The combined effects of the yeast Saccharomyces cerevisiae and Aspergillus niger on the haematological and biochemical parameters of cultured juvenile beluga (Huso huso)

BACKGROUND: Probiotics, in form of microbial supplements, are known to be a suitable alternative for antibiotics and can affect the health indicators of host. Objectives: The present study was conducted to assess the combined effects of dietary autochthonous Saccharomyces cerevisiae and Aspergillus niger on haematological and serumbiochemicalparameters of beluga sturgeon (Huso huso) juveniles. Methods: This study was based on a completely randomized design with 4 treatments and 3 replicates on beluga juveniles with average weight of (mean ±SE) 31.8±2.81g. Beluga Juveniles were divided randomly into 12 fiber glassy tanks with density of 30 fish per tank and were fed with diet contain dietary probiotic with density of 2×106 (Cells/g) for the first treatment, 4×106 (Cells/g) for the second treatment, 6×106 (Cells/g) for the third treatment and basal diet without probiotic for the control group for 8 weeks. Results: Diet supplementing with concentration of 6×106 (Cells/g), significantly improved serum biochemical parameters (p<0.05), however hematological parameters were affected by supplemented diet with probiotics that showed no significant difference in comparison with the control group (p>0.05). Also results indicate that growth factors were improved in experimental treatments in comparison with the control group. Conclusions: The results showed that the use of combination of these species with studied concentrations can improve the performance of some biochemical parameters such metabolites factors, immune, enzymes and serum electrolytes of belugajuveniles. It is recommended that the concentration of A. niger and S. cerevisiae, used for third treatment be used as an immune stimulator for beluga juveniles.

Wound healing and tissue repair are necessary to ensure survival and health of any organism. The aim of this study was to investigate the impact of supplementation with chromium carbochelate (CC) and Saccharomyces cerevisiae (SC) on wound healing in tropical teleost fish Piaractus mesopotamicus. Thus, fish were distributed into four groups: a) control (without supplementation); b) supplemented with 18 mg/kg of chromium carbochelate; c) supplemented with 0.3% of S. cerevisiae and d) supplemented with an association of both supplements. After 105 days of feeding, full-thickness skin incisions (2.0 x 1.0 x 0.25 cm) were performed removing epidermis and dermis. Macroscopic and histologic observations were carried out at 1, 3, 7, 14, 21, 28, and 35 days after wounding to monitor the healing rate. Opposing fronts advanced gradually and faster each day demonstrating a progressive increase in the healing process over time. The inflammatory process was exacerbated and expansive, with an increase in mucous cells and chromatophores. Although no significant differences were observed between groups on wound retraction and microscopic parameters, fish supplemented with CC and SC showed faster re-epithelialization, greater degree of organization of collagen fibers, and higher neovascularization. We concluded that supplementation with S. cerevisiae and chromium carbochelate improves specific aspects of cutaneous healing process in pacu.

Objective: The study was conducted to determine the effect of inoculants of different types and doses on the nutrient quality and in vitro digestibility of fermented rice bran. Materials and Methods: The study was designed using a completely randomized design with a 3 × 3-factorial pattern. The first factor was the type of inoculum, consisting of Saccharomyces cerevi¬siae (SC), Effective Microorganism-4, and Saus Burger Pakan (SBP). The second factor is inoculum doses, which are as follows: levels 2%, 4%, and 6%. The variables measured included chemical composition, fiber fraction content, dry matter digestibility and organic matter digestibility. Results: The results showed that the type of inoculation treatment and the doses of inoculation did not affect the dry matter (DM) content of fermented bran, and the organic matter content of fermented bran was only affected by the inoculation dose factor (p < 0.05). The highest crude protein and Extract Ether (EE) were obtained in the SBP inoculants, which increased linearly with increasing inoculation doses (p < 0.05). While a significant decrease (p < 0.05) occurred in crude fiber content. The cellulose, hemicellulose, lignin, acid detergent fiber (ADF), and neutral deter¬gent fiber (NDF) fractions were significantly lower in the SBP treatment as the dose increased. The SBP inoculant type produced the highest DMD (p < 0.05) but showed a response that was not different from the SC inoculant treatment for OMD. Increasing inoculation doses of 2%, 4%, and 6% linearly increased the DMD and OMD of fermented bran (p < 0.05). Overall, inoculant application on fermented bran showed an interaction effect except for the components of DM, EE, ADF, NDF, and DMD of fermented bran. Conclusions: It was concluded that the SBP at 6% and their combination resulted in the best chemical quality and digestibility of rice bran. [J Adv Vet Anim Res 2022; 9(4.000): 625-633]

