O objetivo do estudo foi verificar a contaminação do solo de peridomicílios de uma comunidade pesqueira localizada no município de Bonito, Mato Grosso do Sul, Brasil, onde é frequente a ocorrência de casos de Larva migrans cutânea. Utilizando-se das técnicas de centrifugo-flutuação, Baermann-Moraes e centrifugo- sedimentação, amostras de solo e fezes de cães foram analisadas e os resultados evidenciaram altos índices de contaminação por ovos de Toxocara spp., Ancilostomídeos e larvas de nematoides. Estes achados podem estar associados aos casos de Larvas migrans cutânea e às elevadas taxas de eosinofilia em escolares daquela comunidade. | The aim of this study was to investigate soil contamination in peridomiciliary areas from a fishing community in the municipality of Bonito, Mato Grosso do Sul state, Brazil, where is often found the occurrence of cases of cutaneous larva migrans. Using the techniques of zinc sulphate centrifugal flotation, Baermann-Moraes and centrifugal sedimentation, soil samples and feces of dogs were analyzed and the results showed high levels of contamination by Toxocara spp., hookworm and larvae of nematodes. These findings may be associated with cases of cutaneous larva migrans and high rates of eosinophilia in school children of that community.

O vírus da cinomose canina (CDV) é um patógeno que afeta cães, causando doença grave e que pode levar a morte. Os cães infectados pelo CDV podem ser diagnosticados pela detecção do RNA utilizando-se a técnica de Nested-PCR. O presente estudo teve como objetivo avaliar o RNA do CDV no sangue, urina e saliva em cães com diagnóstico clínico de cinomose. A técnica de Nested-PCR foi capaz de detectar o RNA em diferentes tipos de amostras biológicas. Obteve-se um maior número de resultados positivos em amostras de urina. | Canine distemper virus (CDV) is a pathogen which affects dogs and causes severe disease leading to death. Dogs infected with CDV can be diagnosed by RNA detection by Nested PCR technique. The following study proposed to valuate CDV RNA in blood, urine and saliva samples. The Nested-PCR technique was able to detect CDV RNA in different types of biologic samples. The higher number of positive results was obtained in urine samples.

<p class="MsoNormal"><span style="mso-ansi-language: EN-US;" lang="EN-US">The Green fluorescent protein (GFP) was first described after being extracted from Aequorea victoria in 1987. Since then, GFP and its derivatives have been widely used in several experiments as cell and protein marker. In the present study it was verified the genotype of the offspring from crosses between heterozygote Lewis LEW-Tg (EGFP) F455.5/ Rrrc rats and analyzed the expression of the enhanced green fluorescent protein (EGFP) in different cell types and genotypes. The genotype of the offspring was assessed by PCR and analysis of EGFP expression in different cells and genotypes, including mesenchymal stem cells (MSC) derived from adipose tissue and calvarial osteoblast cells. Expression of EGFP was verified by flow cytometry, fluorescence microscopy, and immunostaining. Through these methods, it was identified the genotypes of the offspring and determined the levels of expression of EGFP in two cell types. A difference in expression between the (EGFP +/+) and (EGFP +/-) genotypes was also observed in addition to the presence of autofluorescence. Further studies on the natural fluorescence of cells with the (EGFP +/-) genotype and that induced by presence of the EGFP are necessary.</span></p> | <p class="MsoNormal">A proteína fluorescente verde (GFP) foi descrita pela primeira vez após ter sido extraída de Aequorea victoria em 1987. Desde então, a GFP e seus derivados têm sido amplamente utilizados em várias experiências como marcador celular e de proteínas. O objetivo do presente estudo foi o de verificar o genótipo dos descendentes de cruzamentos entre ratos Lewis LEW-Tg (EGFP) F455.5/Rrrc heterozigotos e de analisar a expressão da proteína fluorescente verde melhorada (EGFP) em diferentes tipos celulares e genótipos. O genótipo da descendência foi avaliado por PCR e pela análise da expressão da EGFP em diferentes células e genótipos, incluindo-se as células-tronco mesenquimais (MSC) derivadas de tecido adiposo e de osteoblastos de calvária. A expressão da EGFP foi verificada por citometria de fluxo, microscopia de fluorescência e imunocoloração. Foram, identificados os genótipos da descendência e determinados os níveis de expressão de EGFP em dois tipos de células. Foi também constatada uma diferença de expressão entre os genótipos (EGFP +/+) e (EGFP +/-) além da presença de autofluorescência. Mais estudos são necessários para esclarecer a fluorescência natural de células com o genótipo (EGFP +/-) e aquela induzida pela presença da EGFP.</p>

