The characteristics of immune factors in somatic cells from lactating dairy cows and their association with commensal bacteria in normal milk have not been clarified. This study investigated the relationship between the pathogenic bacteria in milk and somatic cell immune factors in healthy lactating cows. In total 44 healthy Holstein cows were studied on one farm. Milk samples were collected aseptically using a cannula and these samples were cultured for detection of bacteria and analysis of mRNA of immune factors expressed by somatic cells. Cows were divided into two groups based on the microbial status of their milk samples: 12 cows showed bacteria in cultures (positive group), and the other 32 cows did not (negative group). The mRNA levels of IL-6, lactotransferrin, and cathelicidin expressed by somatic cells after milking decreased significantly compared to those before milking in both groups (P < 0.05). There were significantly lower mRNA levels of IL-6 and cathelicidin in the positive group compared to those in the negative group before milking. These results suggest that mRNA levels of IL-6 and cathelicidin expressed by the somatic cells may be affected by the presence of bacteria in healthy lactating dairy cows.

Bovine mastitis is an inflammatory disease of the udder that causes important economic losses in the animal breeding and dairy product industries. Nowadays, the conventional livestock antibiotic treatments are slowly being replaced by alternative treatments. In this context, the main aim of this study was to evaluate the efficacy of natural products in alternative treatment of bovine mastitis. Two natural formulations with previously suggested in vitro antimicrobial effect were tested in vivo on mastitic cows. Animals with a positive diagnosis for mastitis (n = 20) were divided into three treatment groups: two groups (n = 8) were administered formulations of propolis, alcoholic extracts of Brewers Gold and Perle hops, plum lichen, common mallow, marigold, absinthe wormwood, black poplar buds, lemon balm, and essential oils of oregano, lavender, and rosemary designated R4 and R7 (differing only in the latter being more concentrated) and one group (n = 4) a conventional antibiotic mixture. In vivo efficacy of treatments was evaluated by somatic cell and standard plate counts, the treatment being considered efficacious when both parameters were under the maximum limit. R7 was effective in the most cases, being therapeutically bactericidal in six out of eight cows, while R4 gave good results in three out of eight cows, and conventional antibiotics cured one out of four. These results suggest the possible therapeutic potential of these natural products in bovine mastitis.

Comparative analysis of the karyotype structure was made in two hedgehog species: the northern white-breasted hedgehog (Erinaceus roumanicus) and the African pygmy hedgehog (Atelerix albiventris). The cytogenetic analysis used differential staining techniques (DAPI, Ag-NOR, and C-banding/DAPI) and sequential QFQ/FISH banding with NOR20 and TEL20 probes which showed 45S rDNA and (TTAGGG)ₙ repeat sequences, respectively, on hedgehog chromosomes. It was confirmed that the somatic cells of the hedgehogs have a constant number of chromosomes (2n = 48,XY). Differences were observed in the NOR number between the species. NORs were identified on three autosome pairs in the northern white-breasted hedgehog and on only two pairs in the African pygmy hedgehog. Chromosome analysis by C-banding/DAPI showed large segments of heterochromatin rich in A-T pairs on three autosome pairs in both the northern white-breasted and African pygmy hedgehogs. The heterochromatin segments encompassed large fragments of the longer arm of chromosome pairs 13, 14 and 20. The (TTAGGG)ₙ repeat sequences on the hedgehog chromosomes were only observed in the terminal position of all the chromosomes in both species. Our observations provide new information on the level of diversity within the Erinaceidae family.

This study aims to find out the various bacteria that cause mastitis through the 16S ribosomal ribonucleic acid (16S rRNA) gene. Materials and Methods: A total of 150 mastitis somatic cell samples were tested with bacterial nested polymerase chain reaction (PCR) universal primers, targeting the 16S rRNA gene. The primers had both Gram-positive and Gram-negative bacterial specificities. Inflammatory cytokine interleukin (IL-10), IL-4, and interferon-gamma (IFNγ) expression genes were measured and compared in mastitis-free and mastitis-affected animals. Results: Based on the PCR, 70 (46.7%) samples showed positive results. The expression of the IL-10 gene was significantly higher (p &lt; 0.001) in mastitis-affected cows than noninfected animals. Compared to cows diagnosed with clinical mastitis, the IL-4 and IFNγ genes were expressed more strongly in healthy cows (p &lt; 0.0001). Conclusion: Mastitis has been linked to both Gram-positive and Gram-negative bacteria. These genes are strong predictors of mastitis in the states analyzed, as evidenced by the differential expression in mastitis and healthy conditions of the IL-4, IL-10, and IFNγ genes.