In this study, we produced iron-fortified yeast (Saccharomyces cerevisiae) producing Sus scrofa ferritin heavy-chain to provide iron supplementation in anemic piglets. We determined whether iron-ferritin accumulated in recombinant yeasts could improve iron deficiency in mice. C57BL/6 male mice exposed to Fe-deficient diet for 2 weeks were given a single dose of ferrous ammonium sulfate (FAS), ferritin-producing recombinant yeast (APO), or APO reacted with iron (Fe²+) (FER). The bioavailability of recombinant yeasts was examined by measuring body weight gain, hemoglobin concentration and hematocrit value 1 week later. In addition, ferritin protein levels were evaluated by western blot analysis and iron stores in tissues were measured by inductively coupled plasma spectrometer. We found that anemic mice treated with FER exhibited increased levels of ferritin heavy-chain in spleen and liver. Consistently, this treatment restored the iron concentration in these tissues. In addition, this treatment significantly increased hemoglobin value and the hematocrit ratio. Furthermore, FER treatment significantly enhanced body weight gain. These results suggest that the iron-fortified recombinant yeast strain is bioavailable.

This study was conducted to investigate the effects of feeding probiotics (gene modified yeast) on the growth performances in pigs. In pigs, this study investigated the effects of dietary probiotics which contained antibacterial probiotics (OR-7, bacteriocin, gene modified yeast) on growth performances and feed efficiency in pig farm. A total of 150 pigs were studied. The treatments are probiotics yeast (PY) 0.3% (basal diet + 0.3% plasmid modified probiotics), PY 0.5% (basal diet + 0.5% plasmid modified probiotics), yeast (Y) 0.3% (basal diet + 0.3% probiotics), Y 0.5% (basal diet + 0.5% probiotics) and control (basal diet). Weight gain, feed intake and feed efficiency were periodically recorded for 90 days. The treatment group trended higher weight gain, feed intake and feed efficiency than control. And, the PY group trended higher weight gain, feed intake and feed efficiency than Y group.

The main objective of the study reported here was to generate a panel of monoclonal antibodies (MAB) to bovine neutrophil surface antigens, and to identify MAB that modulate neutrophil chemotaxis, respiratory burst, and phagocytosis. A further objective was to study MAB reactivity with resting and activated neutrophils, to identify activation antigens and adhesion molecules. A panel of 14 MAB was generated by producing murine hybridomas. Neutrophils incubated with MAB at 4 C for 2 hours were used in chemotaxis, respiratory burst, and phagocytosis assays. Chemotaxis was evaluated in Boyden chambers, using Escherichia coli endotoxin-activated fetal bovine serum as the chemoattractant. Respiratory burst was determined by measuring chemoluminescence of neutrophils incubated with 5-amino-2,3-dihydro-1,4-phthalazinedione, and serum opsonized zymosan. Phagocytosis was determined by flow cytometry, using fluorescein-labeled Staphylococcus aureus. The MAB S7G8, S5F8G10, S7E10, and S5F8B8 enhanced chemotaxis (to > 125% of control). The MAB S7E10 and S8D9 enhanced respiratory burst activity (to > 125% of control), whereas MAB S2G8, S4G10, S8G10, and S5F8B8 caused inhibition (to < 75% of control). The MAB S2G8, S4G10, S8G10, and S5F8G10 enhanced phagocytosis (to > 125% of control). Chemotaxis, respiratory burst, and phagocytosis values of neutrophils not bound with MAB served as controls for comparison. The MAB binding for nonactivated neutrophils (at 4 C) ranged from 9 to 100%, and for activated neutrophils (at 37 C; at 37 C with phorbol myristate acetate) from 90 to 100%. Binding of MAB S4F5, S5F8B8, S6C6, S7E10, S8D9, and S5F8G10 increased when neutrophils had been incubated at 37 C. Binding of these MAB was further increased after incubation with phorbol myristate acetate (100 ng/ml) at 37 C, indicating recognition of activation antigens by MAB. The MAB generated in this study appeared to be potential candidates for studying mechanisms of neutrophil function and for enhancing neutrophil function in vitro and in vivo.

