Pigeon paramyxovirus type 1 (PPMV-1), a variant of Newcastle disease virus that primarily affects doves and pigeons has been isolated in South Africa since the mid-1980s. Phylogenetic evidence indicates that pigeon paramyxovirus type 1 viruses were introduced in to South Africa on multiple occasions, based on the presence of two separate lineages, 4bi and 4bii, that have been circulating in Europe and the Far East since the early 1990s. During 2006, a PPMV-1 virus was isolated from an African ground hornbil(l Bucorvus leadbeateri) which becamea cutely infected with PPMV-1 and died, probably after scavenging off infected dove carcasses in the region, since a closely-related PPMV-1 strain was also isolated from doves collected nearby. The hornbill isolate had lCPl and MDT values characteristic of PPMV-1s trains. The threat of PPMV-1 to poultry production and biodiversity in southern Africa highlights the importance of monitoring the spread of this strain.

The study is the result of analyzing 16 895 blood smears of cattle collected at 180 sites in the provinces of Manica, Sofala, Zambzia and Tete in Mozambique. Of the blood smears 73.9 % were from Manica, 11.8 % from Tete, 8.5 % from Sofala and 5.8 % from Zambzia; 75.6 % of these were collected from smallholder cattle. Infections with trypanosomes were highest in smallholder cattle from Sofala Province with 36.8 % of the 872 blood smears examined positive for trypanosomes, and lowest in cattle of commercial farmers in Manica Province with only 6.2 % of 2 252 blood smears being positive. Trypanosoma congolense was the predominant species, followed by Trypanosoma vivax and Trypanosoma brucei sensu lato. Trypanosoma brucei, which also infects humans, was more frequent in the districts of Buzi, Mutarara and Morrumbala with 15.1 %, 10.5 % and 9.8 % of all examined cattle in 2005 being infected with it, respectively. The results show a significant increase in the infection rate with trypanosomes compared with results obtained in previous years by the Regional Veterinary Laboratory in Manica Province and by the Regional Tsetse and Trypanosomiasis Control Programme in Zambzia, Tete and Sofala provinces.

The daily requirement for calcium and phosphorus by giraffes to sustain the growth and maintenance of their skeletons is large. The source of sufficient calcium is browse. The source of necessary phosphorus is obscure, but it could be osteophagia, a frequently observed behaviour in giraffes. We have assessed whether bone ingested as a result of osteophagia can be digested in the rumen. Bone samples from cancellous (cervical vertebrae) and dense bones (metacarpal shaft) were immersed in the rumens of five sheep, for a period of up to 30 days, and the effect compared to immersion in distilled water and in artificial saliva for 30 days. Distilled water had no effect on the bones. Dense bone samples were softened by exposure to the saliva and rumen fluid, but did not lose either calcium or phosphorus. In saliva and rumen fluid the cancellous bone samples also softened, and their mass and volume decreased as a result of exposure to saliva, but in neither fluid did they lose significant amounts of calcium and phosphorus. We conclude that although saliva and rumen fluid can soften ingested bones, there is an insignificant digestion of bones in the rumen.

This study investigated the withdrawal periods (WP) of two intramammary antibiotics Cloxamast LC (Intervet SA) and Spectrazol Milking Cow (Schering-Plough Animal Health) in dairy goats and compared them to those recommended for use in cattle. The WP for Cloxamast LC, measured by the Thermo Resistant Inhibitory Substances (TRIS) test, was 60 h in composite samples, 56 h in udder half samples, and the dye was visible for up to 56 h. The WP was significantly shorter than the 72 h recommended WP for use in cattle. It was however significantly longer when the 24 h safety margin (48 h) was subtracted from the recommended WP for cattle. For Spectrazol Milking Cow the antibiotics could be detected by the TRIS test for 61 h in composite samples and 59 h in udder half samples. This did not differ significantly from the recommended 60 h WP for cattle. However, it was significantly longer than that recommended for use in cattle without the 24 h safety margin. There was no significant difference in WP between infected and non-infected udder halves, while there was a weak positive correlation between WP and stage of lactation (R2 = 0.253). There was a moderate positive correlation (R2 = 0.583) between the TRIS test and the presence of dye in milk in udder half samples and between WP in both udder half and composite milk samples (R2 = 0.456). Weak to moderate positive correlations were present between milk yield and the WP in both udder half (R2 = 0.414) and composite (R2 = 0.262) milk samples. Significant differences (P 0.001) were also observed between the milk yield of udder halves with and without palpable udder damage and between samples that tested TRIS positive and negative on both composite (P = 0.008) and udder half samples (P 0.001). There was no significant difference between the milk yield of samples with or without dye. There was a significant difference in milk yield between infected and non-infected udder halves (P = 0.054) and a weak negative correlation between milk yield and stage of lactation (R2 = -0.379).

