Purpose. Analysis of the current state and experience on the loop-mediated amplification (LAMP) use to detect genetically modified plants.Methods. Literature search and analysis.Results. General information on the current state and use of the genetically modified plants is provided. Despite the wide distribution of genetically modified plants, the attitude towards them in society continues to remain somewhat wary. About 50 countries have introduced mandatory labeling of GM feed and products, provided that their content exceeds a certain threshold. In order to meet labeling requirements, effective and sensitive methods for detecting known genetic modifications in a variety of plant materials, food products and animal feed must be developed and standardized. The most common approaches to the detection of genetically modified organisms (GMOs) are the determination of specific proteins synthesized in transgenic plants and the detection of new introduced genes. Methods for the determination of GMOs based on the analysis of nucleic acids are more common, since such methods have greater sensitivity and specificity than the analysis of protein composition. Polymerase chain reaction (PCR) method is the main method of nucleic acid analysis, which is now wide used for the detection of GMOs. Loop-mediated amplification (LAMP), which can occur at a constant temperature and therefore does not require the use of expensive equipment may be an alternative to the PCR. Scientific articles about the use of the loop-mediated amplification (LAMP) for the detection of genetically modified plants were analyzed. Advantages and disadvantages of the polymerase chain reaction and loop-mediated amplification are compared.Conclusions. The main criteria for applying a method of GMO detection analysis are as follow: its sensitivity, time of reaction, availability and ease to use, cost of reagents and equipment, and the possibility for simultaneous detection of many samples. | Мета. Проаналізувати світовий досвід застосування реакції ампліфікації, що опосередкована через петлю (LAMP), для детектування генетично модифікованих рослин.Результати. Наведено загальну інформацію щодо сучасного стан і поширення генетично модифікованих рослин. Попри значне поширення генетично модифікованих рослин, ставлення до них у суспільстві й досі залишається дещо настороженим. Приблизно 50 країн запровадили обов’язкове маркування ГМ кормів та продуктів за умови, що їхній уміст перевищує певне порогове значення. Для того, щоб виконати вимоги до маркування, потрібно розробити та стандартизувати ефективні й чутливі методи визначення відомих генетичних модифікацій у різноманітній рослинній сировині, харчовій продукції та кормах для тварин. Найпоширенішими підходами до детектування генетично модифікованих організмів (ГМО) є визначення специфічних білків, що синтезуються у трансгенних рослинах, та детектування нових привнесених генів. Методи визначення ГМО, засновані на аналізі нуклеїнових кислот, є поширенішими, оскільки мають більшу чутливість та специфічність порівняно з аналізом білкового складу. Основним методом аналізу нуклеїнових кислот, що зараз використовується для детектування ГМО, є метод полімеразної ланцюгової реакції (ПЛР). Альтернативою методу ПЛР убачається реакція ампліфікації, що опосередкована через петлю (LAMP), яка може відбуватися за постійної температури й тому не потребує використання коштовного обладнання. Проаналізовано наукові публікації, що стосуються використання реакції LAMP для детектування генетично модифікованих рослин. Описано переваги та недоліки методів полімеразної ланцюгової реакції та ампліфікації, що опосередкована через петлю.Висновки. Основним критерієм для застосування того чи іншого методу аналізу ГМО є, насамперед, його чутливість, тривалість реакції, доступність та простота виконання, вартість реагентів і обладнання, а також можливість здійснювати одночасне детектування якомога більшої кількості зразків.

