Acacia angustissima intoxication of Menz lambs requires two components Texte intégral

This study sought to determine if sheep suffer neurological symptoms when fed Acacia angustissima leaves, and whether an equivalent amount of 70% acetone extract would have the same effect. In addition, the study tried to determine if treatment of leaves with 70% acetone would destroy the activity of A. angustissima toxins, and whether extraction with 70% aqueous acetone extract would separate two components of a toxic system. Twenty-five Menz lambs were randomly assigned to one of five treatments (1) A angustissima leaves as half the diet, 2) dried extract (70% aqueous acetone) of the same quantity of leaves, 3) a corresponding amount of residues, 4) a recombination of the dried extract and dried residue, or (5) a control diet containing no A angustissima leaves or extract fractions. All animals fed the leaves and the recombined fractions died or were euthanized when they were observed to be dying of severe neurological derangement. None of the other animals showed any neurological signs of impairement. The results of this study indicate that healthy, well-fed sheep can be poisoned by A angustissima, that the toxins are not destroyed by acetone or oven drying, and that severe neurological intoxication requires two components, which can be separated by acetone extraction.

peer reviewed | OBJECTIVES: To compare the values of the urodynamic parameters of the lower portion of the urinary tract and vaginourethral measurements obtained during the phases of the estrous cycle in dogs and determine possible functional or anatomic modifications of the lower portion of the urinary tract associated with those phases. ANIMALS: 7 adult female Beagles. PROCEDURE: Urethral pressure profilometry, diuresis cystometry, and vaginourethrography were performed in each dog during proestrus; estrus; early, mid, and late diestrus; and early and late anestrus. The maximum urethral pressure (MUP), maximum urethral closure pressure (MUCP), urethral functional and anatomic profile lengths (UFPL and UAPL, respectively), integrated pressure, threshold pressure, threshold volume, compliance, urethral length, and vaginal length and width were measured. RESULTS: For all measurements, significant interindividual variation was detected. Integrated and threshold pressures, APL, and each morphometric value significantly increased from late anestrus to proestrus. Compared with other phases, MUP, MUCP, and integrated pressure values were significantly lower in estrus and early diestrus; UAPL and UFPL values were significantly lower in late diestrus. At each cycle phase in old dogs, MUP, MUCP, threshold pressure, and vaginal length and width were significantly lower (except in proestrus for vaginal measurements) and threshold volume and compliance values were significantly higher, compared with middle-aged dogs. CONCLUSIONS AND CLINICAL RELEVANCE: Urodynamic and morphometric measurements of the lower portion of the urogenital tract are affected by the changes in hormonal balance that occur during the estrous cycle. In sexually intact female dogs, estrous phase determination is important for the interpretation of urodynamic data.

peer reviewed | OBJECTIVE: To determine effects of obesity and diet in dogs on plasma lipid and lipoprotein concentrations by assaying plasma leptin and ghrelin concentrations and determining total plasma cholesterol and triglyceride concentrations as well as the concentrations of cholesterol and triglycerides in various lipoprotein classes (ie, very-low-density, low-density, and high-density lipoproteins). ANIMALS: 24 Beagles; 12 lean (mean [+/- SEM] body weight, 12.7 +/- 0.7 kg) and 12 chronically obese (21.9 +/- 0.8 kg) dogs of both sexes, between 1 and 9 years old. PROCEDURES: Total plasma cholesterol and triglyceride concentrations; lipoprotein cholesterol and triglyceride concentrations; and plasma ghrelin, leptin, free fatty acids, insulin, and glucose concentrations were measured and compared between lean and obese dogs, both of which were fed a complete and balanced maintenance diet. Chronically obese dogs were subsequently fed a high-protein low-energy diet to evaluate effects of diet composition on plasma lipid and lipoprotein measurements. RESULTS: Chronic obesity resulted in a significant decrease in plasma ghrelin concentration and a significant increase in plasma leptin, cholesterol, and triglyceride concentrations in dogs. High total plasma cholesterol and triglyceride concentrations resulted from increased cholesterol and triglyceride concentrations in all lipoprotein fractions. In obese dogs, modification of diet composition resulted in beneficial effects on plasma lipid and leptin concentrations, even before weight loss was observed. CONCLUSIONS AND CLINICAL RELEVANCE: Correlations exist between obesity and plasma measurements (ie, lipoproteins, leptin, insulin, and ghrelin) commonly associated with obesity. Modification of diet composition to control energy intake improves plasma lipid and leptin concentrations in obese dogs.

