Efficacy of niclosamide and ivermectin suspension preparations in sheep parasitoses Texte intégral

Objective: This study aimed to examine the efficiency of newly formulated drugs based on mech¬anochemical modification of two widely used anthelmintic substances in suspension formulations against the main classes of helminths and parasitic arthropods. Materials and Methods: Solid-phase co-grinding of two substances, i.e., ivermectin (IM) and niclosamide (NS), with licorice extract and sodium succinate was performed in liquid-phase processing to obtain suspension formulations. Drug preparations were administered to the sheep of the experimental groups (5 experimental groups of 8 heads) with different concentrations of active substances, i.e., 3.0, 5.0, and 10.0 mg/kg of body weight (BW) NS and 0.2-mg/kg BW IM. Aqueous suspensions of the original substances in dosages of 0.2-mg/kg BW IM, 10.0-mg/kg NS, and placebo were included as controls. Efficacy of the formulations against gastrointestinal stron¬gyles, Trichuris spp., Moniezia spp., and nasal bot larvae (Oestrus ovis) was assessed in naturally infected sheep by the number of surviving adult parasites post-treatment in the gastrointestinal tract, nasal passages, and sinuses. Results: Formulated suspensions showed an increased solubility of 5.5–13.1 times compared to the original substances. Treatment of sheep with the SusIN-10 drug containing 0.2-mg/kg BW IM and 10.0-mg/kg NS showed 100% efficiency against gastrointestinal strongyles, Moniezia, and nasal bot larvae. Formulations containing 0.2-mg/kg BW IM and 3.0- and 0.5-mg/kg NS could eliminate all of the Trichuris worms. Administration of original substances of IM and NS with rec¬ommended dosages did not result in the adult parasites' complete elimination. Conclusion: Modification of the anthelmintic substances through mechanochemical methods made it possible to create formulations with a targeted spectrum of action, significantly higher water solubility, and optimal parasiticidal activity. Objective: The study aims to prepare the ash filtrate (AF) from household ashes using an in-house-designed handmade filtration system and to observe the antimicrobial efficacy and sanitizing effects. Materials and Methods: Household ashes from various plant sources were collected, and AF was prepared through a handmade filtration system after adding water. The pH of stock AF was measured, and 20%, 40%, 60%, and 80% AF solutions were prepared by adding distilled water in appropriate proportions to obtain a wide range of pH values. The antimicrobial efficacy of AF against Salmonella spp. in vitro, Newcastle disease virus (NDV), and low pathogenic avian influenza virus (LPAIV) H9N2 in ovo were analyzed. Contaminated eggs were individually sprayed or dipped with AF to detect the bacterial load on the eggshell surface. Further experimental use of AF as an egg sanitizer in routine biosecurity operations in broiler sheds was also evaluated. Results: The prepared AF showed high alkalinity; pH varied from 10.7 to 8.20 and contained a higher amount of K, Na, and Cl. The alkaline AF and its dilution gradually inhibited Salmonella growth and showed gradual pH-dependent antibacterial efficacy. Similarly, AF and its dilution showed a gradual decrease in viral titer against the LPAIV (H9N2); however, antiviral activity against the velogenic strain of NDV was quite steady. Applying AF as an egg sanitizer also reduced the bacterial loads significantly on the eggshell surface compared to untreated eggs. Moreover, AF having pH 10.5 experimentally used in routine sanitization practices of a boiler shed resulted in low bird mortality (10/210), higher body weight gain, and a low feed conversion ratio compared to the untreated control flock. Conclusion: The higher alkalinity of the AF is responsible for the antimicrobial activity of commercial disinfectants. Objective: Pigs are used in various biomedical research fields because of their anatomical and physiological similarities to humans. While farm pigs are raised outdoors for several months, minipigs are typically raised in indoor barrier facilities for several years. Although numerous studies have investigated the prevalence of major pathogens, including porcine reproductive and respiratory syndrome virus (PRRSV) and porcine circovirus type 2 (PCV2), in farm pigs, similar studies targeting minipigs are lacking. Materials and Methods: We imported 57 minipigs to our institution from three experimental animal production facilities and immediately assessed the serological and antigenic prevalence of PRRSV and PCV2. Results: PRRSV and PCV2 had seroprevalences of 80.7% and 94.7%, respectively, with 0% antigen positivity rates for PRRSV types 1 and 2 and high-pathogenic PRRSV and PCV2. Two factors could account for the high seroprevalence rates: the majority of individuals may have been vaccinated despite official claims from the origin facilities or exposed to PRRSV and PCV2. Preventing microbial infections is crucial for obtaining accurate and reproducible results. Conclusion: As the first investigation of microbial prevalence in minipigs, our findings indicate that minipigs raised in barrier facilities are not necessarily free from vaccines or infections. Objective: The aim of this study is to determine narrowband UVB (NB-UVB) irradiation's effect on the promotion of skin cancer, particularly its effect