Molecular detection of Mycobacterium tuberculosis from bovine milk samples

Mycobacterium tuberculosis and Mycobacterium bovis are the major causes of tuberculosis. These may infect many animal species, and are likely to be the main source of infection in humans. A total of 181 bovine raw milk samples and 123 pre-scapular lymph node biopsy samples were collected and subjected to acid fast staining, fluorescent staining, isolation and identification. Genus specific PCR to identify the Mycobacterium tuberculosis complex (MTBC) organism, and multiplex PCR (mPCR) were done to differentiate M. tuberculosis and M. bovis. Among the milk samples tested, only one sample was culture-positive for M. tuberculosis. Four samples were positive by MTBC-PCR and mPCR; all these four were proved to be M. tuberculosis. It is quite likely that animals can be infected with human-originated M. tuberculosis, which in turn may act as a source of infection in humans, becoming a reverse zoonosis. Hence, control strategies for human tuberculosis caused by M. tuberculosis should necessarily include the control strategies in animals too.

Pasteurella multocida type A is the etiologic agent of fowl cholera, a highly contagious and fatal disease of chickens. The present research work was performed for the isolation, identification and molecular detection of P. multocida Type A from chickens. Liver, heart and spleen of suspected dead chicken (n=35) were collected from Gazipur and Pabna districts in Bangladesh. The targeted bacteria from the samples were isolated, identified and characterized based on their morphology, staining, cultural, biochemical characters, pathogenicity test, histopathological study and Polymerase Chain Reaction (PCR). The P. multocida organism was isolated from 11.42% (n=4/35) samples. The organisms were gram negative, non-spore forming rod, non-motile, occurring singly or pairs in Gram staining, whereas in Leishman's stain, bipolar shaped organisms were observed. All the isolates were found positive for oxidase and catalase tests, produced indole, and fermented glucose, mannitol and sucrose. Necrotic foci in liver and congestion with hemorrhages in heart were found on necropsy. After pathogenicity test, the pathological changes were reconfirmed by histopathology depicting congestion, hemorrhage and lymphocyte infiltration in heart, liver and spleen tissues. In type specific PCR reaction, the organisms were confirmed as P. multocida Type A. In conclusion, P. multocida type A is prevalent among poultry in the studied regions; thus, care must be taken to control of the disease. [J Adv Vet Anim Res 2015; 2(3.000): 338-345]

Duck plague (DP) is the most feared duck disease in the world. For isolation, identification, molecular detection and characterization of DP virus (DPV), a total of 94 samples were collected from commercial farms (n=6) and households (n=13) from Rajshahi (n=37), Netrokona (n=35) and Mymensingh (n=22) districts of Bangladesh. The samples were processed and inoculated into 11-13 days old embryonated duck eggs for virus propagation. Virus was identified using agar gel immunodiffusion test (AGIT) and passive hemagglutination (PHA) test, and was confirmed by polymerase chain reaction (PCR) targeting DNA polymerase and gC genes, followed by sequencing. Pathogenicity tests were performed using duck embryos, ducklings and ducks. Among the 94 samples, 17 isolates were confirmed as DPV by PCR amplification of partial DNA polymerase (446-bp) and gC genes (78-bp), respectively. One of the isolates (Anatid herpes 1 BAU DMH) was sequenced and found to be closely related with a Chinese variant of DPV (GenBank: JQ647509.1). In the last two decades, the chemical and biological contaminants in livestock and livestock products worldwide have been considered as one of the causes that threatens life of both man and animals. In Sudan very little work has been done in this field. This study was carried out to detect Oxytetracycline (OTC) residues in sheep liver and muscle samples in Khartoum State, Sudan. A total of 150 samples comprising of liver (n=75) and muscle (n=75) of sheep were collected from three different localities in Khartoum State during the period from March to September 2013. The OTC residues were detected using high performance liquid chromatography (HPLC) and one plate test (OPT). The results of HPLC showed that the level of OTC residues in liver samples above maximum residue limit (MRL) was 4% (n=3/75) and under MRL was 96%. Regard to muscle samples, the level of OTC residues above MRL was 60.3% (n=35/58), whereas, under MRL was 39.7%. Using OPT, 14.7% (n=11/75) and 6.7% (n=5/75) of liver and muscle samples showed inhibition zone, respectively. It is concluded that OTC residues are detected in sheep liver and muscle sample with different levels in Khartoum state. Further investigation on level of OTC residues in other farm animals and other organs throughout the country is recommended. [J Adv Vet Anim Res 2015; 2(3.000): 321-325]

This study was undertaken for isolation, identification and determination of antibiogram profile of Aeromonas hydrophila in farm raised fresh water prawn (Macrobrachium rosenbergii) on five commercial ghers. Fresh water prawns (n=25) were collected from five ghers located at Satkhira, Bagerhat and Khulna districts of Bangladesh. Brain (n=25), muscle (n=25) and intestine (n=25) samples were collected aseptically from fresh water prawn and inoculated into alkaline peptone (APW) water for enrichment at 37℃ for 8 h. Enriched cultured was streaked into Thiosulfate Citrate Bile Salt Sucrose (TCBS) agar to isolate bacteria. Bacteria were identified by cultural, Gram staining, biochemical properties and polymerase chain reaction (PCR) assay. The antibiogram profiles of bacteria were investigated against 5 commonly used antibiotics (Gentamicin, Cefalexin, Ampicillin, Azithromycin and Ciprofloxacin) by disc diffusion method. Thirteen A. hydrophila isolates were identified and the prevalence of the A. hydrophila in fresh water prawn was 17.33%. All 13 (100%) isolates were sensitive to Ciprofloxacin, Gentamicin, Azithromycin and resistant to Ampicillin and Cefalexin. The results of this study indicate that farm raised fresh water prawn harbor multidrug resistant A. hydrophila which might causes public health problem if enter into human food chain.

