Assessment of tissue-specific cortisol activity with regard to degeneration of the suspensory ligaments in horses with pituitary pars intermedia dysfunction

OBJECTIVE To identify signs of tissue-specific cortisol activity in samples of suspensory ligament (SL) and neck skin tissue from horses with and without pituitary pars intermedia dysfunction (PPID). SAMPLE Suspensory ligament and neck skin tissue samples obtained from 26 euthanized horses with and without PPID. PROCEDURES Tissue samples were collected from 12 horses with and 14 horses without PPID (controls). Two control horses had received treatment with dexamethasone; data from those horses were not used in statistical analyses. The other 12 control horses were classified as old horses (≥ 14 years old) and young horses (≤ 9 years old). Standard histologic staining, staining for proteoglycan accumulation, and immunostaining of SL and neck skin tissue sections for glucocorticoid receptors, insulin, 11β hydroxysteroid dehydrogenase type 1, and 11β hydroxysteroid dehydrogenase type 2 were performed. Findings for horses with PPID were compared with findings for young and old horses without PPID. RESULTS Compared with findings for old and young control horses, there were significantly more cells stained for glucocorticoid receptors in SL samples and for 11 β hydroxysteroid dehydrogenase type 1 in SL and skin tissue samples from horses with PPID. Insulin could not be detected in any of the SL or skin tissue samples. Horses with PPID had evidence of SL degeneration with significantly increased proteoglycan accumulation. Neck skin tissue was found to be significantly thinner in PPID-affected horses than in young control horses. CONCLUSIONS AND CLINICAL RELEVANCE Results suggested that tissue-specific dysregulation of cortisol metabolism may contribute to the SL degeneration associated with PPID in horses.

OBJECTIVE To investigate effects of prednisolone administration on gallbladder emptying rate and gallbladder bile composition in dogs. ANIMALS 6 healthy Beagles. PROCEDURES Prednisolone was administered (2 mg/kg, SC, once daily for 2 weeks) to each dog and tapered over 2 weeks. Gallbladder emptying rate and bile composition were evaluated before and after administration of prednisolone for 2 weeks as well as 1 week after cessation of prednisolone administration. RESULTS Gallbladder emptying rate decreased significantly after prednisolone administration (median, 27%; range, 0% to 38%), compared with rate before administration (median, 59%; range, 29% to 68%), but then increased 1 week after cessation of administration (median, 45%; range, 23% to 48%). Gallbladder bile mucin concentration decreased significantly after prednisolone administration (median, 8.8 mg/dL; range, 6.2 to 11.3 mg/dL), compared with concentration before administration (median, 13.1 mg/dL; range, 10.7 to 21.7 mg/dL), but then increased 1 week after cessation of administration (median, 14.3 mg/dL; range, 9.6 to 26.7 mg/dL). Gallbladder taurochenodeoxycholic acid concentration decreased significantly after prednisolone administration (8.1 mmol/L; range, 6.8 to 15.2 mmol/L), compared with concentration before administration (median, 27.2 mmol/L; range, 22.0 to 31.9 mmol/L), but then increased 1 week after cessation of administration (median, 26.4 mmol/L; range, 15.1 to 31.5 mmol/L). CONCLUSIONS AND CLINICAL RELEVANCE A lower gallbladder emptying rate caused by prednisolone administration may be involved in the pathogenesis of gallbladder disease in dogs. Further studies are required to determine the clinical importance of lower gallbladder bile mucin concentrations caused by glucocorticoid administration in the pathogenesis of gallbladder disease in dogs.

Monvalent inactivated oil-emulsion, Newcastle disease virus (NDV), infectious bronchitis virus (IBV) and Haemophilus paragallinarum vaccines as well as a trivalent one (NDV + IBV + HP) were prepared. The cellular and humoral immune responses of chicken to these vaccines were evaluated by using lymphocytes blastogenesis assay, haemagglutination-inhibition, serum neutralization, tube agglutination and challenge tests. The aim of this study was to investigate the immunogenicity of a plasmid deoxyribonucleic acid (DNA) vaccine encoding the G1 epitope of bovine ephemeral fever virus (BEFV) G glycoprotein in mice. A plasmid DNA carrying the G1 gene was constructed and designated as pcDNA3.1-G1. The expression of the target gene was confirmed in human embryonic kidney 293 (HEK 293) cells transfected with pcDNA3.1-G1 by indirect immunofluorescent staining. Immunisation experiments were intramuscularly carried out by vaccinating 6-week-old female mice in four groups, including the pcDNA3.1-G1 construct, pcDNA3.1 (+) plasmid alone, BEF-inactivated vaccine and phosphate-buffered saline (PBS) (1X) three times with 2-week intervals. Fourteen days after the last immunisation, the animals were bled and the resulting sera were tested for anti-G1-specific antibodies by immunoblotting analysis, indirect enzyme-linked immunosorbent assay (ELISA) and virus neutralisation (VN) test. Serological assays showed that the pcDNA3.1-G1 construct expressing G1 protein was able to elicit specific antibodies against this antigen. Virus neutralisation test showed that pcDNA3.1-G1 could induce anti-BEFV-neutralising antibodies in mice. Our findings indicated that a new dimension can be added to vaccine studies for bovine ephemeral fever (BEF) using eukaryotic expression plasmids encoding the G1 antigen in the future.