Мета. Створити сорт промислових конопель середньоєвропейського еколого-географічного типу універсального напряму господарського використання з підвищеним умістом олії в насінні та поліпшеною якістю волокна.Методи. Селекційні (самозапилення, сортолінійна гібридизація в умовах вегетаційного будинку, добір), польові, лабораторні, інструментально-технологічна оцінки оцінка якості волокна, статистичні.Результати. У результаті гібридизації сорту ‘Глесія’ із самозапиленою лінією шостого покоління сорту ‘Золотоніські 15’ і наступного добору на закріплення ознак високої продуктивності та поліпшення якісних ознак коноплепродукції створено новий сорт ‘Артеміда’. Сорт належить до середньостиглої групи: вегетаційний період до настання фази технологічної стиглості становить 94 доби, біологічної – 118 діб. За вирощування на зеленець забезпечує високі показники врожайності волокна (2,56 т/га) та виходу всього волокна (30,4%, зокрема довгого волокна – 27,6%). У разі вирощування на двобічне використання сорт ‘Артеміда’ формує вищі, порівняно із сортом-стандартом ‘Гляна’, урожаї волокна (2,01 т/га) та насіння (1,29 т/га) з підвищеним умістом в останньому олії (36,8%). При цьому він істотно поступається за висотою рослин, що позитивно для збирання насіння зернозбиральним комбайном. Аналіз відповідності емпіричного й теоретичного розподілу ознаки вмісту олії в насінні елітних рослин сорту ‘Артеміда’ свідчить про її високу стабільність. Примітною особливістю сорту є формування дружних сходів та інтенсивний ріст рослин на початку вегетації, що сприяє зменшенню забур’яненості посівів.Висновки. ‘Артеміда’ – новий сорт конопель універсального напряму господарського використання. Належить до середньоєвропейського еколого-географічного типу, хоча і створений у результаті сортолінійної гібридизації різних типів з наступним поліпшувальним селекційним добором за ознаками продуктивності. Характеризується повною відсутністю канабіноїдних сполук, підвищеним умістом олії, поліпшеним її жирнокислотним складом та високою якістю волокна. Рекомендується для вирощування з метою отримання волокна й насіння. Завдяки високій потенційній урожайності продукції є конкурентоздатним на ринку промислових конопель. | Purpose. Creation of industrial hemp variety of multiple purposes with the absence of cannabinoid compounds, high oil content in seeds and fiber quality.Methods. Breeding (self-pollination, varietal-linear hybridization in the conditions of a vegetation house, selection), field, laboratory, instrumental-technological assessment of fiber quality, mathematical statistics.Results. The ‘Artemida’ variety was created as a result of hybridization of the ‘Hlesiia’ variety with the self-pollinated line of the sixth generation of the ‘Zolotoniski 15’ variety and selection for stabilization of high productivity traits and improvement of quality trait of hemp production. The variety belongs to the medium-ripe group; the growing season is 94 days before the phase of technological maturity and 118 days before the phase of biological maturity. When grown for fiber, the variety had a higher fiber yield (2.56 t/ha), the yield of all fiber (30.4% and including long fiber 27.6%). When grown for fiber and seeds, the plant is significantly inferior in height, which is positive for harvesting seeds with a combine harvester, has a significantly higher seed yield (1.29 t/ha), oil content (36.8%) and fiber yield (2.01 t/ha) in comparison with the standard of the variety ‘Hliana’. The analysis of the correspondence between the empirical and theoretical distribution of such a trait as the oil content in the seeds of elite plants of the ‘Artemida’ variety indicates its high stability. A notable feature of the variety is the formation of friendly seedlings and intensive plant growth at the beginning of the growing season, which helps to reduce the weediness of crops. Conclusions. The new variety of hemp ‘Artemida’ of multiple purposes belongs to the Central European ecological and geographical type, although created as a result of varietal-linear hybridization of different types with selection on the basis of productivity, is characterized by complete absence of cannabinoid compounds, high oil content and fiber quality. The variety is recommended for growing for fiber and seeds. Due to its high yield potential, it is competitive in the industrial hemp market.