<p>Bones are believed to be an effective barrier to obtain sonographic images. In fact, the large difference between acoustic impedances of surfaces of the soft and bone tissues generates significant image artifacts. However, transmission of the ultrasound beam depends on bone thickness and structure. Therefore, the temporal bone has been used as an acoustic window to access the brain of adult patients with ultrasonography. The purpose of this study was to assess the brain of adult dogs using ultrasonography with and without bone interposition, compare the images, and correlate them with the brain anatomy. Ten mesaticephalic adult dogs were used, and the ultrasound examination was performed through the magnum orifice and on the temporal, lateral parietal, and frontal bones. A small craniotomy was performed in the frontal bone for examination without bone interposition. We were able to acquire images of the brain with bone interposition. However, resolution of these images was lower than the ones obtained by craniotomy. Important anatomical structures were identified. Regarding the correlation and the wide availability of ultrasound equipment, it was concluded that ultrasound can be used as a tool for monitoring expansive intracranial lesions or in intraoperative procedures.</p> | <span style="font-size: 11.0pt; line-height: 115%; font-family: 'Calibri','sans-serif'; mso-ascii-theme-font: minor-latin; mso-fareast-font-family: Calibri; mso-fareast-theme-font: minor-latin; mso-hansi-theme-font: minor-latin; mso-bidi-font-family: 'Times New Roman'; mso-bidi-theme-font: minor-bidi; mso-ansi-language: PT-BR; mso-fareast-language: EN-US; mso-bidi-language: AR-SA;">Os ossos são tidos como uma barreira intransponível para obtenção de imagens sonográficas, pois a grande diferença entre as impedâncias acústicas das superfícies de tecidos moles e ósseos gera importantes artefatos de reflexão, contudo, a transmissão do feixe ultrassônico depende da espessura e da estrutura óssea e neste caso o osso temporal tem sido usado como uma janela acústica para o acesso, por ultrassonografia, do encéfalo de pacientes adultos. O presente trabalho examinou o encéfalo de cães adultos com o emprego da ultrassonografia com e sem a interposição do crânio, comparou as imagens obtidas e as correlacionou com a anatomia encefálica. Foram examinados dez cães adultos, mesaticefálicos. O exame ultrassonográfico foi realizado através do orifício magno e dos ossos temporal, parietal (porção lateral) e frontal. No exame sem a interposição óssea foi realizada uma pequena craniotomia no osso frontal. As imagens do encéfalo com a interposição óssea apresentaram resolução inferior às obtidas por craniotomia. Importantes estruturas anatômicas foram identificadas. Devido à correlação e a sua ampla disponibilidade, concluiu-se que o aparelho de ultrassom pode ser usado para o acompanhamento de lesões intracranianas expansivas, bem como em procedimentos intraoperatórios.</span>

<p class="MsoNormal"><span style="mso-ansi-language: EN-US;" lang="EN-US">The occurrence of visceral or cutaneous canine leishmaniasis cases among dermatophatic dogs admitted to the veterinary teaching hospital School of Veterinary Medicine and Animal Husbandry - University of São Paulo from 1997 to 2008 was retrospectively characterized. For the diagnosis of 427 animals, serological (Enzyme-Linked Immunosorbent Assay, Indirect Immunofluorescence and/or Complement Fixation), parasitological and molecular (Polymerase Chain Reaction) tests were performed, which resulted in 117 (27.4%) confirmed positive cases. Of these, 96 (82.1%) were pure breed dogs and the others 21 (17.9%) were cross breed, being 57 (48.7%) females and 60 (51.3%) males. In terms of age, 26 (22.2%) were younger than 36 months, 48 (41%) were between 36 and 84 months, and 43 (36.8%) were older than seven years old. Sixty-four (54.7%) dogs were considered oligosymptomatic, 44 (37.6%) symptomatic, and the remaining nine (7.7%) asymptomatic. The main clinical findings were, in descending order, dermal lesions, lymphadenopathy, anemia, dysorexia, prostration and splenomegaly. As for hematological disorders, relative and absolute lymphopenia was the most important findings. Ninety-six (82.1%) of the positive animals correspond to allochthonus cases from São Paulo municipality.</span></p> | <p class="MsoNormal">Caracterizou-se retrospectivamente, a partir do ano de 1997 até março de 2008, a ocorrência de casos de leishmaniose, visceral ou tegumentar, dentre os cães dermatopatas atendidos em serviço especializado de hospital-escola veterinário de Faculdade de Medicina Veterinária e Zootecnia da Universidade de São Paulo. Para o diagnóstico em 427 animais, foram utilizados testes sorológicos (Ensaio Imunoenzimático Indireto, Imunofluorescência Indireta e/ou Fixação de Complemento), parasitológicos diretos e moleculares (Reação em Cadeia da Polimerase), que resultaram na confirmação de 117 (27,4%) casos positivos. Da amostragem, 96 (82,1%) eram cães de raça definida, os demais 21 (17,9%) não dispunham de plena definição racial, sendo 57 (48,7%) fêmeas e 60 (51,3%) machos. Quanto à faixa etária, 26 (22,2%) apresentavam idade inferior a 36 meses, 48 (41%) entre 36 e 84 meses e 43 (36,8%) ultrapassavam o sétimo ano de vida. Considerou-se 64 (54,7%) cães como oligossintomáticos, 44 (37,6%) sintomáticos e os nove (7,7%) remanescentes como assintomáticos. As principais alterações clínicas observadas foram, em ordem decrescente, lesões tegumentares, linfonodomegalia, anemia, disorexia, prostração e esplenomegalia. Quanto às alterações hematológicas, a mais importante foi a linfopenia relativa e absoluta. Dos animais positivos para a enfermidade, 96 (82,1%) correspondeu a casuística alóctone ao Município de São Paulo.</p>

