Physiologic and antinociceptive effects following intramuscular administration of xylazine hydrochloride in combination with tiletamine-zolazepam in llamas

Objective-To evaluate antinociceptive and selected effects associated with IM administration of xylazine hydrochloride in combination with tiletamine-zolazepam in llamas. Animals-8 adult male llamas. Procedures-Each llama received tiletamine-zolazepam (2 mg/kg) combined with either xylazine (0.1, 0.2, or 0.4 mg/kg) or saline (0.9% NaCl) solution IM (treatments designated as TZ-Xy0.1, TZ-Xy0.2, TZ-Xy0.4, and TZ-Sal, respectively) at 1-week intervals. Selected cardiorespiratory variables were assessed during lateral recumbency and anesthesia, and recovery characteristics were recorded. Duration of antinociception was evaluated by clamping a claw every 5 minutes. Results-Interval between treatment administration and lateral recumbency for TZ-Xy0.4 was shorter than that for TZ-Xy0.1 or TZ-Sal. Mean ± SEM duration of antinociception was longer for TZ-Xy0.4 (51.3 +/- 7. 0 minutes), compared with findings for TZ-Xy0.2 (31.9 +/- 6.0 minutes), TZ-Xy0.1 (8.1 +/- 4.0 minutes), and TZ-Sal (0.6 +/- 0.6 minutes). Interval between treatment administration and standing was longer for TZ-Xy0.4 (112 +/- 9 minutes) than it was for TZ-Xy0.2 (77 +/- 9 minutes) or TZ-Sal (68 +/- 9 minutes). Mean heart and respiratory rates during the first 30 minutes for TZ-Sal exceeded values for the other treatments. Administration of TZ-Xy0.2 and TZ-Xy0.4 resulted in Pao2 < 60 mm Hg at 5 minutes after llamas attained lateral recumbency, and values differed from TZ-Sal findings at 5, 10, and 15 minutes; Paco2 was greater for TZ-Xy0.2 and TZ-Xy0.4 than for TZ-Sal at 5, 10, 15, and 20 minutes. Conclusions and Clinical Relevance—Xylazine (0.2 and 0.4 mg/kg) increased the duration of antinociception in llamas anesthetized with tiletamine-zolazepam.

Objective-To evaluate the effect of underwater treadmill exercise on static postural sway in horses with experimentally induced carpal joint osteoarthritis under various stance conditions. Animals-16 horses. Procedures-On day 0, osteoarthritis was induced arthroscopically in 1 randomly selected middle carpal joint of each horse. Beginning on day 15, horses were assigned to either underwater or overground (without water) treadmill exercise at the same speed, frequency, and duration. Two serial force platforms were used to collect postural sway data from each horse on study days −7, 14, 42, and 70. Horses were made to stand stationary on the force platforms under 3 stance conditions: normal square stance, base-narrow placement of the thoracic limbs, and removal of visual cues (blindfolded) during a normal square stance. The mean of 3 consecutive, 10-second trials in each condition was calculated and used for analysis. Results-Displacement of the center of pressure differed significantly depending on the stance condition. Among horses exercised on the underwater treadmill, postural stability in both the base-narrow and blindfolded stance conditions improved, compared with findings for horses exercised on the overground treadmill. Horses exercised on the overground treadmill were only successful at maintaining a stable center of pressure during the normal square stance position. Conclusions and Clinical Relevance-Variations in stance position had profound effects on the mechanics of standing balance in horses with experimentally induced carpal joint osteoarthritis. Underwater treadmill exercise significantly improved the horses’ postural stability, which is fundamental in providing evidence-based support for equine aquatic exercise.