This study aimed to evaluate the effect of probiotics on the microbial and physiochemical quality of chicken nuggets. In the current investigation, different concentrations of Bacillus clausii and Saccharomyces cerevisae were added as probiotics to chicken nuggets. The proximate composition, physicochemical properties, and microbial quality of prepared chicken nuggets were assessed over 21 days at -18oC. The findings of this investigation demonstrated the ability of probiotics such as Bacillus clausii and Saccharomyces cerevisae to depress the growth and multiplication of aerobic and psychotrophic bacteria as well as the yeast and mold count over the storage period. Varying preparations influenced the mean pH, peroxide, and TBA values during the storage of chicken nuggets (p≤0.05). There was a significant decrease in moisture, protein, fat, and ash content compared to the control. In conclusion, additive supplementation of Bacillus clausii and Saccharomyces cerevisae improved the chicken nuggets' shelf life attributes, physicochemical characteristics, and microbial quality.

In this study, we investigated whether β-glucan from Saccharomyces cerevisiae exerts beneficial effects on mucosal immunity in an ovine ruminal explant (ORE) model. Once the ORE model was established, viability was assessed through histological change, E-cadherin expression, CK-18 and Ki-67 distribution. Then, the OREs were co-cultured with β-glucan, following which, gene and protein expression levels of sheep β-defensin-1 (SBD-1), pro-inflammatory interleukin (IL)-6, and anti-inflammatory IL-10 were detected using quantitative real-time polymerase chain reaction (qPCR) and enzyme-linked immunosorbent assay (ELISA). Hematoxylin & eosin staining, qPCR, and immunohistochemistry showed that the overall ORE structure was intact after 96 hours in culture, but explants cultured for more than 24 hours showed epithelial degradation. Therefore, we performed the follow-up test within 24 hours. qPCR and ELISA revealed that the gene and protein expression levels of SBD-1, IL-6, and IL-10 in the OREs significantly increased (P < 0.05) after treatment with β-glucan compared with controls. This study identified the feasibility and optimal conditions of ORE culture and demonstrated that β-glucan activates SBD-1, IL-6, and IL-10 secretion in OREs to promote mucosal immunity.
Afficher plus [+] Moins [-]Comparison of the chemiluminescence responses of bovine neutrophils to differently opsonized zymosan particles

Stimulatory effects of 6 zymosan preparations on luminol-dependent chemiluminescence (CL) responses of isolated bovine neutrophils were compared. Unopsonized zymosan particles and zymosan particles opsonized with bovine IgG1, IgG2, fresh serum, or serum from which zymosan-specific antibodies, but not complement, had been removed (C3- serum) induced strong CL responses, with nearly equal maximal peaks in the presence of extracellular Ca2+ and Mg2+, whereas the response to fetal bovine serumopsonized zymosan particles was markedly low. Removal of extracellular divalent cations almost completely blocked the CL reaction triggered by unopsonized, IgG1-opsonized, C3-opsonized, and fetal bovine serum-opsonized zymosan particles. By contrast, no change in the respiratory burst activity induced by serum-opsonized zymosan and only partial reduction in the response to IgG2-opsonized zymosan were seen under these conditions. Further experiments were performed with 4 zymosan preparations on neutrophils isolated from 2 calves with a genetic deficiency of CD11/CD18 membrane antigens. The unopsonized zymosan-induced CL reaction was absent in these cells. A reduced, but clear, response was observed with C3-opsonized zymosan. Unexpectedly, in the absence of extracellular Ca2+ and Mg2+ , serum-opsonized zymosan failed to generate the respiratory burst, whereas response to IgG2-opsonized zymosan was normal in the CDll/CD18-deficient neutrophils. These findings indicate that unopsonized zymosan may act in a divalent cation-dependent manner at the receptor for C3bi in bovine neutrophils, as it has been shown to do in the human system. In addition, it seems that IgG2-Fc receptors capable of signaling the respiratory burst in the absence of extracellular Ca2+ and Mg2+ exist on bovine neutrophils.