<p class="MsoNormal"><span style="mso-ansi-language: EN-US;" lang="EN-US">The knowledge of the mechanisms that affect the control of the ovarian activity is essential for the success of reproduction biotechnologies. Although a number of studies have been carried out in which the luteinizing hormone (LH) was used to control the ovarian activity, little is known about its influence in the morphology and vascular formation of the corpus luteum, aiming to increase the local blood flow. Thus, the objective of the present experiment was the quantification of the vascular density of corpora lutea (Cls) in animals treated with human chorionic gonadotropin (hCG) just after ovulation. Therefore, eighteen wistar rats were used in this experiment. Eight rats in the treated group and ten rats in the control group. Corpora lutea were divided into two groups: group (A) treated with hCG in the following morning after copulation, and group (B) control animals which received an injection of 0.9% sodium chloride solution. Ovaries from each group were used for preparation of histological sections for vascular density qualification. No statistical significance was found between the two groups tested.</span></p> | <p class="MsoNormal">A compreensão dos mecanismos de controle da atividade ovariana é necessária para o sucesso das biotecnologias da reprodução. Embora existam inúmeros trabalhos a respeito da aplicação do hormônio luteinizante (LH) na função ovariana, pouco se sabe sobre a sua influência na morfologia e formação da vasculatura do corpo lúteo (CL). Diante disto, o presente projeto teve como objetivo a quantificação da densidade vascular dos CLs de animais tratados com Gonadotrofina Corionica Humana (hCG) após a ovulação. Para tanto, foram utilizados ratas wistar, cujos CLs foram divididos em dois grupos: (A) tratado com hCG na manhã seguinte a cópula e (B) controle (solução fisiológica a 0,9 % de NaCl). Foram confeccionadas lâminas dos ovários dos animais para a quantificação da densidade vascular. Os resultados obtidos não revelaram diferenças significantes entre a densidade vascular dos grupos tratado e controle.</p>

To measure florfenicol concentrations in ovine tear fluid after IM and SC administration and determine minimum inhibitory concentrations (MICs) of florfenicol against field isolates of Mycoplasma organisms potentially involved in infectious keratoconjunctivitis. [br/]ANIMALS: 9 healthy adult Lacaune ewes. [br/]PROCEDURES: Animals received an IM and SC administration of florfenicol (20 mg/kg) in a 2-way crossover design. Samples of blood and tear fluid were collected before and for 24 hours after administration. Concentrations of florfenicol in plasma and tear fluid were measured via high-performance liquid chromatography. The MIC of florfenicol for various Mycoplasma strains cultured from sheep and goats was determined via an agar dilution method. [br/]RESULTS: Mean florfenicol concentration in tear fluid for the 24-hour period was significantly higher after IM administration (0.70 μg/mL) than after SC administration (0.22 μg/mL) and was maintained for a longer duration. The lacrimal fluid-to-plasma concentration ratio was not different between the 2 routes of administration, with mean values of 40.2% and 32.5% after IM and SC administration, respectively. The MIC for Mycoplasma agalactiae, Mycoplasma conjunctivae, and Mycoplasma mycoides isolates ranged from 0.5 to 8 μg of florfenicol/mL. Two strains of M agalactiae could be considered resistant to florfenicol. [br/]CONCLUSIONS AND CLINICAL RELEVANCE: Florfenicol readily penetrated the preocular tear fluid of sheep after IM and SC administration. For both routes of administration, doses > 20 mg/kg would be necessary to achieve tear fluid concentrations of florfenicol greater than the MICs for most strains of Mycoplasma organisms.

The outbreak of chlamydiosis in one of the western provinces of Poland, was diagnosed accidentally as a concurrent infection in a commercial laying hen flock during an outbreak of fowl pox. For histological examination, skin and subcutaneous tissue samples from lesions on heads of the birds were collected. Swabs from throat and trachea have been examined by nested PCR, real-time PCR, and partial ompA sequencing. Detailed electron microscopy analysis revealed fowl pox intracytoplasmic inclusions, called Bollinger bodies, and the presence of other intracytoplasmic inclusions; specific for Chlamydia sp. Results of nested PCR confirmed the presence of Chlamydiaceae sp. in two tested samples. Surprisingly, one of the two Chlamydiaceae-positive cases turned out to be infected with a non-classified strain. Results of real-time PCR and sequencing confirmed the presence of a new Chlamydia species that has not been found in Poland to date. Partial sequencing and BLAST analysis of ompA gene sequence confirmed the highest homology to non-classified poultry strains of Chlamydia sp. that were previously detected in Germany and France. The zoonotic potential and the exact taxonomic status of this atypical strain have yet to be defined.