The prevalence and geographic distribution of the fowl tampan, Argas walkerae Kaiser Hoogstraal, 1969 was determined in the eastern region of the Eastern Cape Province, South Africa by inspecting two fowl houses in the vicinity of each of 72 randomly selected communal cattle dip-tanks. Tampans were collected from 102 (70.8 %) of the 144 fowl houses in the neighbourhood of 57 (79.2 %) of the 72 selected dip-tanks, and the localities of the collections were mapped. Argas walkerae was present in fowl houses from the warm coastal regions of the Indian Ocean in the south to the cold and mountainous Drakensberg in the north-east of the Province. Taking into account the probable sensitivity of the sampling method, it is estimated that A. walkerae is likely to be present in fowl houses belonging to between 74 and 84 % of communities making use of cattle dip-tanks in the eastern region of the Eastern Cape Province, and that when it is present, between 64 and 75 % of fowl houses will be infested. The geographic distribution of A. walkerae seemed to be more strongly associated with the presence of fowls and fowl houses containing raw or processed wood in their structure than with climate.

It is suggested that Haemophilus paragallinarum requires at least three haemagglutinins for adhesionduring infection. This paper reports the partial purification and characterization of the HA-L haemagglutininfrom H. paragallinarum strain 46-C3, a heat sensitive, trypsin sensitive haemagglutinin that hasbeen shown to be the serovar specific haemagglutinin in this organism. Using the pI and molecularmass obtained, it was shown that this protein shares similarities with other types of adhesins found in Gram-negative bacteria. The haemagglutination assay conditions were optimized at pH 7.5 at 37 C. It was also shown that activity is enhanced by the addition of Ca2+ and Mn2+ ions.

In 2000 and 2001 Orange River levels were higher than normal: associated serious outbreaks of blackfly had a substantial detrimental impact on the local economy. The poor control was attributed to the suspected development of larval resistance to temephos. A long-term solution to blackfly control, through the identification of a suitable replacement to temephos for use during high flow conditions, was proposed. This study, however, failed to identify or register a suitable larvicide for use during high flow conditions. Although permethrin was highly effective against blackfly larvae, it was rejected because of its detrimental impacts on non-target fauna. Various formulations of locally produced dry Bacillus thuringiensis var. israelensis (B.t.i.) were tested, but these were ineffective against blackflies. The study also confirmed that resistance to temephos has developed among Simulium chutteri in the middle and lower Orange River. The feasibility of "reversing" the resistance to temephos through the use of the synergist piperonyl butoxide (PBO) was investigated, but the results were not favourable. Furthermore, PBO was highly toxic to blackflies and non-target organisms, and was not recommended for further testing. This means that B.t.i. currently remains the only symptomatic measure of treatment currently applied. Although resistance to B.t.i. has not been reported for blackflies elsewhere in South Africa, there is a need to remain vigilant and to implement an operational strategy that minimizes the risks of resistance developing.

Freshwater snails are known to serve as first intermediate hosts for various parasitic diseases such as schistosomosis and fasciolosis. Snails were collected on several occasions in the proximity of Pretoria, South Africa and their cercarial sheddings were studied. This article describes three different types of cercariae shed by the freshwater snail, Lymnaea natalensis, viz. a fork-tailed cercaria of a Trichobilharzia sp., an avian parasite belonging to the family Schistosomatidae, an echinostomatid cercaria of the family Echinostomatidae, also avian parasites and a xiphidiocercaria of the family Plagiorchiidae which parasitise avians and amphibians. The morphology of these cercariae was studied by light and scanning electron microscopy.