Purpose. Grapevines (Vitis spp.) are affected by many viral diseases which cause serious pathological problems. GLRaV-3 is among the most widespread leafroll viruses, while Grapevine Fanleaf Virus (GFLV) is a destructive pathogen which reduces the lifespan of grapevine. Considering the impact and the spread of these diseases, our objective was to analyse the presence of these two viruses in several grapevine varieties in grapevine collection at ATTC Vlore. Data gathered from plant pathogens serve to better understand and prevent the spread of pathogens, as a mandatory rule for the quality control of certified plant material during vegetative propagation. Method. The presence of two common viruses were tested using virus specific primers; LC1/LC2 primer pair designed from the hHSP70 gene for detecting Grapevine Leafroll-associated Virus-3 (GLRaV3) and C3390/H2999 primer pair, designed from coat protein coding regions for detecting Grapevine Fanleaf Virus (GFLV), in six varieties; ?Merlot?, ?Kallmet?, ?Shesh i zi?, ?Shesh i bardh??, ?Debin??, and ?Pul?z?, provided through a randomised sampling procedure. One Step Reverse Transcription Polymerase Chain Reaction assay was used to detect the viral presence. Results showed a high (100%) prevalence of GLRaV3 virus in all of analysed samples, as the most frequent among the two pathogens. Analysis for of GFLV virus showed low infection rate, being present in only one sample. Conclusions. We herein show an efficient, fast and reproducible method for detecting grapevine viruses through one step RT-PCR. Рослини винограду (Vitis spp.) уражуються багатьма вірусними збудниками, що спричиняють їхні серйозні захворювання. До найпоширеніших належать вірус скручування листя винограду (GLRaV-3) та вірус коротковузля винограду (GFLV), деструктивний патоген, який зменшує тривалість життя виноградної лози. З огляду на важливість і поширення захворювань, що спричиняються згаданими вірусами, нашою метою було проаналізувати їхню присутність у сортах винограду з колекції Центру Трансферу Агарних Технологій (ATTC Vlore). Отримані дані про рослинні патогени потрібні для запобігання їхньому поширенню і є обов’язковими для контролювання якості сертифікованого рослинного матеріалу під час його вегетативного розмноження. Методи. Наявність вірусів перевіряли методом одностадійної ЗТ-ПЛР з використанням вірус-специфічних праймерів: пара праймерів LC1 / LC2, що розроблена для детектування гена hHSP70 вірусу скручування листя винограду-3 (GLRaV3), і пара праймерів C3390 / H2999, для визначення кодувальних послідовностей білка оболонки вірусу коротковузля винограду (GFLV). Аналіз шести сортів культури – ‘Merlot’, ‘Kallmet’, ‘Shesh i zi’, ‘Shesh i bardhё’, ‘Debinё’ і ‘Pulёz’ – здійснювали з використанням процедури рандомізованої вибірки. Результати. Найпоширенішим вірусом для досліджених зразків виявився GLRaV3, який траплявся у 100% проаналізованих рослин. Визначення вірусу GFLV показало низький рівень інфікування, вірус був лише в одному зразку. Висновки. Показана можливість використання одностадійної ЗТ-ПЛР як ефективного, швидкого й відтворюваного методу виявлення вірусів виноградної лози.

Features of growth of sorghum [Sorghum bicolor (L.) Purpose. To establish the optimal row spacing and so?wing rate of sorghum seeds of grain varieties ?Dniprovskyi 39? and ?Vinets?, to substantiate their influence on the growing season and biometric parameters of plants in the Right-Bank Forest-Steppe of Ukraine.Methods. Field, laboratory, mathematical and statistical.Results. The most intensive growth and development of sorghum plants was observed when sowing seeds with a row spacing of 45 cm and a seeding rate of 200 thousand pieces/ha. In particular, the duration of the growing season under such conditions was the smallest: 108 days for the ?Dniprovskyi 39? variety and 106 days for the ?Vinets? variety. At the same time, the indicators of field seeds germination, plant height and stem diameter were maximum in the experiment: ?Dniprovskyi 39? ??88.7%, 137.3 cm and 1.7 cm, ?Vinets? ? 86.9%, 121.8 cm?and 1.6 cm, respectively. It was found that an increase in seeding rate reduced indicators of productive tillering, leaf area and weight per plant. The most intense tillering of sorghum plants was observed at a seeding rate of 150 and 200 thousand pieces/ha for all the studied variants of the row spacing: on average, up to two panicles well filled with grain per plant, depending on the varietal characte?ristics. At the rate of 250 thousand pieces/ha, tillering of plants in both varieties was somewhat weaker ? 