on DNA damage, oxidative stress, inflammation, and histological changes in Wistar rat skin. Materials and Methods: Wistar rats were selected for this study and randomly divided into control, dimethylbenzanthracene (DMBA), and DMBA+NB-UVB groups. The rats were given a single dose of DMBA and exposed to NB-UVB 3 times a week for 10 weeks. The radiation dose started with 1 minimal erythema dose, which is equivalent to 3.192 J/cm². In the 11th week, analysis on cyclobutene pyrimidine dimer (CPD), malondialdehyde (MDA), nuclear factor kappa-B (NFκB), inflammatory cytokines, and histopathology examination of the skin tissue was conducted. Results: Higher CPD, MDA, NFκB, tumor necrosis factor a (TNF-a), interleukin (IL)-6, IL-11, IL-10, and IL-12 levels in rats exposed to DMBA+NB-UVB for 10 weeks compared to control and DMBA groups. Macroscopic examination presented erythema, skin thickening, desquamation, ulcer, and crust. Histopathology examination showed hyperkeratosis, acanthosis, atypical keratinocytes,irregular arrangement of the basement membrane, and inflammatory cell infiltration in the DMBA+NB-UVB group. Conclusion: This research has shown that 10 weeks of a combination of DMBA and NB-UVB irradiation induced DNA damage, oxidative stress, inflammation, and histological changes in the Wistar rat skin. Objectives: The current study aimed to investigate the prophylactic potential of EBN compared to EDTA in mitigating Cd's toxic effects on pregnancy rates and embryonic development in rats. Materials and Methods: Ninety-eight female rats (Sprague Dawley) were divided into donor and recipient groups, with donors further divided into seven subgroups, including negative control, Cd-exposed, EBN-treated, and EDTA-treated groups. Embryos from donors were transferred to recipient rats, with EBN and Cd administered for 4 weeks and EDTA given only in the last 5 days for the donor group. Results: Results showed significant differences in pregnancy rates and blastocyst quality. EBN at 120 mg/kg BW led to higher blastocyst production and better quality compared to Cd-exposed groups. The highest pregnancy rates in recipient groups correlated with the highest blastocyst scores from donors. Conclusion: EBN at 120 mg/kg demonstrated significant protection against Cd toxicity and its effect on pregnancy rates, embryo production, quality, and pre- and post-embryo transfer, sur¬passing the effects of both 90 mg/kg EBN and EDTA. Bovine brucellosis is an enduring and formidable challenge in Bangladesh. In this first comprehensive review, we explored the historical, current, and future perspectives of bovine brucellosis outbreaks in Bangladesh. Data spanning from 1984 to 2023 regarding bovine brucellosis in Bangladesh were gathered from literature, reviews, conference papers, and online reports using various search engines and software tools. We considered 29 published documents and analyzed them thoroughly to evaluate the current status of bovine brucellosis for the present comprehensive review. We also suggest policy and other frameworks to mitigate and control the disease on a national scale. Bovine brucellosis in Bangladesh affects various livestock and poses economic burdens due to reduced milk and meat production with potential risks to human health. Over the past decade (2013-2023), the ruminant population in Bangladesh has increased substantially (between 1.46 and 26.95 million individuals), with goats showing the most significant growth, while financial losses due to bovine brucellosis have risen steadily, emphasizing its economic impact (basis on species between Euro currency 12.824 and 298.272). This review highlights brucellosis prevalence, diagnostic challenges, and traditional management practices contributing to its spread. Our findings indicate that bovine brucellosis was reported and prevalent in mild to severe forms across 26 districts of Bangladesh. Bangladesh has initiated measures such as vaccination and awareness campaigns, but effective control remains challenging due to diverse farming systems and resource constraints. A one-health approach is advocated for future strategies, emphasizing community engagement and multidisciplinary efforts to address the complex challenges posed by bovine brucellosis in Bangladesh, ultimately aiming to safeguard livestock health, public health, and economic stability. Objective: The purpose of this study is to evaluate the phenotypic characteristics and typification of Creole goats in five localities of the Ayacucho region in south-central Peru. Materials and Methods: Data from 149 goats (25 males and 124 females) were collected, excluding animals under 2 years of age, pregnant, and sick. Seven qualitative characteristics and 11 zometric measurements were evaluated, and then 9 zometric indices were estimated. Results: In the region, goats with composite colors predominated (76.50%). Additionally, supernumerary (24.20%) and divergent teats (22.60%) were observed in females, while males exhibited a scrotal bifurcation (32.00%). The origin of the flock had a significant effect (p < 0.05) on body measurements, as well as on pelvic index, transverse pelvic index, longitudinal pelvic index, compactness index, and load cannon bone index, except for body index (BOI), proportionality index, dactyl thorax index (DTI), and relative cannon bone thickness index (p > 0.05). In the Ayacucho region, there