In four Kenyan pig breeding units the pregnancy diagnosis of sows has been carried out in two groups: Group 1 (n = 1911): the sows were transrectaly pregnancy tested between Days 1722 post-mating by ultrasound. Sows testing non-pregnant immediately received one dose of 400 IU pregnant mare serum gonadotropin (PMSG) (equine chorion gonadotropin, eCG) and 200 IU human chorion gonadotropin (hCG). On showing signs of oestrous, the animals were subsequently artificially inseminated (AI). Group 2 (n = 1923): sows were pregnancy tested by serum progesterone (P4)-based enzyme-linked immunosorbent assay (ELISA) on Day 17 post-breeding. P4 concentrations were categorized as positive ( 5 ng/ml) or negative ( 5 ng/ml). Sows testing non-pregnant immediately received one dose of 400 IU PMSG and 200 IU hCG by injection, and were subsequently artificially inseminated. The following parameters were evaluated: sows diagnosed non-pregnant, days from first post-weaning insemination until the sows were inseminated at their first return to oestrus; farrowing rate and total piglets born and number of live-born piglets in litters.The percentage of sows diagnosed non-pregnant in the two groups, as well as the totals of born piglets and of live-born piglets in litters did not differ significantly between the two groups. The number of days from the first post-weaning mating until the sows were artificially inseminated at their first return to oestrus and the administration of eCG and hCG was shorter (P 0.01) and farrowing rate was higher (P 0.01) in the ELISA-tested sows.

The percentage of sows diagnosed non-pregnant in the two groups, as well as the totals of born piglets and of live-born piglets in litters did not differ significantly between the two groups. The number of days from the first post-weaning mating until the sows were artificially inseminated at their first return to oestrus and the administration of eCG and hCG was shorter (P < 0.01) and farrowing rate was higher (P < 0.01) in the ELISA-tested sows.

Lactose in normal saline was administered intravenously to a group of Zebu cattle infected with Trypanosoma vivax to determine the bloodplasma kinetics at onset of an experimental infection and its ability to protect tissues against damage as part of preliminary studies to determine its suitability for use in the treatment of trypanosomosis. Significant (P 0.01) higher lactose concentrations were observed in the T. vivax-intecled bulls at 30 min and 1h (P 0.05) post-infectio (p.i.) and by 4 h p.i. the plasma lactose remained above the level prior to infusion, after which it fell slightly below the preinfusion level in the uninfected group. Calculated pharmacokinetic parameters revealed delayed excretion of lactose in the T. vivax-intected group soon after infection. The total body clearance (C/B )was significantly (P 0.05) reduced. The biological half-life (t1/2), elimination rate constant (kel) and apparent volume of distribution (Vd) were relatively decreased (P 0.05) as a result of the T. vivax infection. Retention of lactose in the plasma was attributed to decreased plasma clearance l.t is suggested that the presence of trypanosomes in circulation rather than organic lesions could have been responsible for the delay observed in the excretion of lactose.At 12 weeks p.i., when the experiment was terminated, the group infected and given lactose infusion (despiteh igherp arasitaemia) had no gross or histopathological lesions in the brain, spleen, lymphnodes, heart, kidneys, liver and testes. However, the group infected but not infused with lactose were emaciated, had pale mucosae, watery blood, general muscular atrophy, serous atrophy of coronary fat and other adiposet issue, hepatomegalys, plenomegalys, wollen and oedematous lymph nodes, all of which are suggestive of trypanosomosis. Histopathological lesions included arrowing of Bowman's space and hypercellularity of glomerular tufts in the kidneys with the mean glomerula truft nucleairn dices (GTNs) in the group significantly higher (P 0.01)than the mean GTNs of the lactoseinfused and control bulls. Degenerative changes occurred in the myocardium, spleen, testes and epididymides. The tesicular and epididymal lesions are indicative of male reproductive dysfunction.
