Bacterial loads and antibiotic resistance profile of bacteria isolated from the most popular street food (Phuchka) in Bangladesh Texte intégral

Objective: Phuchka is one of the most common street foods in Bangladesh. It is served with salad, sweet and sour tamarind dispersed water, and minced eggs as topping at places where people usually gather. This makes these foods susceptible to bacterial contamination. Therefore, assessing the bacterial load and antimicrobial profile of organisms isolated from phuchka and other foodstuffs served with it was the focus of this study. Materials and Methods: Bacterial isolates were isolated and identified from the samples after the bacterial loads were assessed as total viable count (TVC), total coliform count (TCC), and total staphylococcal count (TSC). The antibiotic resistance profile of the isolates was obtained using the disk diffusion method. Molecular detection of Escherichia coli isolates and the presence of gene responsible for tetracycline resistance was confirmed by polymerase chain reaction. Results: According to the recommendations, the TVC value of 70% phuchka and egg samples was safe, whereas TSC value illustrated that 80% of both phuchka and egg samples were at safety level. For the TCC value, 80% egg and 70% phuchka samples were found to be safe for consumption. Among all the samples, the microbial loads of the vendors hand wash were least in the safety level. Antibiotic sensitivity tests revealed that both Staphylococcus spp. and E. coli isolates were sensitive to gentamicin and ciprofloxacin but showed resistance to ampicillin. Conclusion: The data of this study indicate that phuchka can pose a public health problem as foodborne bacterial isolates which are antibiotic-resistant are found in it. Objective: The work aimed to determine the state of lipid peroxidation products and the activity of the antioxidant system in cats with aseptic inflammation and purulent-inflammatory processes of varying severity. Materials and Methods: The intensity of the process of lipid peroxidation- antioxidant system processes in blood plasma was evaluated using commercial kits. The level of diene conjugates (DC), the content of malondialdehyde (MDA), the level of medium-weight molecules were deter¬mined from lipid peroxidation indices. The state of antioxidant protection was assessed by indi¬cators of superoxide dismutase (SOD) activity, catalase (CT), ceruloplasmin (CP) concentration, glutathione peroxidase activity, glutathione reductase (GLR), and total antioxidant activity (AOA). Results: With aseptic inflammation in the blood of cats, a slow activation of peroxidation reactions occurred. The DC indicator increased by 1.4 times, the SOD level rose by 1.38 times, the amount of GLR by 1.04 times, and the activation of total AOA by 9.06. In sepsis, the values of DC, MDA, and medium-weight molecules increased by 4.4, 8.4, and 8.8 times, respectively. With abscesses in the blood of cats, an increase in CT, CP, and GLR is observed against a background of a decrease in glutathione peroxidase. With purulent wounds, the total AOA of plasma increases by 2.5 times; with abscesses, the total AOA increases by 1.9 times, and with sepsis, it decreases by 1.5 times. Conclusion: With surgical infections in cats, deep biochemical changes or irreversible biochemical changes (in sepsis) occur that indicate a significantly enhanced catabolic orientation of metabolic processes and the accumulation of toxic metabolites, which leads to damage and a decrease in tissue regenerative capacity. Objective: Natural substances found in dietary sources and medicinal plants have attracted con¬siderable attention in recent years as chemopreventive agents. Spirulina is a blue-green alga that possesses chemopreventive properties. The purpose of this study was to determine the effect of spirulina on rats with inorganic arsenic (As) [sodium arsenite (NaAsO2)]-induced lipid peroxidation. Materials and Methods: 120 rats were randomly assigned to 10 groups and designated T0, T1, T2, T3, T4, T5, T6, T7, T8, and T9. One group was kept as a control (T0) that received no treatment. The seven groups received 3.0 mg of NaAsO2/kg body weight in drinking water and were given spirulina ad