Adipokines as metabolic modulators of ovarian functions in livestock: A mini-review

Adipose tissue is the principal fat storing tissue which secretes various molecules known as adipokines. The major adipokines secreted from adipose tissue are leptin, adiponectin, visfatin, resistin, chemerin and apelin. Adipokines are regarded as the and ldquo;marker of body metabolic status'' which maintains the body energy homeostasis. An adequate energy level is essential for the onset of puberty and ovarian functions. Adipokines act as energy sensor and signal the body energy level to hypothalamic neurons to regulate many physiological activities including ovarian functions such as onset of puberty, estrus behavior, follicular development and ovulation followed by corpus luteum (CL) formation and function in livestock. However, adipose tissue dysfunctions limit adipokines secretion leading to an imbalance in body energy level which ultimately affects the reproduction in livestock. Objective: The study was conducted to evaluate the influence of season on cauda epididymal spermatozoa isolated from bucks. Materials and methods: Testes of 30 mature bucks were collected from local slaughter house, and were processed for the retrieval of cauda epididymal spermatozoa for evaluation. Testes were collected in three seasons (winter, summer and rainy), and each season was having 10 pairs of testicles. Recovered spermatozoa from the cauda epididymis were processed immediately for evaluation of semen attributes (Spermatozoa motility, viability, plasma membrane integrity, acrosomal status and DNA integrity). Results: Physiological effect of season was observed on progressive motility, percent of live spermatozoa, spermatozoal membrane integrity (HOST), acrosomal integrity, capacitation status and DNA integrity. Progressive motility, percent live spermatozoa, HOST positive spermatozoa, were found significantly (P<0.05) high in summer season, whereas, significantly (P<0.05) lower comet positive spermatozoa were found in summer season as compared to rainy and winter. Compromised acrosomal status was seen in winter and rainy seasons as compared to summer. Conclusion: Compromised acrosome along with plasma membrane and higher percentage of spermatozoa with damaged DNA in cauda spermatozoa were observed during winter and rainy seasons as compared to summer season. Summer season was found to be the most suitable season for collection of cauda epididymal spermatozoa and can effectively be used for assisted reproduction with further investigations of associated mechanisms. Objective: This study was aimed to evaluate the effects of daily intake of vitamin C or selenium against deleterious effects of nicotine toxicity on pregnant albino rats. Materials and methods: Forty albino pregnant rats were equally distributed into four groups. Group A was considered as control. Group B was administered with nicotine dosed at 1 mg/kg body weight (bwt) daily for 7 weeks (wks) from 1st day of gestation until the postnatal 4 wks. Group C was treated with nicotine and vitamin C dosed at 1 mg/kg bwt orally for 7 wks, group D was treated with nicotine and sodium selenite dosed at 1 ug/100 g bwt concurrently for 7 wks. The levels of catalase (CAT), superoxide dismutase (SOD), protein carbonyl (PC) and thiobarbituric acid reactive substances (TBARS), were estimated in homogenates of the lung, kidney and liver. Histopathological studies using hematoxylin and eosin as well as immunohistochemical studies using p53 antibody were also done. Results: Nicotine significantly elevated the levels of TBARS and PC as compared to control rats. Groups C and D showed decrease in these levels significantly. CAT and SOD activities of group B were decreased significantly. Significant elevation of CAT and SOD activities was detected in both groups C and D. Vitamin C elevated the antioxidant enzymes activities to normal levels, however selenium administration improved these levels but still lower than those of group A. Expression of p53 was decreased in group B as compared to group A. Vitamin C completely reversed the expression of p53 as group A. However, group D did not showed any significant changes in expressions as compared to group B. Conclusion: It is concluded that vitamin C intake was useful than selenium in prevention against nicotine-induced oxidative stress including p53 expression in the lung, kidney and liver of pregnant rats. Objective: Lighting requirements of birds have to be considered for choosing the optimal light intensity. Therefore, the experiment was carried out to study the effects of the different light intensities on performance, welfare and behavior of turkey poults. Materials and Methods: A total of 81 turkey poults aging 3 weeks were randomly selected and divided into three equal groups. The birds were subjected for 3 different light intensities (5, 25 and 50 lux) with 12 h photoperiod in 3 separate rooms. The growth performance parameters of the poults were measured; the parameters were body weight (BW), average feed intake (FI), average body weight gain (ABWG), relative growth rate (RGR), and feed conversion ratio (FCR). Besides, blood parameters and behavioral characters were observed. Results: Our results revealed that poults reared in low intensity (5 lux) had better performance (body weight gain, relative growth rate and feed intake). Also, final body weight was significantly higher in 5 lux. The cortisol level was lower in 5 lux than other 25 and 50 lux. Behavior of poults is affected by light intensities in our study, where feather preening, feather pecking and aggressive behaviors were significantly higher in birds housed in the highest intensity (50 lux). In the same way, drinking time was significantly higher in 25 lux. The poults tended to lay and rest on perches more under the lowest intensity (5 lux). Conclusion: It was concluded that, it is better to use moderate light intensity to improve previous parameters with avoiding abnormal behavior. [J Adv Vet Anim Res 2016; 3(1.000): 18-23]

