrequently isolated in all poultry breeder types. Temporal clusters of different serovars were identified in all poultry breeder types. Clusters of Salmonella Heidelberg, Typhimurium, and Hadar from environmental samples from breeder flocks were detected during a similar period to clusters from hatchery fluff samples from the same population. Therefore, interventions at the breeder flock-level might help to reduce transmission of Salmonella from breeder flocks to hatcheries and possibly, to lower levels of the poultry production chain.

Objective: To determine whether the concentration of airborne virulent Rhodococcus equi varied by location (stall vs paddock) and month on horse farms. Sample: Air samples from stalls and paddocks used to house mares and foals on 30 horse breeding farms in central Kentucky. Procedures: Air samples from 1 stall and 1 paddock were obtained monthly from each farm from January through June 2009. Concentrations of airborne virulent R equi were determined via a modified colony immunoblot assay. Random-effects logistic regression was used to determine the association of the presence of airborne virulent R equi with location from which air samples were obtained and month during which samples were collected. Results: Of 180 air samples, virulent R equi was identified in 49 (27%) and 13 (7%) obtained from stalls and paddocks, respectively. The OR of detecting virulent R equi in air samples from stalls versus paddocks was 5.2 (95% confidence interval, 2.1 to 13.1). Of 60 air samples, virulent R equi was identified in 25 (42%), 18 (30%), and 6 (10%) obtained from stalls during January and February, March and April, and May and June, respectively. The OR of detecting virulent R equi from stall air samples collected during May and June versus January and February was 0.22 (95% confidence interval, 0.08 to 0.63). Conclusions and Clinical Relevance: Foals were more likely to be exposed to airborne virulent R equi when housed in stalls versus paddocks and earlier (January and February) versus later (May and June) during the foaling season.

This study assessed the protective efficacy of oral vaccination with 2 experimental attenuated Salmonella Typhimurium-vectored vaccines for necrotic enteritis in protecting chickens against intestinal colonization by common serovars of Salmonella belonging to the 4 major serogroups affecting chickens. Birds were vaccinated orally with 1 × 108 colony-forming units (CFU) of 1 of the vaccine strains χ9241 and χ9352, which express a plasmid-encoded partial recombinant hypothetical protein gene (tHP) of Clostridium perfringens, at days 1 and 7 of age, and then were challenged at 14 d of age with 106 CFU of Salmonella serovars Anatum, Enteritidis, Heidelberg, Kentucky, or Typhimurium (representative serovars of serogroups B, C, D, and E). Birds were necropsied at 4 wk of age, and samples were collected to determine reduction in tissue and intestinal colonization. The chickens vaccinated with χ9241-tHP showed reduced colonization by Salmonella Enteritidis (serogroup D) and by Salmonella Heidelberg and Salmonella Typhimurium (serogroup B) compared with the control birds. No reduction in colonization was observed in the chickens vaccinated with χ9352-tHP. There was an association between the efficacy of these vaccine strains in protecting against necrotic enteritis, assessed on an earlier occasion, and their efficacy in protecting against Salmonella colonization. Thus, the choice of an attenuated Salmonella Typhimurium vaccine vector for delivery of heterologous antigens to chickens should be based partly on the vaccine’s value in protecting against colonization by serovars within serogroups B and D. Such vectors would have the additional benefit of reducing colonization of important Salmonella serovars.

The efficacy of febantel paste formulation (6 and 12 mg/kg) against natural infections of gastrointestinal helminths in lambs (n = 33) in Kentucky was evaluated in a controlled test. For the test, 23 lambs were treated orally (17 at 6 mg/kg and 6 at 12 mg/kg) and 10 lambs were not treated. Removals at both dosages in treated lambs were 95% to 100% for species of immature and mature Haemonchus, Trichostrongylus, and Cooperia; and mature Ostertagia females, Nematodirus, and Strongyloides. For immature Nematodirus, removals were 92% and 77% at the dosages of 6 and 12 mg/kg, respectively. Only a few specimens (av less than 100) of some other species or stages were found in the nontreated group and removal of them (at both dosages) were 94% to 100% for Ostertagia (immature and males), Strongyloides (immature), Oesophagostomum (immature), and Monieiza (mature); and 61% (at 6 mg/kg) and 100% (at 12 mg/kg) for Capillaria (mature), 0% for Trichuris (mature, at both dosages), and 67% (at 6 mg/kg) and 100% (at 12 mg/kg) for Oesophagostomum (mature).

Observations were completed over an 18-year period (1966 through 1983) in Thoroughbred mares (15/year) and yearlings (11 to 24/year) on a farm where benzimidazole-resistant small strongyles had emerged previously (1962 to 1965). This farm was operated as a closed, nonboarding type, which included a racing stable for its home-bred foals. At 2-week intervals, counts of worm eggs per gram of feces (EPG) and larvae per gram of feces were done to monitor strongyle infections and efficacies of bimonthly (every 8 weeks) antiparasitic treatments that were administered by stomach tube or were fed (dichlorvos pellets) to 1 group of yearlings during a 7-year period (1970 through 1976). The study included several drugs or mixtures, including thiabendazole (TBZ), phenothiazine (PTZ) + piperazine (PPZ)-carbon disulfide (CS2) complex, PTZ + PPZ + trichlorfon (TCF), dichlorvos, and pyrantel pamoate. These were used selectively in treated subgroups (usually 3/year) of mares and yearlings. The horses in drug treatment subgroups grazed together on common pastures; thus, efficacy comparisons between drugs were limited or nullified. However, annual mean EPG and larvae per gram of feces counts of mares and yearlings tended to increase over time, and for the yearlings treated with TBZ + PPZ and PTZ + PPZ-CS2, the buildup of these mean counts was statistically significant (P less than 0.05 for regression coefficients). These trends of annual mean counts to increase were a general index of loss of anthelmintic activity of the 3 drug mixtures containing PPZ (TBZ + PPZ, PTZ + PPZ-CS2, and PTZ + PPZ TCF); however, trends were indicated more definitively by 2 other indices: (1) loss of effectiveness in reduction of EPG counts 2 weeks after treatment and (2) decrease in performance for complete clearance of strongyle eggs (EPG, 0) 2 weeks after treatment. Loss of effectiveness over time was statistically significant (P less than 0.001) for the mares only when data was pooled for all 3 treatments (TBZ + PPZ, PTZ + PPZ-CS2, and PTZ + PPZ + TCF); however, the decreasing effectiveness was statistically significant (P less than 0.05) for TBZ + PPZ and PTZ + PPZ-CS2 treatment of yearlings. Statistically, clearance performance in mareswas significant for treatments with TBZ + PPZ (P less than 0.003) and PTZ + PPZ + TCF (P less than 0.005) and approached significance with PTZ + PPZ-CS2 (P greater than 0.06). Clearance performances in yearlings were highly significant (P less than 0.0003) for treatments with TBZ + PPZ and PTZ + PPZ-CS2. These analyses of data indicate that the intense and prolonged (greater than or equal to 18 years) use of PPZ as a common component in several of the drug mixtures had resulted in the selection of a segment of small strongyles resistant to PPZ. Multiple resistance of this population of small strongyles includes several drugs: PTZ, the benzimidazoles (except oxibendazole and albendazole), possibly pyrantel, and PPZ. Side effects of drug treatments were not apparent during the prolonged period of this study.
