2017
Daghigh Kia, Hossein | Olfati Karaji, Rasteghar
BACKGROUND: Semen cryopreservation process with increased reactive oxygen species levels (ROS) plays a decisive role on the sperm cellular organelles (cell membrane, mitochondria and DNA). OBJECTIVES: The aim of this study was to determine the effects of adding different levels of superoxide dismutase (SOD) and reduced glutathione (GSH) antioxidants before freezing on motility, total progressive motility, viability and positive response to hypo-osmotic swelling test (HOST) parameters after Bull semen was frozen. METHODS: In this study, four bulls aged 4-5 years were used and 20 times the semen collection (5 ejaculates per bull) was performed. SOD (100 and 150 IU/ml) and GSH (5 and 7.5 mM) antioxidants were added to Tris - egg yolk extender. Diluted semen samples with or without additives were manually filled with straw and the free end of the straws were sealed by sealing machines; then, semen were frozen by semi-automatic freezing machine, using liquid nitrogen. After thawing the samples, the total and progressive motility, sperm viability and positive response to the HOST parameters were determined at zero and 2 h of incubation. RESULTS: Adding SOD and GSH antioxidant levels did not reveal significant differences between treatments in any of the semen parameters in the initial hour of incubation after thawing. Two hours incubation of post-thawed semen samples containing SOD (100 IU/ml) significantly improved total motility, viability and membrane integrity of the sperm (p<0.05). Also, in samples containing SOD (150 IU/ml), sperm membrane integrity was maintained (p<0.05). In the semen samples with antioxidant GSH (5 mM), total motility and membrane integrity of the sperm are considerably improved after thawing (p<0.05). CONCLUSIONS: This study showed that using SOD and GSH antioxidants as freezing semen extender additives can improve post-thawed bull semen quality.

Canine brachycephalic obstructive airway syndrome (BOAS) is a conformation-related respiratory disorder of dog breeds having congenitally flattened facial and skull anatomy. The aim of the study was to determine oxidative stress parameters, the lipid peroxidation product malondialdehyde, and antioxidant enzymes superoxide dismutase (SOD) and glutathione peroxidase in BOAS patients before and after surgical treatment and in healthy brachycephalic dogs. Nine healthy brachycephalic dogs that had not undergone surgery and 39 BOAS patients were included in the study. The BOAS patients were classified as grade 1 (5/34), grade 2 (16/34), and grade 3 (13/34) based on the decrease in the radius of the airway in the larynx. In BOAS patients, oxidative stress parameters were determined before and two weeks after surgery, while in control dogs, blood samples were collected only on inclusion to the study. All BOAS patients showed various degrees of improvement in clinical signs after surgery. Significantly lower (P < 0.05) SOD activity was found in grade 2 and 3 BOAS patients than in grade 1 patients. Two weeks after surgery, a significant (P < 0.05) increase in SOD activity in grade 2 and 3 patients was observed. Antioxidant enzyme SOD may play an important role in BOAS and can be used as a biomarker of antioxidant status assessment in BOAS patients.

The aim of the study was to demonstrate a link between uncomplicated Babesia canis infection in dogs and blood concentrations of zinc and copper and erythrocytic antioxidant defence – activities of glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT). The study was based on 15 naturally occurring cases of canine babesiosis with anorexia, pyrexia, depression, pale mucous membrane, splenomegaly and dark red urine. Microscopic examination of Giemsa-stained peripheral blood smears and the results of PCR confirmed B. canis infection. Seven apparently healthy dogs brought in for either a check-up or vaccination were used for comparison. The levels of the erythrocytic antioxidant enzymes - SOD and CAT - were significantly higher in the infected dogs than in cytologically negative dogs. The levels of blood micronutrients were significantly lower in the infected dogs (0.478 μg of zinc per mL vs 1.241 μg/mL and 0.722 μg of copper per mL vs 1.392 μg/mL). Oxidative stress can be posited as one of the mechanisms leading to anaemia in dogs with babesiosis, and therefore antioxidant biomarker and copper and zinc concentrations could be used as indicators of disease severity and prognostic markers.

The aim of the study was to investigate the mitigative effects of bisoprolol (BIS) in cadmium-induced myocardial toxicity on oxidative stress and its inhibitive effect on nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB) signalling in rats. Male albino Wistar rats were assigned to control, Cd, BIS 2 (2 mg/kg b.w.) and BIS 8 (8 mg/kg b.w.) groups with nine rats in each. Over four weeks, the control group was administered 1% gum acacia, all other groups received 3mg/kg b.w. CdCl₂ dissolved in distilled water, and the BIS groups were additionally given bisoprolol in gum acacia. Blood samples were collected for biochemical estimations. Blood pressure and serum biomarker (lactate dehydrogenase, aspirate transaminase, alanine transferase and creatine kinase-MB, enzyme (superoxide dismutase, lipid hydroxy peroxidase, catalase and malondialdehyde), and tumour necrosis factor alpha (TNF-α) concentrations were measured. Western blot analysis was conducted for NF-κB and glutathione S-transferase (GST). After sacrificing the rats, cardiac tissue samples were examined histopathologically. Our findings pointed to a significant decrease (P < 0.05) in the studied serum biomarkers and levels of the relevant enzymes in the BIS 8 group compared to the Cd group. A significant decrease (P < 0.05) in NF-kB p65 expression and TNF-α levels was noted in the BIS 8 group relative to the BIS 2 and Cd groups, indicating a reduction at a higher dose. In microscopy, histopathological changes in the cardiac muscles of the BIS 8 group were evident compared to those of the Cd group. BIS seemed to have protective effects against cardiac injury induced by cadmium and could be considered a novel therapeutic drug and prognostic biomarker in the pathology of the many cardiovascular diseases caused by heavy metal intake.