Objective: To determine the prevalence of Schistosoma infection in cattle in Maiduguri Metropolis (MMC) and Jere Local Government Areas (LGAs) of Borno State, Nigeria.Materials and Methods: Blood samples (n=200) were collected from cattle consisting of one hundred (100) each from five (5) ward levels each of MMC and Jere LGAs. DNA samples were extracted from the serum samples, analysed and quantified using a Nano-drop machine. The extracted DNA were subjected to polymerase chain reaction (PCR).Results: The overall prevalence of Schistosoma infection was 2% (n=200). Jere LGA had 3% (n=100) while MMC had 1% (n=100). There was no statistical significant association in prevalence rate in the two LGAs studied (P=0.621) (P>0.05). At the ward levels, Custom Area in Jere LGA had 15%, Jiddari ward in MMC had 5%, and the remaining ward levels had no cases. Of the 103 female and 97 male cattle screened, the prevalence in female was 1(0.97%) and 3(3.09%) in the male. Of the 177 serum samples from above 1year (adult) examined, 4 (2.26%) were positive and none in the young. There was no statistical significant association in prevalence rate among ward levels, sex groups and age groups in the study areas (P=0.621) (P>0.05), (P=0.356) (P>0.05) and (P=1.000) (P>0.05) respectively. Of the eight (8) breeds screened, Kuri had 2.7%, Sokoto Gudali (1.82%), Abore (2%), Red Bororo (2.63%), White Fulani, Porland, Mbala and Wafara recorded no cases. The difference in prevalence rates among the breeds based on the trend of occurrence of Schistosoma infection were not significantly associated statistically (P=1.000) (P>0.05). Conclusion: There is a prevalence of Schistosoma infection in cattle in the two LGAs of Borno state. It is recommended that a system be established to maintain preventive and control programs. Objectives: This study was designed to investigate the Sero-prevalence of Schistosoma species in cattle in Maiduguri Metropolis (MMC) and Jere Local Government Areas (LGAs) of Borno State, Nigeria. Materials and Method: Blood samples (n=200) from cattle were collected using a multistage sampling technique; 100 samples each were collected from MMC and Jere LGAs, respectively. The samples were subjected to screening for Schistosoma antibodies using Enzyme Linked Immunosorbent Assay (ELISA). Age, sex, breed and location of cattle were recorded. Results: The overall prevalence of Schistosoma infection among cattle in MMC and Jere LGAs was 10%. Jere LGA had a prevalence rate of 14% and MMC had 6%. At the ward levels, Custom Area in Jere LGA had the highest number of Schistosoma positive (50%). Out of 103 female and 97 male cattle screened, the prevalence of Schistosoma infection in female and male were 9.71% (n=10/103) and 10.31% (n=10/103). Out of the 177 serum samples from cattle aging >1-year (adult) examined, 16 (9.04%) were positive, while only 4 (17.39%) out of 23 serum samples from cattle aging <1-year (young) were positive. Out of the eight (8) breeds screened, the highest number of cases was recorded in Kuri breed (16.22%). This was followed by Sokoto Gudali (10.9%) breed. The prevalence in other breeds was as follows: Abore- 10%, Red Bororo- 5.26%, and White Fulani- 6.67%. Conclusion: It is concluded that schistosomiasis in cattle was prevalent in MMC and Jere LGAs of Borno State. A regular checking program is suggested to constantly check out whether the prevalence rate is increasing, so that effective control measures can be strenthened. The only way to diagnose hepatic fibrosis in dogs is by histological assessment of a liver biopsy specimen. As this technique is invasive and susceptible to sampling variation, serum biomarkers are used to detect hepatic fibrosis in humans. The objective of this study was to assess the utility of hyaluronic acid (HA), procollagen type III N-terminal peptide (PIIINP), and tissue inhibitor of matrix metalloproteinase-1 (TIMP-1) as serum markers of canine hepatic fibrosis. Serum samples were collected from 47 dogs with histologically confirmed hepatobiliary disease and 24 healthy dogs in order to measure concentrations of HA, PIIINP, and TIMP-1. Hepatic fibrosis was staged using a 5-point scoring scheme. There was no correlation between serum concentrations of HA or PIIINP and the severity of hepatic fibrosis. There was a negative correlation between serum concentration of TIMP-1 and the severity of hepatic fibrosis (r s = −0.33; P = 0.036). It was not possible to use serum concentrations of HA, PIIINP, or TIMP-1 to discriminate between dogs with absent-to-moderate hepatic fibrosis and those with marked-to-very-marked fibrosis. The results of this study do not support the utility of measuring serum concentrations of HA, PIIINP, or TIMP-1 for diagnosing canine hepatic fibrosis. Further studies are needed to support this finding.