O amitraz possui efeitos sistêmicos resultantes da atividade intrínseca com receptores adrenérgicos do tipo a-2. Nesse estudo, avaliou-se os efeitos das injeções epidurais de amitraz (0,1 e 0,4 mg.kg-1) ou DMSO 10% (5,0 mL) sobre a freqüência cardíaca (FC), pressão arterial sistólica (PAS), freqüência respiratória (FR), motilidade ruminal (MR), temperatura retal (TR), altura de cabeça (AC) e as latências da resposta a estímulos nociceptivos nas regiões da coxa (LECC) e coroa do casco (LRRM) de vacas. Houve uma diminuição das variáveis FC e MR nos animais que receberam amitraz. Não houve diferenças significativas na FR, PAS e AC entre os grupos estudados. A LECC e a LRRM foram maiores no grupo amitraz (0,4 mg.kg-1) entre 25 e 120 minutos. Concluiu-se que o amitraz aumenta a latência de resposta a estímulo nociceptivo, sem induzir efeitos colaterais sistêmicos severos nas doses utilizadas, por via epidural em vacas. | The amitraz is widely used in veterinary medicine as an acaricide in small and large animals. Several reports showed that the systemic effects caused by epidural administration of this substance are the result of its interaction with a-2 adrenoceptors. In the present study, the effects of epidurally amitraz (0,1 or 0,4 mg.kg-1) or 10% DMSO (5,0 mL) on the heart rate (HR), systolic arterial pressure (SAP), respiratory rate (RR), ruminal motility (RM), rectal temperature (RT), head height (HH), and latency to heat-evoked reflexes (LSPR - skin of perineal region; LHWR - hoof withdrawal reflex) were evaluated. The HR and RM decreased in amitraz groups. There were no statistical differences in RR, SAP and HH among groups. LSPR and LHWR were prolonged in amitraz 0,4 group (T25-T120). We concluded that epidural amitraz does not produce severe systemic effects, and that the higher dose induces prolongation of the latency to heat-evoked reflexes, in cows.