1.0?1.1 panicles per plant. Мета. Установити оптимальні ширину міжрядь та норму висіву насіння сорго зернового сортів ‘Дніпровський 39’ й ‘Вінець’, обґрунтувати їхній вплив на тривалість вегетаційного періоду й біометричні показники рослин в умовах Правобережного Лісостепу України. Методи. Польовий, лабораторний, математично-статистичний. Результати. Найінтенсивніший ріст і розвиток рослин сорго зернового відзначено за сівби насіння із шириною міжрядь 45 см та нормою висіву 200 тис. шт./га. Зокрема, тривалість вегетаційного періоду за таких умов була найменшою: 108 діб у сорту ‘Дніпровський 39’ та 106 діб у сорту ‘Вінець’. Водночас показники польової схожості насіння, висоти рослин та діаметра стебла були максимальними в досліді: ‘Дніпровський 39’ – 88,7%, 137,3 та 1,7 см, ‘Вінець’ – 86,9%, 121,8 та 1,6 см відповідно. Установлено, що зі збільшенням норми висіву насіння зменшуються показники продуктивної кущистості, площі листкової поверхні та маси однієї рослини. Найінтенсивніше рослини сорго зернового кущились за норми висіву насіння 150 та 200 тис. шт./га за всіх досліджуваних варіантів ширини міжрядь: у середньому до двох добре виповнених зерном волоті на одну рослину залежно від сортових особливостей. За норми 250 тис. шт./га кущіння рослин в обох сортів відбувалося дещо слабкіше – 1,0–1,1 волоті на рослину. Найбільшими показники площі листкової поверхні та маси однієї рослини були за ширини міжрядь 45 см: 1528–2320 см2 і 169,2–185,6 г у сорту ‘Дніпровський 39’ та 1476–2180 см2 і 143,1–162,3 г у сорту ‘Вінець’ залежно від густоти посіву. Зменшення ширини міжрядь до 15 см і збільшення до 70 см призводило до зниження основних параметрів росту й розвитку рослин. Висновки. Найліпше розвивалися рослини сорго зернового за сівби із шириною міжрядь 45 см та нормою висіву 200 тис. шт./га, які й рекомендуються для вирощування культури в умовах Правобережного Лісостепу України.

Purpose. To establish the optimal row spacing and sowing rate of sorghum seeds of grain varieties ‘Dniprovskyi 39’ and ‘Vinets’, to substantiate their influence on the growing season and biometric parameters of plants in the Right-Bank Forest-Steppe of Ukraine. Methods. Field, laboratory, mathematical and statistical. Results. The most intensive growth and development of sorghum plants was observed when sowing seeds with a row spacing of 45 cm and a seeding rate of 200 thousand pieces/ha. In particular, the duration of the growing season under such conditions was the smallest: 108 days for the ‘Dniprovskyi 39’ variety and 106 days for the ‘Vinets’ variety. At the same time, the indicators of field seeds germination, plant height and stem diameter were maximum in the experiment: ‘Dniprovskyi 39’ – 88.7%, 137.3 cm and 1.7 cm, ‘Vinets’ – 86.9%, 121.8 cm and 1.6 cm, respectively. It was found that an increase in seeding rate reduced indicators of productive tillering, leaf area and weight per plant. The most intense tillering of sorghum plants was observed at a seeding rate of 150 and 200 thousand pieces/ha for all the studied variants of the row spacing: on average, up to two panicles well filled with grain per plant, depending on the varietal characteristics. At the rate of 250 thousand pieces/ha, tillering of plants in both varieties was somewhat weaker – 1.0–1.1 panicles per plant. The drought resistance of plants in the field was assessed according to S. S. Pyatnitsky 6-point scale (1961); experiments in the laboratory were to determine the water hol?ding capacity of leaves, their water deficiency, the ability to restore turgor, hydration of tissues according to the unified method of the Institute of Horticulture NAAS (Kytaiev et al., 1998, 2009).Results. According to visual observations in periods with low moisture supply, leaf turgor did not decrease, so in the field, drought resistance of hybrid tea roses was estimated at 5 points on a 6-point scale. In the laboratory, water deficiency in the leaves was calculated (as a percentage of the total water content in the state of full saturation). According to the indicators of the water-holding capacity of leaf tissues, varieties with the level of moisture loss in the exposure after 12 hours from the lowest to the highest, were selected.Conclusions. All varieties of hybrid tea roses from the collection of M. M. Gryshko NBG of NAS of Ukraine, included in the experiment, in field conditions were rather drought-resistant. According to the indicators of drought resistance of leaves from 44 model varieties, 9 with low (up to 22.30%), 5 with medium (up to 24.37%), 30 with high and very high level of moisture loss were identified (27.23?46.47%). Analysis of the research results shows that the physiological processes associated with water loss are a variety-specific, genetically inherited trait. Varieties of hybrid tea roses of diffe?rent geographical origin, which showed the criterion of drought resistance from medium to highest, can be recommended for cultivation in the Right-Bank Forest-Steppe of Ukraine.