libitum. T1 was treated with NaAsO2 but not with spirulina. Two groups of rats (T2 and T3), on the other hand, were treated with spirulina without receiving any As (NaAsO2). T2 received agro-based spirulina (Ab-Sp; grown in 1.5% soybean meal media and harvested on day 12 of seed inoculation) at 2.0 gm/kg feed, whereas T3 received commercially available spirulina (Com-Sp) at 2.0 gm/kg feed. T4, T5, and T6 were concurrently treated with Ab-Sp at 1.0, 1.5, and 2.0 gm/ kg of feed. On the other hand, T7, T8, and T9 induced by NaAsO2 were concurrently treated with Com-Sp at 1.0, 1.5, and 2.0 gm/kg feed. All groups received treatment for 90 days. Results: The efficacy of both spirulina in preventing lipid peroxidation caused by As was deter¬mined quantitatively by measuring the rats serum malondialdehyde (MDA). The results indicated that As supplementation increased serum MDA levels, whereas both types of spirulina signifi¬cantly decreased them. The highest dose of Ab-Sp (2.0 gm/kg feed) was found to be the most effective in preventing lipid peroxidation in rats treated with inorganic As. Conclusion: Ab-Sp could be a natural, cost-effective, and safe measure to mitigate As toxicity. [J Adv Vet Anim Res 2021; 8(2.000): 330-338]

Objective: Humpback (hpbk) mice harbor a pathogenic mutation in the Notch3 gene and can serve as a beneficial animal model for investigating human myopathy, kyphosis, and developmen¬tal disorders, including lateral meningocele syndrome. Detection of the point mutation in hpbk mice is important for maintaining strains and scrutinizing genetic rescues, especially considering that homozygous mice are infertile and indistinguishable from their littermates at a young age. This study aimed for the development of a novel, precise, and time-saving genotyping method to identify the mutation in hpbk mice. Materials and Methods: In order to study the hpbk mouse line, we describe how we applied sev¬eral tools, including quantitative polymerase chain reaction (qPCR), multiplex tetra-primer ampli¬fication-refractory mutation system (ARMS-PCR) and Sanger sequencing, toward the recognition of heterozygous and homozygous mice. Results: The Notch3 mutation was clearly identified using qPCR and ARMS assays, but the latter was a more precise and cost-effective approach. The lengths of the ARMS-PCR amplicons are 210 bp and 164 bp for the wild-type and hpbk alleles, respectively. Moreover, the genotyping results for each mouse were corroborated by Sanger DNA sequencing. Conclusion: Our newly developed PCR-based ARMS system affords a swift and precise way to genotype the hpbk mice. ARMS-PCR does not rely on any advanced equipment and is useful as a genotyping method for other model organisms that harbor a pathogenic variant. [J Adv Vet Anim Res 2021; 8(1.000): 7-13]

Objective: The purpose of this work was to investigate the combination of early-age acclimation and linseed dietary inclusion in enriching polyunsaturated fatty acids (PUFAs) in broilers meat as a strategy to mitigate heat stress. Materials and Methods: A total of 400 broiler chicks were assigned to four experimental groups with four duplicates (25 animals each): C: control (basal diet), AC: early-age acclimated (basal diet), Cl: fed 5% ground linseed, and Acl: early-age acclimated and fed 5% ground linseed. The lipid and fatty acid contents of different parts (breast, thigh, liver, subcutaneous, and abdominal fat) of broilers were determined. Results: Low levels of lipids and unsaturated fatty acids have been found in the meat of accli¬mated broilers. Higher levels of linolenic acids were noted in Cl thigh meat compared to C (6% vs. 2.68%, respectively). The results showed that oleic and linoleic acids constitute a large part of the PUFAs of different meats. The most elevated levels of monounsaturated fatty acids were recorded in the breast meat of AcL animals. The highest content of omega-3 was recorded in the liver of AcL animals compared to that of C (14.98% vs. 7.8%, respectively). Conclusion: We suggest that the combination of treatments during hot conditions has led to the reversion of the environment-affected variables to accepted values, and yields better ther¬moresistance, PUFA-enriched meat, and safeguard animal