is a predominance of light animals (57.72%) with a significant DTI (p < 0.05) among the populations, including goats with good balance and capable of walking long distances. Furthermore, there are compact animals (47.65%) with a significant BOI (p < 0.05) among the flock populations, which animals are of the biotype of meat. Conversely, the strongest positive correlation (r = 0.89) was discovered between chest girth and body weight. Conclusion: In the Ayacucho region, there are various creole goat biotypes range from light to very compact heavy goats, with a predominance of meat biotype animals, as well as a marked size dimorphism between localities of origin. Objectives: The research was aimed at determining the staining patterns of Peanut agglutinin (PNA) lectin and Ulex europaeus agglutinin I (UEA-I) lectin in postnatal developing (day 1 to 8 weeks) male genital excurrent duct epithelium in mice. Materials and Methods: Lectin staining was performed on testis and epididymis of 1-day-old and 1-, 2-, 3-, 4-, 5-, 6-, and 8-week-old ICR mice. Results: Rete testis epithelia were UEA-I- and PNA-negative at all postnatal ages. PNA lectin unclearly stained the epithelial apical surface in efferent ductules (ED) and ductus epididymis up to 2 weeks, while UEA-I was negative in those epithelia. By contrast, at 3 weeks of age, PNA and/or UEA-I turned to clearly label epithelia in the excurrent duct system. At 5 weeks, efferent ductules epithelium was UEA-I-negative but strongly PNA-positive; segment I was PNA-negative but faintly UEA-I positive; segment II was weakly PNA-positive but strongly UEA-I-positive; segment III was strongly-PNA- and UEA-I-positive; segment IV was weakly UEA-I-positive but strongly PNA positive; and segment V was faintly UEA-I-positive but strongly PNA-positive. At 5 and 8 weeks (the adult), the staining patterns of PNA and UEA-I lectins were nearly identical. Moreover, an increasing staining intensity was observed in both lectins with the advancement of age until 5 weeks of age. Additionally, it was noted that, with the exception of segment I, the staining intensity of PNA gradually increased while that of UEA-I gradually decreased towards the distal section of the ductus epididymis. Conclusion: Lectin PNA and UEA-I staining, in conclusion, indicated epithelial segments in the male genital duct of mice from the postnatal developing age of 3 weeks, and up until 5 weeks, their staining patterns were nearly identical to those in adults. Objective: The study aimed to identify the effect of pineapple hump extracts in different doses on increasing the chemical and physical quality of rabbit meat. Materials and Methods: The research stages were carried out with maintenance for two months using a completely randomized design consisting of four treatments and five replicates. P0 = complete ration without pineapple hump extract; P1 = complete ration with the addition of 0.2% pine¬apple hump extract; P2 = complete ration with the addition of 0.4% pineapple hump extract; and P3 = complete ration with the addition of 0.6% pineapple hump extract. The variables observed were the chemical quality of meat (pH, moisture, carbohydrate, fat, and protein content in rabbit meat), and the physical quality (water holding capacity (WHC), cooking loss). Results: The data were analyzed using SAS, with significantly different results subjected to Duncan's further testing. The addition of 0.4% pineapple hump extract in rabbit complete rations had a very significant effect on the WHC of 71.62%. The addition of 0.6% pineapple hump extract also had a significant effect on the protein content of meat, increasing it by 19.17%. Conclusion: The effects of pineapple hump extract up to 0.6% in a complete diet of weaned rab¬bits have a positive effect on the physical and chemical quality of rabbit meat, especially on the protein and water-holding capacity of rabbit meat. Objective: We aimed to evaluate the potential of Ampiang-Dadih (AD), a combination of Indonesian traditional fermented buffalo milk (Dadih) and black glutinous rice flakes (Ampiang) as an anti-hypercholesterolemic agent and to prevent liver-cell degeneration using a rat model. Materials and Methods: A mixture of black glutinous rice powder (0.3 gm/gm feed) and fer¬mented buffalo milk (3.74/200 gm BW) was prepared to obtain AD. Fifteen adult male Sprague Dawley rats were divided into three groups of five animals each: (A) negative control group (dis¬tilled water; 5 weeks), (B) hypercholesterolemia group (1% cholesterol per feed; 5 weeks), and (C) preventive AD group (1% cholesterol and AD; 5 weeks). The blood lipid profiles were measured at weeks 2, 4, and 5. The liver enzyme activity, cholesterol level, and histology were observed at the end of week 5. Results: AD administration simultaneously with cholesterol in Group C significantly prevented an increase in total plasma cholesterol and low-density lipoprotein levels compared to Group B. Alanine transaminase and aspartate transaminase were maintained at normal levels in Group C. Furthermore, the levels of liver cholesterol and liver cell degeneration in Group C were also maintained because of AD administration compared to that in Group B. Conclusion: This study demonstrated that AD has the potential to be developed as a functional food for hypercholesterolemia prevention. [J Adv Vet Anim Res 2024; 11(2.000): 275-283]