OBJECTIVE To determine population pharmacokinetics of enrofloxacin in purple sea stars (Pisaster ochraceus) administered an intracoelomic injection of enrofloxacin (5 mg/kg) or immersed in an enrofloxacin solution (5 mg/L) for 6 hours. ANIMALS 28 sea stars of undetermined age and sex. PROCEDURES The study had 2 phases. Twelve sea stars received an intracoelomic injection of enrofloxacin (5 mg/kg) or were immersed in an enrofloxacin solution (5 mg/L) for 6 hours during the injection and immersion phases, respectively. Two untreated sea stars were housed with the treated animals following enrofloxacin administration during both phases. Water vascular system fluid samples were collected from 4 sea stars and all controls at predetermined times during and after enrofloxacin administration. The enrofloxacin concentration in those samples was determined by high-performance liquid chromatography. For each phase, noncompartmental analysis of naïve averaged pooled samples was used to obtain initial parameter estimates; then, population pharmacokinetic analysis was performed that accounted for the sparse sampling technique used. RESULTS Injection phase data were best fit with a 2-compartment model; elimination half-life, peak concentration, area under the curve, and volume of distribution were 42.8 hours, 18.9 μg/mL, 353.8 μg•h/mL, and 0.25 L/kg, respectively. Immersion phase data were best fit with a 1-compartment model; elimination half-life, peak concentration, and area under the curve were 56 hours, 36.3 μg•h/mL, and 0.39 μg/mL, respectively. CONCLUSIONS AND CLINICAL RELEVANCE Results suggested that the described enrofloxacin administration resulted in water vascular system fluid drug concentrations expected to exceed the minimum inhibitory concentration for many bacterial pathogens.

OBJECTIVE To evaluate optimal isolation of endothelial colony-forming cells (ECFCs) from peripheral blood of horses. SAMPLE Jugular and cephalic venous blood samples from 17 adult horses. PROCEDURES Each blood sample was divided; isolation was performed with whole blood adherence (WBA) and density gradient centrifugation (DGC). Isolated cells were characterized by uptake of 1,1’-dioctadecyl-3,3,3’,3’-tetramethylindocarbocyanine perchlorate–labeled acetylated low-density lipoprotein (DiI-Ac-LDL), vascular tubule formation, and expression of endothelial (CD34, CD105, vascular endothelial growth factor receptor-2, and von Willebrand factor) and hematopoietic (CD14) cell markers by use of indirect immunofluorescence assay (IFA) and flow cytometry. RESULTS Colonies with cobblestone morphology were isolated from 15 of 17 horses. Blood collected from the cephalic vein yielded colonies significantly more often (14/17 horses) than did blood collected from the jugular vein (8/17 horses). Of 14 cephalic blood samples with colonies, 13 were obtained with DGC and 8 with WBA. Of 8 jugular blood samples with colonies, 8 were obtained with DGC and 4 with WBA. Colony frequency (colonies per milliliter of blood) was significantly higher for cephalic blood samples and samples isolated with DGC. Cells formed vascular tubules, had uptake of DiI-Ac-LDL, and expressed endothelial markers by use of IFA and flow cytometry, which confirmed their identity as ECFCs. CONCLUSIONS AND CLINICAL RELEVANCE Maximum yield of ECFCs was obtained for blood samples collected from both the jugular and cephalic veins and use of DGC to isolate cells. Consistent yield of ECFCs from peripheral blood of horses will enable studies to evaluate diagnostic and therapeutic uses.

OBJECTIVE To determine effects of oral administration of metronidazole or doxycycline on olfactory function in explosives detection (ED) dogs. ANIMALS 18 ED dogs. PROCEDURES Metronidazole was administered (25 mg/kg, PO, q 12 h for 10 days); the day prior to drug administration was designated day 0. Odor detection threshold was measured with a standard scent wheel and 3 explosives (ammonium nitrate, trinitrotoluene, and smokeless powder; weight, 1 to 500 mg) on days 0, 5, and 10. Lowest repeatable weight detected was recorded as the detection threshold. There was a 10-day washout period, and doxycycline was administered (5 mg/kg, PO, q 12 h for 10 days) and the testing protocol repeated. Degradation changes in the detection threshold for dogs were assessed. RESULTS Metronidazole administration resulted in degradation of the detection threshold for 2 of 3 explosives (ammonium nitrate and trinitrotoluene). Nine of 18 dogs had a degradation of performance in response to 1 or more explosives (5 dogs had degradation on day 5 or 10 and 4 dogs had degradation on both days 5 and 10). There was no significant degradation during doxycycline administration. CONCLUSIONS AND CLINICAL RELEVANCE Degradation in the ability to detect odors of explosives during metronidazole administration at 25 mg/kg, PO, every 12 hours, indicated a potential risk for use of this drug in ED dogs. Additional studies will be needed to determine whether lower doses would have the same effect. Doxycycline administered at the tested dose appeared to be safe for use in ED dogs.