Nanoparticles have many characteristics that make them suitable for biological and medical applications. Uptake of thesenano particulates into animals and humans bodiesthrough different routes may exhibit potential side effects. Titanium dioxide (TiO2) is a common additivethat is increasingly used in consumer products, food,pharmaceutical dosage forms and cosmetic articles. In this study, the effects of oral administration of TiO2 nanoparticles (500 mg| kg .bw) for 60 days were investigated on kidney function and histopathological changes. The body weight gain and kidney/body weight ratio showed no significant changes in comparison with control group. There was a significant decrease in total thiol levels in kidney homogenate, the biochemical changes was supported by histopathologicalultration. In conclusion the data shows that the oral administration of TiO2 NPs may lead to renal toxicity in experimental rats.

Ozobranchus spp. are leeches that feed solely on turtle blood. They are common ectoparasites found on a range of marine turtle species, with some species of the leech being implicated as vectors of fibropapilloma-associated turtle herpesvirus (FPTHV). Green (Chelonia mydas) and hawksbill (Eretmochelys imbricata) turtles are the two commonly occurring species in the inner granitic islands of the Seychelles. Routine monitoring of nesting turtles on Cousine Island, Seychelles, allowed for opportunistic sightings of leeches on two hawksbill females. In both cases infestation was low, with three leeches collected off one female turtle and five off the other. No obvious signs of papillomas secondary to infection of FPTHV were seen. All of the turtle leeches collected were determined to be Ozobranchus margoi as they had five pairs of lateral digiform branchiae. The specimens were deposited in the Seychelles Natural History Museum on Mahé. To the best of our knowledge this is the first record of Ozobranchus margoi recorded in the inner granitic Seychelles on hawksbill turtles.

Foot-and-mouth disease (FMD) is one of the major trans-boundary animal diseases in East Africa causing economic loss to farmers and other stakeholders in the livestock industry. Foot-and-mouth disease occurs widely in both Uganda and Tanzania with annual outbreaks recorded. With the recent introduction of the Progressive Control Pathway for FMD control (PCP-FMD) in eastern Africa, knowledge of the spatial and temporal distribution of FMD at the border area between Uganda and Tanzania is helpful in framing engagement with the initial stages of the PCP. Retrospective data collected between 2011 and 2016 from four districts located along the border areas of Uganda and Tanzania, recorded 23 and 59 FMD outbreaks, respectively, for the entire study period. Analysis showed that 46% of the 82 recorded outbreaks occurred in 20% of sub-counties and wards immediately neighbouring the Uganda–Tanzania border and 69.5% of the outbreaks occurred during the dry months. While the serotypes of the FMD virus responsible for most outbreaks reported in this region were not known, previous research reported South African Territory (SAT) 1, SAT 2 and O to be the serotypes in circulation. The results from this study provide evidence of the endemic status of FMD on the Uganda–Tanzania border and emphasise that the border area should be given due consideration during FMD control drives and that cross-border coordination should be prioritised. With the limited data on circulating serotypes in this area, there is a need for more vigilance on FMD case detection, laboratory diagnostic confirmation and provision of more complete documentation of outbreaks. This work further recommends more studies on cross-border livestock movement coupled with phylogenetics in order to understand the spread of the FMD in the border area.

A number of 50 Ardeola ibis ibis birds were found harboring six nematodes species; Tetrameres species, Microtteramere species, Synhimantus invaginatus, Synhimantus equispeculatus, Ascaridia species, Paracamallanus species,and five species of trematodes; Euclinostomum heterostomum, Nephrostomum ramosum, Apharyngostrigea ibis, Apatemon gracilis and Centrocestus armatus. The most common infection by nematodes was (46%) in which highest infection rate Synhimantus invaginatus recorded (30 %) while the trematode infection was (24 %) and Apatemon gracilis was the most prevalent (16 %). Experimental infection of buff backed heron by encysted metacercaria (EMC) and exysted metacercaria (ExMC) of Clinostomum complanatum from freshwater fish Tilapia nilotica, resulted in adult worms formed after 6 days. Where the infection by EMC recorded higher worm burden (14-18 worm) and hatching percent (78%) while the infection by ExMC gave lower worm burden (7-10 worm / bird ) and hatching (48 %). In the present study, it is worthy to mention that buff backed heron act as final host model for Clinostomum complanatum and this will be helpful in further biological and immunological studies for this trematode to decrease its economic losses in fish intermediate host.Fifty random samples of Kareish cheese were collected from different localities in Bani-suef Governorate. All samples were examined chemically for acidity, salt and moisture percent and bacteriologicaly for the presence of Escherichia coli, Staphylococcus aureus, Enterococci, Bacillus cereus, Clostridium perfringens, Yersinia enterocolitica, Salmonella and Shigella species. The obtained results revealed that the mean values of acidity, salt and moisture % were 1.63 ± 0.095,3.55 ± 0.299 and 58.54 ± 0.599 in the examined kareish cheese samples, respectively.Escherichia coli, Staphylococcus aureus, Enterococci, Bacillus cereus, Clostridium perfringens were recovered from 16 (32%), 12(24%), 46 (92%), 25 (50 %) and 3 (6%) with a mean value of 4.86x102 ±4.21x10 2, 4.84x 10 5 ± 2.91x10 5, 3.74x10 6±1.55x10 6, 7.08x10 4±2.61x10 4 and 9.5x10 1 ± 7.37x10 1 of the examined samples , respectively. Yersinia enterocolitica could be isolated from 12% of the examined samples. Salmonella and Shigella species could not be detected in any of the examined samples. The isolated Escherichia coli were examined for serological identification, Enterotoxigenicity and the susceptibility of the isolated serovars to various chemotherapeutic agents. The public health significance and economical importance of the isolated organisms and the recommendations to be followed in the processing, handling and storage of such dairy product were discussed