Several lyssavirus species occur in Africa (Rabies virus, Lagos bat virus, Mokola virus, Duvenhage virus, Shimoni bat virus and Ikoma lyssavirus), displaying a high sequence diversity between isolates belonging to the same species. There is limited information about comparative pathogenesis of these African lyssaviruses and this precludes authoritative opinion on the potential public and veterinary health impact. In this study, an analysis of representative African lyssaviruses attempted to correlate viral genomic sequence similarities and differences with the corresponding pathogenic profiles observed in mice. The study demonstrated that the virus isolates evaluated could be lethal to mice when introduced intramuscularly and that different isolates of the same lyssavirus species differ in their virulence. Using real-time polymerase chain reaction (PCR), viral RNA was detected in brain tissue, but no viral RNA was detected in the salivary glands or blood of mice that succumbed to infection. Comparison of known pathogenic domains indicated that pathogenicity is likely to be dependent on multiple domains. Cumulatively, our results re-emphasised the realisation that the pathogenicity of a lyssavirus species cannot be deduced based on studies of only a single isolate of the species or a single pathogenic domain.

The metacercarial (larval) stages of diplostomid digeneans are known to inhabit freshwater fish, causing tissue damage in the process. Due to their widespread diversity, little is known about their life cycle. The classification of these parasitic stages to the species level using only the morphology is very challenging due to the lack of genitalia; they are regarded to be the most important structures in the identification of these organisms. In this study, additional morphological information through light and scanning electron microscopy is given for two different diplostomids found in the cranial cavity of Clarias gariepinus and the vitreous chambers of Tilapia sparrmanii and Pseudocrenilabrus philander. The diplostomid metacercaria inhabiting the cranial cavity of Clarias gariepinus was morphologically identified as Diplostomulum (Tylodelphys) mashonenseand an unknown metacercaria of the genus Diplostomumwas found in the vitreous chambers of Pseudocrenilabrus philander and Tilapia sparrmanii. Both parasitic species’ 28S recombinant deoxyribonucleic acid genomic regions were successfully amplified using Dig 125/1500R primer pairs. The assay yielded a product of approximately 1300 base pairs as seen on the gel images. There were 14 nucleotide differences over the entire analysed sequences resulting in a 1.1% (14/1273) nucleotide difference. In line with the morphological characteristics of these parasites, there seemed to be a slight difference in their genetic makeup. The application of molecular techniques on digenetic trematodes seems very promising and may yield great potential in future descriptions of morphologically similar parasitic species.

Changes in serum gamma globulin levels, numbers of replete female ticks and engorged tick mass were used as parameters to monitor the acquired immune response (antibody mediated immune response) elicited by Rhipicephalus appendiculatus adult tick infestations. Three consecutive Rhipicephalus appendiculatus adult tick infestations were applied to South African Indigenous goats (Nguni), Saanen goats and cross-bred goats (Saanen goats crossed with South African Indigenous goats [Nguni]) under laboratory conditions. During the three consecutive Rhipicephalus appendiculatus adult tick infestations the serum gamma globulin levels increased in all three breeds, whilst the mean replete female tick numbers and engorged tick mass decreased. Even though all three goat breeds exhibited an acquired immune response, the South African Indigenous goats (Nguni) response was significantly higher than that of the Saanen and cross-bred goats. However, the acquired immune response elicited by Saanen goats was significantly lower when compared with cross-bred goats.

A cross-sectional study was conducted to determine the prevalence of sub-clinical and clinical mastitis and the associated factors in cows from selected smallholder dairy farms in Zimbabwe. Physical examinations were conducted on all lactating cows for evidence of signs of clinical mastitis. Composite milk samples were collected from all lactating cows for bacterial culture and somatic cell counting. Cows were categorised as clinical if they exhibited clinical features of mastitis, or sub-clinical if no apparent signs were present but they had a positive bacterial isolation and a somatic cell count of at least 300 x 103 cells/mL. Farm-level factors were obtained through a structured questionnaire. The association of mastitis and animal- and herd-level factors were analysed using logistic regression. A total of 584 animals from 73 farms were tested. Overall, 21.1%(123/584) had mastitis, 16.3%(95/584) had sub-clinical mastitis and 4.8% (28/584) had clinical mastitis. Herd-level prevalence was 49.3%. Coagulase-negative staphylococci (27.6%), Escherichia coli (25.2%), Staphylococcus aureus(16.3%), Klebsiella spp. (15.5%) and Streptococcus spp. (1.6%) were the most common isolates. In individual cows, pure dairy herds (OR = 6.3) and dairy crosses (OR = 3.1) were more likely to have mastitis compared to Mashona cows. Farms that used pre-milking teat dipping were associated with reduced mastitis prevalence. Further research is needed on the prevalence of mastitis and a comparison of data for both smallholder and commercial dairy farms in all regions of Zimbabwe should be undertaken.