health which conferred to birds better solutions to reduce fatigue and hypoxic activities, which induces a considerable consumption of oxygen. Objective: The effect of seasonality needs to be considered in designing future studies because global warming has caused a rise in ambient temperatures. The objective of the present study is to investigate the effect of high ambient temperatures on fecal score and fecal microflora in dairy cows during summer. Materials and Methods: During the 7 days before the sampling of feces, the daily mean tempera¬tures were 19.9°C in early summer and more than 27.5°C in late summer. Fecal samples were collected from the rectum of cows and the fecal score was evaluated on a 4-point scale. The equalized samples were used to extract the genomic deoxyribonucleic acid (DNA) of the bacteria (Escherichia coli, Salmonella, Lactobacillus, and Bifidobacterium). Results: There was no significant difference in fecal scores between the sampling times in early and late summer. In the populations of the bacteria, there was no significant difference between sampling days in the DNA level of Salmonella, and E. coli in late summer increased to more than three times the level in early summer. However, both levels of Lactobacillus and Bifidobacterium in early summer significantly decreased after 2 months. Conclusion: These data suggest that the increase in temperature in late summer may adversely affect the populations of bacteria in the intestinal environment of dairy cows. In addition, the method used in the present study was sufficient to evaluate the changes in internal and external environmental conditions of dairy cattle. Objective: Fast foods are often responsible for staphylococcal foodborne illness. The present study was carried out to isolate Staphylococcus spp. from various fast foods sold in Mymensingh and to determine their antibiogram. Materials and Methods: Overall, 60 samples of fast foods sold in different restaurants were screened by culture, biochemical tests, and polymerase chain reaction (PCR) to isolate and iden¬tify Staphylococcus spp., followed by employing of disk diffusion method to determine their anti¬biotic resistance patterns. Results: Among these 60 samples, 8 [13.33%, 95% confidence interval (CI): 6.91%24.17%] were positive for Staphylococcus spp. by cultural and biochemical properties. By PCR, four (6.67%, 95% CI: 2.62%15.93%) isolates were confirmed as Staphylococcus aureus by targeting nuc gene. Additionally, all the S. aureus isolates were coagulase-positive. By antibiogram profiles, all the Staphylococcus isolates exhibited resistance to azithromycin and erythromycin (95% CI: 67.56% 100.00%), and frequently resistance to cefixime (75%, 95% CI: 40.93%95.56%), ampicillin (50%, 95% CI: 21.52%78.48%), and amoxicillin (37.5%, 95% CI: 13.68%69.43%); moderate to lower resistance was found against cefotaxime, gentamicin, and doxycycline. In addition, all the isolates were sensitive to ciprofloxacin and chloramphenicol. Interestingly, 75% (6/8; 95% CI: 40.93% 95.56%) isolates were multidrug-resistant (MDR) in nature. Furthermore, the indices of multiple antibiotic resistance (MAR) were ranged from 0.2 to 0.6. Conclusion: This study revealed that fast foods sold in restaurants were contaminated with MDR and MAR Staphylococcus isolates, having potential public health significance. Objective: Duck virus enteritis is a severe viral disease that kills ducks and swans worldwide. The clinical manifestations, gross pathology, molecular detection, and characterization of the duck virus enteritis virus (DVEV) in Australian black swans at a safari park in Bangladesh were described in this case report. Materials and Methods: On a safari park in Bangladesh, an Australian black swan flock exhibited clinical signs of anorexia, greenish watery diarrhea, increased thirst, partial paralysis, and death. Postmortem examinations of deceased swans revealed extensive pathological abnormalities in the trachea, liver, and spleen. To isolate DVEV, a viral inoculum produced from the liver and spleen of dead swans was implanted into 913-day-old embryonated duck eggs via the chorioallantoic membrane (CAM) route. DVEV was confirmed using a polymerase chain reaction (PCR) assay. Phylogenetic analysis was used to determine the genetic relationship between the DVEV isolates from Australian black swans, and 16 DVEV isolates previously described in the GenBank. Results: Hemorrhage was noted in the annular ring of the trachea, as well as an enlarged and hemorrhagic liver and spleen. The PCR assay amplified a 446-bp fragment of the DVEV DNA polymerase gene in the liver, spleen, and CAM homogenates. The phylogenetic analysis found that the DVEV isolates from swans were comparable to those from Bangladesh, India, Vietnam, China, Germany, the USA, and Egypt. Conclusion: According to the findings of this study, the DVEV was the cause of illness and mortality in an Australian black swan flock. [J Adv Vet Anim Res 2021; 8(4.000): 557-562]

Objective: This study was conducted to generate data on the prevalence, species composition, and factors associated with small ruminant nematode infection in the South Omo zone, in South-western Ethiopia. Material and Methods: A cross-sectional study through fecal nematode parasite examination was conducted. Simple floatation test and coproculture, followed by Baermann technique, were used for nematode egg and third-stage larvae (L3) separation and identification. The McMaster method was used to calculate the egg per gram of feces (EPG). Results: From a total of 242 sheep and goats examined, 72.34% were infested with single or mixed nematode parasites. District, Kebele, species, body condition score, and age were significantly (p < 0.05) associated with nematode infestation. Simple logistic regression analysis indicated that nematode infestation in Bena-Tsemay district (78.33%) was significantly (p < 0.05) higher by a factor of 0.54 (OR 95% CI: 0.300.96) than Hamer district (66.39%). Among the species, caprine (79.43%) was significantly (p < 0.05) infested than ovine (62.37%) by a factor of 0.45 (OR 95% CI: 0.250.81). Moreover, infestation on poor (79.12%) and medium (70.96%) body conditioned animals was higher by a factor of 2.94 (OR 95% CI: 1.416.26) and 1.76 (OR 95% CI: 0.883.53) than on good body conditioned animals (63.79%). Likewise, infestation in age groups of 13 years (78.66%) and >3 years (84.40%) was significantly (p < 0.05) higher by a factor of 4.83 (OR 95% CI: 2.3110.46) and 8.23 (OR 95% CI: 3.9817.75) than younger age groups (41.37%), respectively. A moderate parasitic burden was observed on 52.90% of gastrointestinal nematodes (GIN)-infested animals with significantly (p < 0.05) higher EPG in females than males. Furthermore, mixed infestation of Trichostrongylus axei and Eimeria (6.19%), Haemonchus contortus and Eimeria (5.78%), and Trichostrongylus vitrines and Eimeria (5.78%) were dominantly identified. On the contrary, T. axei (15.70%), Eimeria (8.67%), H. contortus (7.43%), and Trichostrongylus colubriformis (7.02%) were dominant single infestations. Conclusion: The current study revealed the highest prevalence of GIN in the study area, which needs strategic control, needs to enhance community awareness toward GIN control and prevention, and to implement further investigation into anthelminthic efficacy to solve the problem. Objective: The influence of the degree of pigmentation, shine, weight, and form index on the hatchability rate of quail eggs (Coturnix coturnix japonica) was examined. Materials and Methods: Three thousand three hundred eggs were incubated after they were classified according to the degree of shell pigmentation: high (HP), medium (MP), and low (LP); shell shine: shiny (SS), and opaque; Egg weight: 13 gm; form index: 78%. The MannWhitney non-parametric test for independent samples was used to evaluate hatching data. Results: Hatchability was best in eggs with HP and SS (p < 0.05): 69.2% and 75.7%, respectively; eggs with MP had the lowest hatchability rate (63.1%). The highest risk of embryo death (17.8%) was observed in eggs containing LP (p < 0.05). Eggs weighing between 12 and 12.9 gm had the highest hatchability rate (p < 0.05) (79.11%). Eggs with a form index >78% had the highest hatch¬ability rate (p < 0.05) (67.62%). Conclusions: HP, MP shell eggs, and SS eggs with a weight of between 12.0 and 12.9 gm and a shape index of greater than 78% have a higher hatchability rate.
