Comparative histopathology of the lymph nodes, spleen, liver and kidney in experimental ovine trypanosomosis

The infection of Yankassa rams with three important trypanosome species affecting livestock, namely, Trypanosoma congolense, T. vivaxand T. bruceiproduced both acute and chronic fatal conditions. Chronic infections were induced in the three infections by the application of subcurative doses of diaminazene aceturate (Berenil®). Pathological changes in the infected animals included splenomegaly and hepatomegaly which were more pronounced in acute than in chronic T. congolense infection. However, these changes were more severe in chronic than in acute T. vivax infection. While splenomegaly was more pronounced in chronic T. brucei infection than in acute, hepatomegaly and lymphadenopathy were more severe in acute than in the chronic condition. The increases in size of the spleen, lymph nodes and liver were associated with congestion, increases in cell density related to increased immunological reactions in the spleen and lymph nodes as well as increase in numbers, size and activity of the phagocytic cells in these organs.

Abortions in domestic ruminants cause significant economic losses to farmers. Determining the cause of an abortion is important for control efforts, but it can be challenging. All available diagnostic methods in the bacteriology laboratory should be employed in every case due to the many limiting factors (autolysis, lack of history, range of samples) that complicate the investigation process. The purpose of this study was to determine whether the recovery of diagnostically significant isolates from domestic ruminant abortion cases could be increased through the use of a combination of the existing aerobic culture and Brucella selective method with methods that are commonly recommended in the literature reporting abortion investigations. These methods are examination of wet preparations and impression smears stained by the modified Ziehl-Neelsen method, anaerobic, microaerophilic, Leptospira, Mycoplasma and fungal culture. Samples of placenta and aborted foetuses from 135 routine clinical abortion cases of cattle (n = 88), sheep (n = 25) and goats (n = 22) were analysed by the new combination of methods. In 46 cases, bacteria were identified as aetiological agents and in one case a fungus. Isolation of Brucella species increased to 7.4% over two years compared with the previous 10 years (7.3%), as well as Campylobacter jejuni (n = 2) and Rhizopus species (n = 1). Salmonella species (5.9%) and Trueperella pyogenes (4.4%) were also isolated more often. In conclusion, the approach was effective in removing test selection bias in the bacteriology laboratory. The importance of performing an in-depth study on the products of abortion by means of an extensive, combination of conventional culture methods was emphasised by increased isolation of Brucella abortus and isolation of C. jejuni. The combination of methods that yielded the most clinically relevant isolates was aerobic, microaerophilic, Brucella and fungal cultures.

The present study describes the use of electrochemiluminescenceimmunoassay (ECL) with the specific human kit for measuring plasma testosterone and progesterone in ovine. This study wascarried out on 16 mixed age animals of the local breed. They were divided into two groups of both sexes, namely Male-1 Group,Male-2 Group, Female-1 Group and Female-2 Group. Blood samples were collected from the jugular vein into tubes containing EDTAand centrifuged at 1500 rpm for 20 min. Plasma was rapidly separated and stored at -20° C until assayed. The reproducibilityinter- and intra-assay of the P4- and T-ECL is satisfactory (2.11-7.3 %). The accuracy (93-102 %) and the test of parallelism were largely acceptable. No cross-reaction was observed with the different hormones including PMSG,hCG, progesterone,testosterone, oxytocin and PGF2α when concentrations of 10 UI/ml and 10-3 UI/ml were used. T concentrations were higher in Male-1 Group than in Male2 Group (1.67 ± 1.15 and 0.38 ± 0.45 ng/ml, respectively). The female-1 Group showed very high P4 concentrations (15.17 ± 4.91 ng/ ml) compared to the Female-2 Group (0.08 ± 0.04 ng/ml). In this study a total of 2230 sheep (one-three years of age) were serologically surveyed in three selected areas in Libya (Western, Middle and Southern areas) to specify foci of infection and determination of the prevalence of ovine brucellosis using Rose Bengal Plate Test and Rivanol test. Prevalence of brucellosis in this study revealed 4%, 0%and 0%, respectively. Only the western area showed positive cases, while the Middle and Southern areas showed no serological evidence of brucella infection.

The objective of this study was to compare the efficacy of tulathromycin (TUL) with a combination of florfenicol (FFC) and long-acting oxytetracycline (LAOTC) in the treatment of naturally occurring undifferentiated respiratory diseases in large numbers of sheep. In this study, seven natural outbreaks of sheep pneumonia in Garmsar, Iran were considered. From these outbreaks, 400 sheep exhibiting the signs of respiratory diseases were selected, and the sheep were randomly divided into two equal groups. The first group was treated with a single injection of TUL (dosed at 2.5 mg/kg body weight), and the second group was treated with concurrent injections of FFC (dosed at 40 mg/kg bwt) and LAOTC (dosed at 20 mg/kg bwt). In the first group, 186 (93%) sheep were found to be cured 5 days after the injection, and 14 (7%) sheep needed further treatment, of which 6 (3%) were cured, and 8 (4%) died. In the second group, 172 (86%) sheep were cured after the injections, but 28 (14%) sheep needed further treatment, of which 10 (5%) were cured, and 18 (9%) died. This study revealed that TUL was more efficacious as compared to the combined treatment using FFC and LAOTC. As the first report, this field trial describes the successful treatment of undifferentiated respiratory diseases in large numbers of sheep. Objective: A retrospective survey was designed to determine the prevalence and factors involved in surgical cases at the Veterinary Teaching Hospital (VTH), Bangladesh Agricultural University from June 2014 to June 2017. Materials and materials: In total, 1042 surgical cases of food animal (large ruminants: n=564, and small ruminants: n=493) and 26 non-food animal (mono-gastric animal) were recorded from patient register book and case recording card. Data were analyzed by Epi Info TM software and frequencies were calculated for different variables using Statistical Package for Social Science (SPSS) software. Result: In large ruminant, hernia (16.13%) ranked top (90% umbilical and 10% lateral) followed by fracture (14.89%), abscess (14.54%), umbilical myasis (10.46%), atresia ani (5.85%) and naval ill (4.07%). Among the reported cattle, 87.41% were crossbred and 12.59% were indigenous. Calf, heifer and adult cattle were 55.32, 10.29 and 34.39% respectively. In small ruminants, castration (32.94%; n=138) ranked top followed by myasis (10.55%) naval ill (10.31%), abscess (7.44%), dystocia (6.24%) and urolithiasis (5.49%). Based on surgical classification in large and small ruminants, 37.56 and 42.50% were reported for general surgery, whereas 28.71 and 7.15% for congenital, 11.18 and 13.12% for gynecological and 2.65 and 37.23 % for andrological problems, respectively. Male and female ratio was 1:1.31 and 2:1 respectively in large and small ruminants, respectively. Conclusion: This study emphasizes the factors related to successful surgical cases management at VTH. The results may help in controlling surgical related cases in Mymensingh region of Bangladesh. [J Adv Vet Anim Res 2018; 5(1.000): 81-87]