Dairy cows may infrequently give milk tinged with blood after calving, which is a condition termed haemolactia. Economic losses for dairy farmers are caused by cases of haemolactia because of the condemnation of such milk, potential contamination of good bulk tank milk with haemolactic milk, and need for veterinarian intervention. This study was performed to elucidate the oxidative status of dairy cows with haemolactia during the peripartum period. Plasma glutathione peroxidase, malondialdehyde (MDA) and superoxide dismutase concentrations along with serum vitamin A, C and E concentrations were determined as indices of oxidative stress. The sampled dairy cows comprised two haemolactic (n = 11 and n = 6) and two non-haemolactic (n = 11 and n = 6) groups. On the first day when haemolactia was identified in colostrum (at mean 2.1 days after parturition), a significantly increased concentration of plasma MDA was noted in the haemolactic group. During the prepartum period, low levels of serum vitamin E were continuously observed from prepartum week 4 to the parturition day but only in the haemolactic group. These results demonstrate that continuous low levels of serum vitamin E in the prepartum period may play a pivotal role as a requisite factor in the onset of haemolactia after calving.

The aim of the study was to describe the process of neuron death in the cerebral cortex caused by embryonic carbofuran exposure. 81 mouse foetuses from 27 breeding mice were used in the study. Carbofuran was administered by gavage from the 6ᵗʰ to the 15ᵗʰ day of gestation to two groups: one at 0.0208 and the other at 0.0417 mg/kg b.w. On the 17ᵗʰ day, the mice were sacrificed and the foetuses were taken to measure the ROS (malondialdehyde/MDA and superoxide dismutase/SOD) activity in brain tissue, the number of apoptotic embryonic cerebral cortex neurons using a TUNEL assay, and necrotic cells using HE staining. Examination of p53 and caspase 3 expression was done by immunohistochemistry. Data were analysed using analysis of variance (ANOVA) and Duncan’s test. Increased activity of cerebral ROS characterised by significant elevation of the MDA level (P < 0.05), decreased SOD (P < 0.01), increased p53 and caspase 3 expression, and cerebral cortical neuron death either by necrosis or apoptosis (P < 0.05) were found. At the low dose carbofuran increased expression of p53, caspase 3, and apoptosis. At the high dose it increased levels of MDA and necrosis. Increased expression of p53 and caspase 3 and apoptosis indicated that carbofuran may cause apoptosis through the intrinsic pathway. The increased apoptosis grants an opportunity to prevent and treat the effect of ROS due to gestational carbofuran exposure.

2016
Kurhaluk, Natalia | Tkachenko, Halyna
Introduction: The main goal of the study was to investigate the effect of KATP channel modulators on development of oxidative stress in the heart of rats showing different resistance to hypoxia.Material and Methods: The study has been performed on rats showing high- (HR) or low-resistance (LR) to hypoxia under modulators of ATP-sensitive potassium (KATP) channel opener pinacidil (0.06 mg/kg) and blocker glibenclamide (1 mg/kg) upon cobalt (Co) treatment (30 mg of cobalt chloride/kg b.w., 3 h). Changes in the oxidative stress parameters of the heart tissue, such as lipid peroxidation (LPO), level of oxidatively modified protein (OMP), and antioxidant defence system (superoxide dismutase - SOD, catalase -CAT, glutathione peroxidase - GPx, glutathione reductase - GR) as well as total antioxidant activity (TAA) were analysed. Results: Co treatment caused a significant decrease in SOD and CAT activity in the heart of LR rats and GPx activity in HR rats. It also led to a decrease in OMP level in the heart of rats with HR in comparison with controls. Conclusion: The obtained results suggest that individual resistance to hypoxia plays a crucial role in Co actions and provides evidence that the effects of KATP channel opener pinacidil in the heart are mediated through different pathways of the antioxidative system, depending on the individual resistance to hypoxia. Pinacidil exerts a protective effect on the heart tissue by preventing the LPO decrease and significantly reducing OMP levels, as well as increasing TTA in rats with LR.

This study was designed to evaluate the antioxidative status of serum by measuring its total antioxidant capacity, as well as the antioxidant enzyme activity (superoxide dismutase, catalase, and glutathione reductase), in dogs with various stages of degenerative mitral valve disease (DMVD) compared to healthy controls. In total, 71 client-owned dogs in different stages of DMVD, which included healthy controls, took part in the study. Following an anamnesis, clinical examination, standard transthoracic echocardiograpic examination, chest X-ray, complete blood (cell) count, and serum biochemistry, dogs were divided into 2 study groups. Blood was drawn from each dog once at the time of presentation and selected antioxidant parameters were measured using commercially available assay kits. The activity of superoxide dismutase gradually decreased in the more advanced stages of DMVD, while the activity of catalase was significantly higher in the group of dogs with asymptomatic DMVD compared to healthy controls and dogs with symptomatic DMVD. No significant changes were noted in total antioxidant capacity and the activity of glutathione reductase. Results suggested that DMVD has a significant impact on the activity of superoxide dismutase and catalase in the serum of the tested dogs. Knowledge of changes in the activity of antioxidative enzymes may warrant further studies, possibly to evaluate the potential role of compounds with antioxidative properties in the clinical outcome of dogs with DMVD.