O objetivo deste estudo foi identificar mudanças do epitélio vaginal em fêmeas de "Cerdo Pelón Mexicano", que ovularam durante o lactação, estágio causado pelo efeito da presença de macho e retirada da leitegada. A avaliação do estro foi feita através de citologia de raspado vaginal, observação do comportamento das fêmeas e por determinação de 17 ß estradiol e de progesterona no 8º dia após o início de lactação. Foram formados quatro grupos de fêmeas: Grupo 1 não sofreu estímulo; Grupo 2 permaneceu com o macho; Grupo 3 foi separado sua leitegada por 4 h e grupo 4 recebeu ambos estímulos. Amostras de raspado vaginal foram coletadas a cada 24 horas durante 5 dias após o estímulo. ANOVA para amostras repetidas foi realizada durante os 5 dias do teste. O estímulo utilizado no Grupo 4 causou modificações significativas (P < 0·001) quando comparado aos Grupos 1, 2 e 3. Os níveis de estradiol foram mais altos que 30 pg/ml no Grupo 4 no 10º dia pós parto e 4.5 ng/ml de progesterona nos 11º e 12º dias pós parto. Ficou evidente que 100% das fêmeas nos Grupos 1, 2 e 3 não mostraram atividade de estro quando foi relacionado citologia vaginal com o comportamento estral e determinação hormonal da fase de lactação das fêmeas, ao passo que 100% das fêmeas no Grupo 4 apresentaram estro 72 horas após os estímulos e ovularam 24 a 36 horas do início do cio, o que foi comprovado pela determinações de estradiol e progesterona, respectivamente. | The objective of this study was to identify changes of the vaginal epithelium in Mexican hairless sows, which ovulated during lactation, caused by the effect of the boar presence and the litter withdrawal. In order to determine the oestrus stage, an exfoliative vaginal cytology and 17b estradiol and progesterone determinations were carried out on the 8 day after the onset of lactation out accompanied with behaviour observations. Four groups of sows were used: Group 1 was not stimulated; Group 2, remained with the boar; Group 3 was separated from its litter for 4 h and Group 4 got both stimuli. Vaginal smear samples were collected every 24 h. for 5 days after stimulus. An ANOVA statistical analysis was performed for repetitive samples during the 5 days of the test. Stimuli used in group 4 caused significant modifications (P< 0·001) when compared to Groups 1, 2 y 3. Estradiol levels were higher than 30 pg/ml in Group 4 on day 10 post partum and 4.5 ng/ml of progesterone on day 11 and 12 post partum. It was evident that 100% of the sows in Groups 1, 2 and 3 did not show oestral activity when relating vaginal cytology with the oestral behaviour and hormone determination of the lactating sows, whereas 100% of the sows in group 4 presented oestrus 72 h. after the stimulus and ovulated 24 to 36 h after the oestrus onset, this was corroborated by estradiol and progesterone determinations, respectively.

A ultraestrutura da glândula de Mehlis de Metamicrocotyla macracantha, parasita de brânquia coletado de Mugil liza do Rio de Janeiro, Brasil, foi estudado através da microscopia eletrônica de transmissão. A glândula de Mehlis consiste de dois tipos de células secretoras, S1 e S2, cada uma produzindo um corpo secretor diferente. Os corpos S1 são esféricos, em forma de lamelas e observados em diferentes estágios de desenvolvimentos no citoplasma dessas células. Os corpos S2 são esféricos a ovais com conteúdos densos, apresentando uma estrutura cristalina. O citoplasma das células da glândula de Mehlis apresenta também ribossmas livres, retículo endoplasmático granular e complexo de Golgi, organelas características de células secretoras. | The ultrastructure of Mehlis' gland of Metamicrocotyla macracantha, a gill parasite collected from Mugil liza from Rio de Janeiro, Brazil, was studied by transmission electron microscopy. The Mehlis' gland consists of two types of secretory cells, S1 and S2, each producing a different secretory body. The S1 bodies are spherical, lamellae-like and were observed in different stages of development in the cytoplasm of these cells. The S2 bodies are spherical to ovoid with dense content, showing a crystalline structure. The cytoplasm of Mehlis' gland cells contains also free ribosomes, granular endoplasmatic reticulum and Golgi complex, characteristic organelles of secretory cells.

