Microvascular circulation of the ascending colon in horses.

Microvascular circulation of the ascending colon in healthy horses was studied using microangiography, light microscopy, and scanning electron microscopy. The pelvic flexure with 30 cm of ventral and dorsal colon attached was removed from 14 adult horses immediately after horses were euthanatized. The lumen was flushed with warm water, and this section of the ascending colon was placed in a 37-C bath of isotonic NaCl. In sections from 8 horses, colic vessels were perfused with a radio-opaque medium for microangiography. After angiographic evaluation, tissue sections were prepared for light microscopic observation, using standard histologic methods. In sections from 6 horses, injection replicas were made by perfusing the vessels with 2 types of plastics. The results of microangiography, light microscopy, and scanning electron microscopy of vascular replicas were correlated, providing acomprehensive documentation of the microvasculature of the ascending colon at the pelvic flexure. Arteries branched from mesenteric colic vessels approximately every 2 cm toward the colonic tissue. Immediately after branching, arterial vessels formed an anastomotic plexus, the colonic rete. However, each branch from the colic vessel eventually continued into the colonic tissue. A second set of vessels originated from the colonic rete and supplied the mesenteric lymph nodes. Arterial vessels penetrated the tunica muscularis into the sub-mucosa 3 to 4 cm toward the antimesenteric border forming a submucosal vascular network. From the submucosal arterioles, branching took place at right angles to supply the mucosal capillaries. Capillaries surrounded the colonic glands and anastomosed at the luminal surface, forming a superficial luminal honeycomb-appearing vascular plexus. Venules, sparsely distributed, drained the superficial plexus. Arterial venous anastomoses were not observed within the mucosa.

In this study, to understand the pathogenesis of new rabbit hemorrhagic disease virus (RHDVa) serotype, we carried out to administrate RHDVa to rabbits, and to examine sequential electron microscopic changes and relationship between pathogenesis and apoptosis. TUNEL-positive cells began to be observed from 24 hours after inoculation (HAI) and the number of positive cells was slightly increased with the course of time. Whereas marked increase of positive cells was seen in the liver from the rabbits died acutely. Typical viral particles with cup-like projections and a diameter of 30~40 nm were detected in homogenized liver samples and tissues at 36 and 48, and 48 HAI, respectively. Ultrastructurally, glycogen deposition was observed from the first stage of hepatocellular degeneration by RHDVa infection and then, swelling and disruption of cristae of mitochondria by viral particles, swelling of smooth endoplasmic reticulum, vacuoles and vesicles were detected. Condensation, margination and fragmentation of chromatin were observed in degenerative hepatocytes at 36 and 48 HAI, indicating apoptotic bodies. These data offer that hepatocytic apoptosis by RHDV infection could be closely related with mitochondrial impairment in the hepatocytes.

Scanning electron microscopy on Ixodes signatus nymphs with particular reference to major physiological sensory organs

Ixodes signatus nymphs were subjected to the scanning electron microscopy for the observation of the major physiological sense organs including Haller's organ. Funger prints and variable sensory setae were found on the body surface, and especially well developed setae were identified on the article IV and on the anal lobes. A central button-like organelle and numerous small holes were found in the spiracular plate. The Haller's organ was identified at the distal point of the tarsus I and regarded as the major sensory organ, chemoreceptor. Two types of sensory setae were observed, the shorter ones in front of the organ and the longer ones in distal front as well as behind the organ. The fully opened organ was ellipsoidal and a total of seven basiconic sensory pegs rooted deeply and shaped like well arranged hooks were found inside the organ

Scanning electron microscope (model; SEM ISI-DS-130) was used to observe the fine surface structures of the predominant cattle tick Haemaphysalis longicornis Neumann 1901 (Acarina; Ixodidae) in Korea R. and some SEM microphotographs were presented in this pictorial monograph. A total of 18 photographs was selected from the numerous SEM photographs of the female and male specimens of H. longicornis for the observations of the fine surface structure and the comparison between the female and male structures