The distribution of interstitial cells of Cajal (ICC), the probable pacemakers in gastrointestinal motility, was investigated using an antigenic marker of gastric ICC known as C-Kit. Antiserum raised against the general neuronal marker protein gene peptide 9.5 (PGP) as well as the nitrergic neuronal marker neuronal nitric oxide synthase (nNOS) were used to investigate the distribution of gastric nerves. Polyclonal goat anti-human C-Kit was reliable in labelling ICC in the stomach. Two classes of ICC were identified according to their distribution: ICC-MY distributed around the periphery of myenteric ganglia and ICC-IM in the circular and longitudinal muscle layers. The neuronal marker PGP was reliably consistent in revealing the density and distribution of the enteric nervous system. Density of nerve fibres was higher in circular smooth muscle than in longitudinal smooth muscle. From nNOS immunohistochemistry, it is evident that inhibitory (nitrergic) nerves constitute a substantial fraction of the enteric nervous system.

This study was conducted to study the UV protective effect against Haemonchus contortus infection in goats. Sixteen male goats were divided into 5 groups, control infected, control uninfected and UV 30minutes; UV 60minutes and UV booster 60minutes exposure. The UV groups were exposed to UV irradiation at wave length 254nm for 30 and 60 minutes. The UV booster 60min was administrated 2 doses of exposed larvae with an interval of one month. All groups except the control negative one were challenged for 42 days from the beginning. In UV booster 60min had reduction in egg count per gram feces and worm burden (93% & 34 % respectively). These parameters were similar in control infected, UV 30min and UV 60min groups. Increases in levels of antibodies were found in goats of UV booster 60min group the other groups. Finally, 2 doses of UV 60min exposure could protect goats from H. contortus.

Assessment of the neutralizing antibody response to the attenuated strain of peste des petits ruminants Nigerian strain (75/1) in 5 pregnant ewes revealed a geometric mean titre of 194, four weeks post Vaccination. Such an appreciably high titre was found to be dropped to 73.5, throughout 48 hours post parturn. Suckling kids born to these dams passively acquired a titre of 84.5 throughout their first month of life that dropped to 48.5 at the age of 2 months. At their 5th month of age their immune titre was found to be only 4. Data generated from this study might be of value in launching vaccination campaigns against peste des petits ruminants disease.

Micronization of chemical compounds is a new promising field as it reduces the size of the particles thus it increases their penetration power. Many theories prove that micronized particles are more effective even in very low concentration as compared to its normal size. Paracetamol is a commonly used effective analgesic and antipyretic agent for relief of mild and moderate pain. However, deliberate overdose or accidental overdose can cause hepatotoxicity. In the present study, we have evaluated and compared between the hepatoprotective and antioxidant effects of vitamin C and micronized vitamin C against paracetamol induced hepatotoxicity in rats. Activities of liver enzymatic markers (Alanine amino transferase "ALT", Aspartate amino transferase "AST", Alkaline Phosphatase "ALP") and total protein "TP" concentration were estimated in serum. Lipid peroxidation "MDA" and antioxidant status (reduced glutathione "GSH " concentration, glutathione reductase "GR", catalse and super oxide dismutase "SOD" activities) were measured in tissue homogenates. Paracetamol administration (600 mg/ Kg B.wt.) significantly increased the liver enzymatic markers and decreased the total protein level. It also increased hepatic lipid peroxidation “MDA” and the activities of both catalase and SOD while it decreased GSH content and glutathione reductase (GR) activity. Treatment of vitamin C and micronized vtamin C restore the measured parameters nearly to their normal levels. Finally, micronized vitamin C has a more potent effect than ordinary vitamin C.