Perfect detection of estrus is crucial for good husbandry practice of cow. Estrus synchronization is the alternative strategy to bypass the critical problem of estrus detection. Synchronization program allows fixed timed artificial inseminations (FTAI) to evade the estrus detection. The most recently developed programs include protocols for re-synchronization following first or subsequent inseminations. These re-synchronization protocols may involve selected forms of hormonal intervention during the diestrus and pro-estrus periods following FTAI, or following pregnancy diagnosis by ultrasound from 28 days after FTAI. Almost all programs involve strategically timed treatment of prostaglandin F2α (PGF) and gonadotropin releasing hormone (GnRH). Treatment of an estradiol ester and progesterone supplementation per vaginum may be included in some programs. The basic program is the “Ovsynch” regimen. This mini-review discusses a number of key points related to external hormonal stimulation on ovarian follicular wave to improve pregnancy rate following timed AI.

The purpose of this study was to provide fundamental information on both the common toxicants and poisoning episodes in dogs within Effurun/Warri municipality region of Delta State of Nigeria from 2011 to 2014. The collected data were grouped according to age, sex, breed, year of occurrence, and toxicant type. The study revealed that a total of 76 dogs comprising 12 breeds were poisoned by several toxicants such as pesticides (69%; n=52/76), household products (12%; n=9/76), food and ndash;poisons (9%; n=7/76), and snakebite envenomation (3%; n=2/76). Mixed breeds (29%; n=22/76), Alsatian (25%; n=19/76), Rottweiler (16%; n=12/76) and indigenous breed (7%; n=5/76) ranked the topmost breeds accounted for most poison emergencies with human and ndash;related factors incriminated. This study provides useful information on episodes of poisoning in dogs in the studied region. The knowledge of agents involved can help veterinarians for accurate diagnosis, and pet owners to cautiously protect their pets from potentially poisonous substances. Tetanus in animals is sporadic in nature but difficult to combat even by using antibiotics and antiserum. It is crucial to understand the fundamental mechanisms and signals that control toxin production for advance research and medicinal uses. This review was intended for better understanding the basic patho-physiology of tetanus and neurotoxins (TeNT) among the audience of related field.

In Nigeria, knowledge on the epidemiology of diseases of dogs is limited. A retrospective study of data from clinical records of six veterinary clinics was undertaken to determine the incidence of disease in owned dogs in Delta State, Nigeria from 2012 to 2014. Association between the diagnosed diseases and the studied variables was explored using Chi-Squared test statistics. This study revealed that most of the conditions presented to the veterinary clinics were preventable. Thirty-one (31) clinical conditions were diagnosed from 571 cases recorded, involving nonspecific (21.6%), infectious (70.9%; P=0.001) and non-infectious (7.5%) diseases. The most occurring clinical conditions comprised helminthoses (21.4%), mange (10.5%), parvovirosis (8.4%), babesiosis (7.9%), septicemia (7.2%), gastroenteritis (7.0%), myiasis (7.0%), trauma (6.3%), poisoning (6.0%), ectoparasitism (3.7%), ascites (2.5%), dermatitis (2.3%), aural hematoma (1.2%), and orchitis (1.1%). Disease incidence was highest in Alsatian (40.3%), mixed/cross (33.1%), Rottweiler (7.0%) and toy breeds (4.6%). Details on the least occurring diseases and the association between disease and the studied variables are given. The outcomes demonstrate the prevalence of the clinical conditions diagnosed, inadequate husbandry and veterinary care accorded to owned dogs in the State. Education of dog owners on preventive measures is paramount in alleviating some of these health problems.

The present investigation was carried out to evaluate three potential complete rations made from locally available ingredients through in vitro digestibility (DM, CP, NDF and ADF) and in vitro total volatile fatty acids (TVFA) production. Three iso-nitrogenous complete rations comprising of maize stover (T1), red gram straw (T2) or black gram straw (T3) and concentrate in 60:40 ratio were prepared and the cost/kg diets were 5.2, 5.5 and 5.7 rupees/Kg diet, respectively. Three fistulated buffalo bulls (5 yrs, 350 ± 9.36 kg BW) used as donors of rumen inoculums were fed the same three complete rations (T1, T2 and T3) after proper adaptation. The three complete rations were iso-nitrogenous with CP content ranging from 12.62 to 12.82, while T3 contained higher EE (1.68 %) and TA (9.75 %) and higher NFE (34.38 %) in T1 than in other complete rations. However, variation was apparent in the fiber fractions of the three complete rations attributable to varying sources of the fibrous crop residues. Overall in vitro DM digestibility (IVDMD) and in vitro NDF digestibility (IVNDFD) % was higher (p<0.05) in T1 (58.59 ± 1.55 and 56.33 ± 0.24) followed by T3 (57.75 ± 0.46 and 54.20 ± 0.64) and in T2 (55.45 ± 0.69 and 53.23 ± 1.25). However, the differences for in vitro CP digestibility (IVCPD) and in vitro ADF digestibility (IVADFD) % were higher in T1 than in other complete rations, the values did not differ statistically. Overall TVFA (meq/L) production was higher in T1 (87.65 ± 7.17) than in T2 (83.00 ± 5.74) and T3 (84.00 ± 5.52). It is concluded that T1 is superior to T2 or T3 in terms of in vitro digestibility (DM, CP, NDF and ADF), in vitro TVFA production and cost of formulation.