Com objetivo de avaliar o efeito de dois protocolos de descongelação do sêmen canino congelado em três diluidores foram coletados ejaculados de nove cães. Cada ejaculado foi dividido em 3 amostras, centrifugado e os pellets tratados sob diferentes protocolos: 1º - meio á base de glicina e gema de ovo; 2º - meio á base de TRIS e 3º - meio M9. Foram envasadas 28 palhetas de 0,5ml segundo cada protocolo. Amostras tratadas sob cada protocolo foram avaliadas (M1) quanto a motilidade, vigor e integridade das membranas. As palhetas foram refrigeradas a 5ºC por 60 min. e congeladas em N2. Uma amostra de cada protocolo foi avaliada quanto: motilidade, vigor e integridade das membranas após o equilíbrio (M2). Eram sempre descongeladas três pares de palhetas, sendo que cada par havia sido congelado sob um mesmo protocolo. Uma das palhetas de cada par era descongelada a 37ºC por 30 segundos e outra a 72ºC por 8 segundos. Cada grupo foi avaliado após o descongelamento (M3) quanto a motilidade e vigor espermático, avaliação computadorizada do movimento, integridade das membranas e avaliação acrossomal por meio de Fitc-PNA e PI. As amostras foram mantidas em banho Maria a 37ºC por 1h para o teste de longevidade espermática e reavaliadas (M4) quanto à motilidade e vigor espermáticos, integridade das membranas. Após análise estatística concluímos que as células espermáticas caninas descongeladas a 72ºC por 8 segundos apresentaram um maior somatório de bons resultados quanto aos testes de viabilidade espermática in vitro empregados. | To verify the effect of three different extenders and two thawing temperatures on frozen-thawed canine sperm characteristics, one ejaculate from nine dogs were separately collected and processed (n=9). Each ejaculate was divided into 3 equal samples and centrifuged. The pellets were re-suspended using: 1st pellet - Glicyne-egg yolk extender (GEY), 2nd pellet - TRIS extender and 3rd pellet - M9 extender and all samples were filled in 0.5ml straws. A total of 84 straws (28 for each protocol) were done. The sperm motility, vigour and plasma membrane integrity from each protocol were immediately evaluated (M1) and the straws were brought to a refrigerator at 5ºC for 60 minutes. After that, a sample from each protocol was warmed up in a water bath 37ºC for 5 min. and sperm motility, vigour and plasma membrane integrity were evaluated (M2). The straws were frozen in liquid nitrogen. One straw from each protocol was thawed at 37ºC for 30 sec and another at 72ºC for 8 sec and the sperm motility; vigour, CASA and plasmatic membrane integrity and acrossomal status using FITC-PNA stain were evaluated (M3). Plasma membrane, sperm motility and vigour were evaluated after a 1-hour incubation at 37ºC (M4). Statistical analysis showed that the higher temperature had positive effect on froze-thawed canine sperm characteristics. The best results were taken when canine semen was frozen in GEY extender and thawed 72ºC for 8 sec.

Foi estudada a atividade ovariana de fêmeas de onça-pintada (Panthera onca; adultas n=2 e pré-púberes n=3) mantidas em cativeiro, pela extração e quantificação de estrógenos e progestinas fecais. Foram colhidas amostras fecais de 2-7 vezes por semana durante 16-18 meses. Foi realizada a validação dos radioimunoensaios em fase sólida, para progesterona e 17b-estradiol, para uso em extratos fecais em onça-pintada. A concentração média de estrógenos fecais (ng/g de fezes secas) para o grupo dos animais pré-púberes foi de 10,97 (variando de 0,28 a 59,16) e para o grupo dos animais adultos foi de 68,99 (variando de 3,50 a 609,37). A concentração média de progestinas fecais (mg/g de fezes secas) para o grupo dos animais pré-púberes foi de 0,26 (variando de 0,02 a 4,44) e para o grupo dos animais adultos foi de 0,85 (variando de 0,08 a 6,51). | Ovarian function of captive jaguars (Panthera onca; adults n=2 and pre-pubertal n=3) was assessed by extraction and quantification of fecal estrogens and progestins. Fecal samples were obtained 2-7 times per week during 16-18 months. Validation of solid phase radioimmunoassay for progesterone and 17b-estradiol was performed for jaguar fecal extracts. Mean concentration the fecal estrogen (ng/g of dry feces) in pre-pubertal animals was 10,97 (range, 0,28 - 59,16) and adults animals was 68,99 (range, 3,50 - 609,37). Fecal progestins (mg/g of dry feces) had a mean concentration of 0,26 (range, 0,02 - 4,44) in pre-pubertal animals and adults animals was 0,85 (range, 0,08 - 6,51).