The expression of milk proteins in vitro is essential to exploit the mammary gland cells as a biological model. Enzymatic tissue disaggregation has been widely used to establish mammary cell culture, but its effect in long-term ovine mammary cell culture is not completely elucidated. This study aimed at comparing mechanical/enzymatic and mechanical dissociation methods to establish ovine mammary cell culture. We compared cellular differentiation induced by lactating ewe sérum or fetal bovine serum based on the gene expression levels of milk proteins (beta-lactoglobulin, alpha s1-casein, and betacasein). Mechanically dissociated cells were positive immunostaining for cytokeratin 8.13, such as mammary epithelial cells. These cells are responsible for milk protein expression and they are low immunostaining for vimentin, mesenchymal marker. Mechanical/enzymatic dissociation cells were positive for vimentin. The fastest cell growth (cell/hour) was observed in the mechanical dissociation group cultured with 10% fetal bovine serum medium. Mechanically and mechanically/enzymatically derived cells were able to express beta-casein and beta-lactoglobulin, but not alpha s1-casein. The relative expression of beta-lactoglobulin was not affected by the tissue dissociation method or culture media, beta-casein relative expression was down regulated in mechanically dissociated cells cultured in the presence of lactating ewe serum, (P = 0.019). Beta-casein relative expression was also down regulated in mechanically/enzymatically dissociated cells cultured with fetal bovine sérum (P = 0.021). In the present conditions, we conclude that mechanical dissociation followed by culture with 10% of fetal bovine serum was the most efficient method to induce milk proteins’ mRNA expression by ovine mammary epithelial cells in vitro.

Sonography is a safe and non-invasive imaging modality, with the added benefit of providing dynamic images. Power Doppler sonography allows blood vessel identification regardless of flow intensity or direction. A one-year-old Ile de France ewe was admitted to the FMVZ-USP Veterinary Hospital with a complete tibial fracture. The animal was submitted to general inhalation anesthesia and fracture repair using a 3.5 mm locking compression plate. Power Doppler sonography was used to assess neovascularization over the course of the bone healing process. Formation of new vessels was noted between postoperative days 14 and 21; new vessels regressed progressively on subsequent assessments (postoperative days 28 and 35). Power Doppler sonography enabled early detection of newly formed blood vessels at the fracture site. The non-invasive nature of this imaging modality prevented patient stress and provided useful information on the progression of bone healing. Early neovascularization was thought to reflect successful postoperative healing of the tibial fracture described.

The present study aimed to estimate the prevalence of antibodies against Brucella ovis-epididymitis, smooth-Brucella, leptospirosis, toxoplasmosis and Maedi-visna in sheep slaughtered in Minas Gerais, Brazil and to study their simultaneous occurrence, including caseous lymphadenitis, at sheep and flock levels. The study was conducted at a sheep slaughterhouse with Federal Inspection Service. Sera from 594 animals from 21 flocks were collected, in 2007. The agar gel immunodiffusion (AGID) was employed to detect anti-B. ovis and anti-Maedi Visna antibodies, whereas Rose Bengal (RB) and the 2-mercaptoethanol test (2ME) were used to test anti-smooth Brucella antibodies. For the detection of anti-Leptospira antibodies, sera were examined by microscopic agglutination test (MAT), while for the detection of IgG antibodies to Toxoplasma gondii ELISA was used. Prevalence of antibodies against smooth Brucella, B. ovis-epididimitis, Leptospira spp., toxoplasmosis and Maedi-Visna found in sheep from Minas Gerais was 0.00%, 24.04%, 25.96%, 10.46% and 3.08%, respectively; whereas the seroprevalence in flocks was 0.00%, 80.95%, 90.48%, 71.43% and 23.81%, respectively. Moreover, when data on antibodies anti-Corynebacterium pseudotuberculosis, previously obtained, were included, about 60% of the flocks showed animals that were exposed to four or more of the studied agents. However, only 25.47% of the sheep exhibited simultaneously antibodies against more than one pathogen. Thus, data from the present study on sheep slaughtered in Minas Gerais, Brazil, showed no antibodies to smooth-Brucella and a low frequency of antibodies anti-Maedi Visna lentivirus, and a high and widespread seroprevalence of B. ovis, Leptospira spp., and T. gondii among animals and flocks.