Мета. Установити посухостійкість сортів троянд чайно-гібридної садової групи у польових і лабораторних умовах. Методи. Польовий, біометричний, лабораторний, статистичний. Посухостійкість рослин у польових умовах оцінювали за 6-баловою шкалою С. С. П’ятницького (1961), досліди в лабораторних умовах полягали у визначенні водоутримувальної спроможності листків, їх водного дефіциту, здатності відновлювати тургор, оводненості тканин за уніфікованою методикою Інституту садівництва НААН (Китаєв та ін., 1998, 2009). Результати. За візуальними спостереженнями в періоди з низьким рівнем вологозабезпечення тургор листків не знижувався, тож у польових умовах посухостійкість сортів чайно-гібридних троянд оцінено у 5 балів за 6-баловою шкалою. У лабораторних умовах вираховували водний дефіцит у листках (у відсотках від загального вмісту води в стані повного насичення). За показниками водоутримувальної здатності тканин листків сортів чайно-гібридних троянд, виділено сорти з рівнем втрати вологи в експозиції через 12 годин – від найнижчого до найвищого. Висновки. Усі сорти чайно-гібридних троянд колекції НБС ім. М. М. Гришка НАН України, які були у досліді, в польових умовах є достатньо посухостійкими. За показниками посухостійкості листків із 44-х модельних сортів виділено 9 із низьким (до 22,30%), 5 – із середнім (до 24,37%), 30 – з високим і дуже високим (27,23–46,47%) рівнем утрати вологи. Аналіз отриманих результатів досліджень свідчить про те, що фізіологічні процеси, пов’язані з утратою вологи є сортоспецифічною, генетично спадковою ознакою. Сорти чайно-гібридних троянд різного географічного походження, що показали критерій посухостійкості від середнього до найвищого, можна рекомендувати для вирощування в умовах Правобережного Лісостепу України

Purpose. Analysis of species and varietal diversity of the genus Salvia L. cultivated flora of the world, including Ukraine; prognostic assessment and determination of the directions of ornamental representatives of the genus introduction to the conditions of the Forest-steppe of Ukraine.Methods. Analysis and synthesis, comparison and generalization of information data, introduction forecast.Results. The modern assortment of the genus Salvia is presented in quantitative, taxonomic, arealogical and ecological-cenotic terms. Belonging of the species of the genus to 6 main centers of origin of cultivated plants was determined, and a significant diversity of life forms (nanophanerophytes, microphanerophytes, hamephites, hemicryptophytes, theophytes), naturally formed under the influence of factors determined by different geographical and ecological-phytocenotic conditions of plant growth was reflected. Species assortment of the genus Salvia from the collection fund of the M.?M.?Gryshko National Botanical Garden of the National Academy of Sciences of Ukraine was analyzed in comparison with the collections of other Ukrainian botanical gardens. The list of decorative species of the collection, found in the natural flora of Ukraine, was given. Three groups of species have been identified by origin, what determines the conditions for plant cultivation in the Forest-Steppe of Ukraine. In terms of taxonomic structure, introduced sage species belong to eight (according to Bentham, 1833) out of twelve sections, which represent biomorphological diversity and confirm the high introduction potential of these plants.? The species and interspecific hybrids most fully used in breeding work and represented by a significant varietal diversity were distinguished, as well as the sage assortment of the collection of ornamental plants of the NBG was presented.Conclusions. It has been revealed that the species and varietal diversity of the genus Salvia of the world cultivated flora has a significant introduction potential for the Forest-Steppe zone of Ukraine. It was determined that the main base for the introduction of the ornamental species of the genus Salvia in Ukraine is the collection of the M.?M.?Gryshko National Botanical Garden of the National Academy of Sciences of Ukraine. Purpose. Analysis of species and varietal diversity of the genus Salvia L. cultivated flora of the world, including Ukraine; prognostic assessment and determination of the directions of ornamental representatives of the genus introduction to the conditions of the Forest-steppe of Ukraine. Methods. Analysis and synthesis, comparison and generalization of information data, introduction forecast. Results. The modern assortment of the genus Salvia is presented in quantitative, taxonomic, arealogical and ecological-cenotic terms. Belonging of the species of the genus to 6 main centers of origin of cultivated plants was determined, and a significant diversity of life forms (nanophanerophytes, microphanerophytes, hamephites, hemicryptophytes, theophytes), naturally formed under the influence of factors determined by different geographical and ecological-phytocenotic conditions of plant growth was reflected. Species assortment of the genus Salvia from the collection fund of the M. M. Gryshko National Botanical Garden of the National Academy of Sciences of Ukraine was analyzed in comparison with the collections of other Ukrainian botanical gardens. The list of decorative species of the collection, found in the natural flora of Ukraine, was given. Three groups of species have been identified by origin, what determines the conditions for plant cultivation in the Forest-Steppe of Ukraine. In terms of taxonomic structure, introduced sage species belong to eight (according to Bentham, 1833) out of twelve sections, which represent biomorphological diversity and confirm the high introduction potential of these plants. The species and interspecific hybrids most fully used in breeding work and represented by a significant varietal diversity were distinguished, as well as the sage assortment of the collection of ornamental plants of the NBG was presented. Conclusions. It has been revealed that the species and varietal diversity of the genus Salvia of the world cultivated flora has a significant introduction potential for the Forest-Steppe zone of Ukraine. It was determined that the main base for the introduction of the ornamental species of the genus Salvia in Ukraine is the collection of the M. M. Gryshko National Botanical Garden of the National Academy of Sciences of Ukraine. The main directions of further introduction and breeding work with representatives of the genus Salvia in the Forest-Steppe conditions of Ukraine were highlighted. | Мета. Аналіз видового та сортового різноманіття роду Salvia L. культивованої флори світу, зокрема й України; прогностична оцінка та визначення напрямів інтродукції декоративних представників роду в умови Лісостепу України. Методи. Аналіз і синтез, порівняння та узагальнення інформаційних даних, інтродукційний прогноз. Результати. Представлено сучасний асортимент роду Salvia в кількісному, таксономічному, ареалогічному та еколого-ценотичному відношенні. Визначено приналежність видів роду до шести основних центрів походження культурних рослин, а також відображено значне розмаїття життєвих форм (нанофанерофіти, мікрофанерофіти, хамефіти, гемікриптофіти, терофіти), які закономірно сформувалися під впливом чинників, що визначалися різними географічними та еколого-фітоценотичними умовами зростання рослин. Проаналізовано видовий асортимент роду Salvia колекційного фонду Національного ботанічного саду імені М. М. Гришка НАН України (НБС) порівняно з колекціями інших ботанічних садів України. Наведено перелік декоративних видів колекції, що трапляються в природній флорі України. Виділено три групи видів за походженням, які й визначають умови культивування рослин у Лісостепу України. За таксономічною структурою інтродуковані види шавлій належать до восьми (згідно з Bentham, 1833) з дванадцяти секцій, які представляють біоморфологічне різноманіття та підтверджують високий інтродукційний потенціал цих рослин. Виділено види й міжвидові гібриди, які найбільш задіяні в селекційній роботі та репрезентовані значним сортовим розмаїттям, а також представлено сортимент шавлій колекції квітниково-декоративних рослин НБС. Висновки. Установлено, що видове та сортове різноманіття роду Salvia культивованої флори світу має значний інтродукційний потенціал для лісостепової зони України. Визначено, що основною базою для інтродукційного вивчення декоративних видів роду Salvia в Україні є колекція Національного ботанічного саду імені М. М. Гришка НАН України. Виокремлено основні напрями щодо подальшої інтродукційної та селекційної роботи з представниками роду Salvia в умовах Лісостепу України.
